ABSTRACTBurn wound is damaged tissue caused by contact with a heat source which causes infection, so needed some drug for this burn wound. Ointment is a pharmaceutical dosage form used on the skin which is intended for topical effect. In African leaves or Vernonia amygdalina  which has many chemical contents and useful effects. This study aimed to test the quality preparation of ointments made from African leaves ethanol extract with hydrocarbon and absorption and its effect on burn wound in rabbits in order to find out effective ointment base for burn wound. The procedures applied were to dry the samples and extract them by maceration. After that, the samples were soaked using ethanol and diaterbath to obtain their thick extract. Later, the base ointment was mixed with thick extract using hot mortar, and tested for its physical properties and effectiveness of burn wound. The results showed that the hydrocarbon and absorbtion ointment base has good quality except for absorption’s poor distribution capacity due to its small dispersive power. African leaves extract ointment with hydrocarbon base and absorption has good quality preparations except for the absorption base’ dispersion power for due to its small dispersive power. To conclude, this study proves that the African leaf extract ointment with hydrocarbon base is very effective for burn wound healing. Key Words: African Leaves, Burn Wound, Ointment. ABSTRAKLuka bakar merupakan rusaknya jaringan yang disebabkan kontak dengan sumber panas yang dapat menimbulkan infeksi, maka dari itu dibutuhkan sediaan yang dapat menangani luka bakar tersebut. Sediaan salep merupakan bentuk sediaan farmasi yang digunakan pada kulit yang dimaksudkan untuk efek topikal. Pada daun Afrika (Vernonia amygdalina) dapat ditemukan banyak kandungan kimia dan efek yang baik juga. Pada penelitian ini bertujuan untuk menguji salep ekstrak etanol daun Afrika dengan basis hidrokarbon dan absorbsi terhadap sediaan mutu dan juga terhadap luka bakar pada kelinci kemudian menemukan basis salep yang efektif untuk luka bakar. Sampel dikeringkan dan diekstraksi secara maserasi dan direndam menggunakan etanol dan diwaterbath untuk mendapatkan ekstrak kental, bahan basis salep dicampurkan dengan ekstrak kental menggunakan mortar panas, diuji sifat fisik dan efektifitas terhadap luka bakar. Basis hidrokarbon sangat efektif untuk luka bakar. Untuk basis hidrokarbon bersifat melunakkan lapisan kulit karena occlusive sehingga akan meningkatkan hidratasi kulit dengan menghambat penguapan air pada lapisan kulit, akibat hidratasi lapisan kulit, mungkin juga akan meningkatkan aktivitas obat. Absorbsi menunjukkan kemampuan basis dalam menyerap air, bukan kemampuan obat diabsorpsi oleh kulit. Salep ekstrak daun Afrika dengan basis hidrokarbon dan absorbsi memiliki sediaan mutu yang baik kecuali pada bagian daya sebar untuk basis absorbsi karena memiliki hasil daya sebar yang kecil, basis hidrokarbon salep ekstrak daun Afrika sangat efektif terhadap penyembuhan luka bakar. Kata Kunci: Daun Afrika, Luka Bakar, Salep.

Objective: Macrophages are essential cells in burn wound healing. The primary function of macrophages is phagocytosis of microbes and necrosis tissue. Migration of macrophages into the wound area is affected by interleukin-1 (IL-1). Ethanol extract of Cincau or green grass jelly leaves or Cyclea barbata Miers contains pectin, flavonoids, and alkaloids. These compounds influence the inflammatory process. The purpose of this study was to reveal the effect of ethanol Cincau leaves extract on the expression of IL-1 and the number of macrophages in second degree burn wounds tissue. Methods: This study used 30 rats divided into six groups based on different treatments: NaCl 0.9% (K), silver sulfadiazine/(SSD) (P1), hydrogels (P2), Cincau extract 40% (P3), Cincau extract 50% (P4), and Cincau extract 60% (P5). Treatment was conducted over 4 days. On the 5th day, burn wounds tissues were taken, then immunohistochemistry staining was applied. Results: One-way ANOVA test on the expression of IL-1 showed a difference among groups (p<0.05) and on the number of macrophages (p<0.05). Post hoc tests on the expression of IL-1 showed Cincau extracts 40% and 50% treated-groups significantly differ with the NaCl 0.9% group but not with hydrogel treated group. The similar result showed Cincau extracts 40% and 50% treated-groups significantly differ from the control group but not with SSD and hydrogel groups. Conclusions: It can be concluded that ethanol extract of Cincau leaves can decrease IL-1 expression and increase the number of macrophages which have an important role in inflammation resolution of burn wounds.

