Introduction: Antioxidant and α-Amylase inhibitory activity of methanolic extract of Calotropis gigantea (L.) Dryand leaves were evaluated. Methods: The antioxidant activity was evaluated by DPPH assay. The extract was fractionated in Silica gel loaded column chromatography (CC). All fractions were evaluated for their purity by TLC. Out of 11 fractions from CC, one fraction was analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). Results: The antioxidant activity of methanolic extract was found satisfactory (IC50268.80 µg/ml) as compared with ascorbic acid (141.82 µg/ml). TLC of a fractions showed a compound at Rf value at 0.45 in toluene: chloroform: methanol with mobile phase ratio 7:2:1 respectively. Conclusions: Total 17 compounds were identified by GC-MS of ethyl acetate fraction and 5-hydroxyl methyl furfural was major furan compound (59.49%). α-Amylase inhibitory activity of the same fraction showed IC50 value of 0.94 mg/ ml. The Nepalese originated C. gigentea (L.) Dryand possesses antioxidant and α-Amylase inhibitory property.

Introduction: Antilipase from natural resources are a potential tool for the treatment of obesity while antioxidant-rich plants are essential in combating degenerative diseases. The aim of this study is to determine the antilipase and antioxidant activity of Orthosiphon stamineus methanolic extract. Methods: The inhibitory activity against pancreatic lipase was determined by measuring the hydrolysis of p-nitrophenyl butyrate to p-nitrophenol at 405 nm. Antioxidant activity of O. stamineus extract was measured by 2, 2, diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity assay. Results: The O. stamineus crude extract exhibited strong lipase inhibitory activity with an IC50 value of 34.7 µg/ml. The inhibition mode study disclosed that O. stamineus could act as uncompetitive inhibitor. O. stamineus showed high antioxidant activity with an EC50 value of 26.3 µg/ml. Conclusions: The results suggest that O. stamineus has shown potential as a source of natural antilipase and antioxidant.

This study aims to find the treatment of diabetes using natural materials by exploring plants in the province of East Nusa Tenggara. his research was conducted out by extracting the Strychnos nitida G.Don stem using a method of maceration by ethanol 70%. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Simplicia from maceration and fractionation results were then tested for antioxidant activity, α-glucosidase inhibition activity and identification of active compounds. The results showed that ethyl acetate fraction had the lowest IC50 value of 86.83 μg / ml. Results of the α-glucosidase activity test showed that ethyl acetate fraction and n-heksan fraction at 900 ppm had the highest percentage of inhibition of 34.23% and 33.89%. Identification using LCMS/MS method showed that ethyl acetate fraction consist of Benzenemethamine, N, N-dioctyl- as an antioxidantcompound and compound 24-methyl-5-cholestone-hexol as an antidiabetic compound. From the results of this study, we concluded that the extract of kayu ular Strychnos nitida G.Don stem has inhibition activity toward α-glucosidase enzyme.

Samama (Anthocephalus macrophyllus) is one of a pioneer indigenous fast growing species in Indonesia which have been used as traditional medicine for various diseases. The objectives were to determine the yield extract, to analyze their antidiabetic activity by inhibition assay for α-glucosidase enzyme activity and chemical analysis with GCMS. Extraction of leaves, bark and wood samples were done by using ethanol 95%. Fractionation the most active  ethanol extract was conducted by using n-hexane and ethyl acetate. The yield of ethanol extracts in leaves, bark and wood were 13.90%, 12.87%, and 2.18% respectively. Based on antidiabetic activity assay, the bark ethanol extract was the most active extract by the IC50 value of 5.86 μg mL-1. Phytochemical analysis on bark ethanol extracts showed that they contained flavonoid, quinon, triterpenoid, saponin and tannin which were assumed have high contribution in antidiabetic activities. The result of fractionation ethanol extract bark showed that the ethyl acetate fraction was the most active fraction (IC50 6.82 μg mL-1). GCMS analysis indicated the presence of dominant phenolic compounds such as pyrocatechol, antiarol, isopropyl myristate and phenol in which were suspected have antidiabetic activity. These results strongly suggested that ethyl acetate fraction of Samama bark was a potential natural source for antidiabetic agents.