Eupatorium odoratum L. is traditionally used to treat open wounds. Aim of this study was toinvestigate the healing activity of ointment containing extract of E. odoratum in burns induced in malealbino mice. Mice were divided into 5 groups, all were induced for burn wound using a heat stamp intemperature 80°C for 20 minutes. Group I was treated with ointment base (control), group II, III and IVwere treated with extract ointment in concentration of 5%, 10%, and 20% w/w respectively, and the lastgroup V was treated with ointment reference. Observations were made during 21 days exactly on the 7th,14th and 21st day, which included parameters: percentage of healed area, epithelialization time andcollagen scores. The result showed that on 14th day, mean of healed area in group III (75.89%±7.76%)and IV (76.29%±6.981%) were significantly higher than other groups (P<0.05). On the 14th and 21th day,collagen scores in groups III (2.33±0.577 and 2.67±0.577) and IV (2.67±0.577 and 2.33±0.577) are equalwith reference group, these score are significantly higher than in groups I and II (P<0.05). Meanwhile,though epithelialization time in group III and IV (both 14.33 days±0.577days) are more quickly thanothers, statistical analysis showed that there is no significant difference on time between all groups(P>0.05).

Areca seeds contain flavonoid, tannin, and alkaloid which can help the healing process of burn wounds on the back skin of mice. Areca seeds are used for treatment because it has  antimicrobial activity. Areca Seeds are one of Indonesian plants which the fruit in tranditional way is used as burn medicine. The objective of this study was to know theeffect of ethanol extract of areca seeds against burn wound healing on the back skin of mice. Fifteen mices were used in this experiment and were divided into 5 groups : vaseline flavum as a control negative, branded ointment as a control positive , ethanol extract of areca seeds in vaseline flavum with consentration 20%, 40 % and 60 %. Each group consisted of 3 mices. The back skin of mice was induced using hot iron size 1×1 cm for 2 seconds then given a treatment for 14 days. The percentage of healing of burns  by calculating the surface area of wound. The results of its preliminary phytochemical analysis showed the presence of alkaloids, flavonoids, tannins and saponins. The percentage of burn wound healing of ethanol extract in concentration 20 % was 84,33 %, 40 % was 87,67%; and 60% was 89,67%. Data was analyzed using one-way Analysis of Variance (ANOVA) which showed that data has significant differences (P < 0,05) was 0,000. LSD test showed that each treatment group had siginicant differences with significant value < 0,05.

ABSTRACT This study evaluated the effect of mupirocin topical spray on burn wound healing in a rat model. Fifteen male Sprague Dawley rats were used to create full-thickness burns on the rat dorsum using a cylindrical stainless steel rod. The rats were topically treated with normal saline solution (NSS), mupirocin spray, ointment, and solution. The wound size and morphological evaluation were investigated by photographs and clinical criterions for wound healing. The histology was observed by hematoxylin and eosin (HandE) staining assay. The immunohistochemical study was evaluated by detection of transforming growth factor-beta 1 (TGF-β1), and the ratio of matrix metalloproteinase-9 to the tissue inhibitor of matrix metalloproteinase-1 (MMP-9/TIMP-1) was quantified using the enzyme-linked immunosorbent assay (ELISA) assay. A complete healing was observed at 28 days in all treatments. Mupirocin formulations accelerated the wound healing faster than NSS in size. However, the clinical criteria indicated a desirable skin appearance in the mupirocin spray and ointment treated groups. The histological evaluations showed no differences between the treatments while the immunohistochemical study revealed that all treatments reduced the level of TGF-β1 over time, particularly on day 28 in the mupirocin spray and ointment treated groups. The MMP-9/TIMP-1 ratio was significantly lower in the mupirocin spray and ointment treated groups than in the NSS and mupirocin solution groups. This study shows the safety and efficacy in the use of mupirocin topical spray. The topical mupirocin spray is an alternative suitable for development as a human topical anti-infective and wound protection spray.

Papaya leaves has been shown to have efficacy as an analgesic. Analgesic effect caused by the flavonoids in leaves of papaya. To enhance the analgesic effi cacy papaya into dosage formulatio of nanoparticles. The purpose of this research was to create and characterized nanoparticles from ethanol extract of papaya leaves then tested for effectiveness as an analgesic. This study used polymer chitosan and NaTPP to created nanoparticles through ionic gelation method and analgesic activity tested using writhing test method and statistical tested. Optimal formula of nanoparticles were produced at concentration of 0.2% chitosan, Na-TPP extract 0.1% and 4% with a ratio of 6: 1: 1. The nanoparticles was formed a size of 255.4 nm with polidispers index of 0.7, the zeta potential of + 11,1mV, the adsorption effi ciency by 87% and rounded morphology (spherical). Analgesic effectiveness tested by comparison between the groups ethanol extract of papaya and the groups nanoparticles of chitosan-ethanol extract papaya statistically using the T-test Independent. The results of this research indicate that there are statistically signifi cant differences (p <0.05). This results shows that the preparation nanoparticles of chitosan-ethanol extract papaya is more effective as an analgesic than the ethanol extract of papaya leaves.