Plant derived substances and herbal medicines have recently attracted the great interest towards their versatile application, as medicinal plants used in traditional and modern medicine. The aim of the current study was to investigate the antioxidant activity of Ziziphus mauritiana leaves extract followed with the formulation of herbal creams based on ZM methanolic extract then evaluate the physical characteristic and stability of the creams. After the methanolic extract was obtained by using Soxhlet extraction, the extract was assessed for antioxidant activity by using stable 2,2- Diphenyl-1-picrylhydrazyl (DPPH) which showed the IC50 value of the ZM extract is 10.57 µg/ml, while IC50 value for ascorbic acid is 4.19 µg/ml. Four formulations of water in oil (w/o) emulsion based cream was formulated based on Ziziphus mauritiana leaves extract which are F1, F2, F3 and F4. Several physical properties were evaluated such as organoleptic, pH, viscosity, homogeneity, washability and emmolliency which proved that all formulations have good homogeneity, non greasy and under suitable pH and viscosity. Based on thermal stability test (45 °C ± 70 % RH, for 48 hours), it showed that F4 is not stable at high temperature compared to other formulations. Therefore, the present study indicated that the Ziziphus mauritiana leaves extract has great potential for personal care product development.

Oxidative stress plays a major role in diabetic physiopathology; hence, the interest of using natural antioxidants as therapeutic tools exists. The aim of this study was the evaluation of in vitro antioxidant activity and inhibitory potential of organic extracts from Aristolochia longa roots against key enzymes linked to hyperglycemia. Antioxidant activity was performed using 2,2′-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radicals and ferric reducing/antioxidant power (FRAP) methods. The α-Glucosidase and β-Galactosidase inhibitory activities were investigated using an in vitro model. Moreover, phytochemical analysis of tested extracts was carried out. The aqueous fraction of this herb exhibited the highest antioxidant activity for both DPPH and ABTS methods, IC50=125.40±2.40 μg/mL and IC50=65.23±2.49 μg/mL, respectively. However, the ethyl acetate fraction possessed the strongest inhibitory effect towards α-Glucosidase (IC50=1.112±0.026 mg/mL). Furthermore, the result showed high levels of phenolic content. The results showed that this plant could be a significant source of medically important natural compounds.

Objective: The present study aims to analyze the potential of Aglaia edulis Roxb. leaf extract to induce cytological aberrations in Allium cepa root meristem and to determine the phytoconstituents in the extract. Methods: Cytotoxicity evaluation of the leaf methanolic extract was done using Allium cepa assay using various concentrations. Volatile phytoconstituents in the extract were determined using gas chromatography–mass spectrometry analysis. Results: Considerable number of cytomictic cells along with other aberrations was observed. The occurrence of cytomixis was found to be dose dependent where it ranged from 6.58±0.35 to 29.45±0.45. The percentage of cytomictic cells among the total aberrant cells was observed between 35.19±1.67 and 77.39±1.39. The phytochemical analysis of the plant extract revealed the presence of active secondary metabolites. Conclusion: The synergistic action of the active compounds might have triggered the phenomenon of cytomixis which, in turn, could be exploited for the production of polyploids.

This study was carried out to screen the amount and the classes of secondary metabolites and to evaluate the antioxidant, cytotoxic, antifungal, and antibacterial activities of the methanolic, ethanolic, and water extracts of the roots, leaves, and flowers of Nepeta juncea Benth. The results show that the highest total phenol (69.54 ± 0.31 mg gallic acid equivalents (GAE)/g dry weight), total flavonoid (41.37 ± 0.17 mg quercetin equivalents (QE)/g dry weight), anthocyanin (6.52 ± 0.21 mg cyanidin/100 g dry weight), and tannin (47.36 ± 0.33 mg catechin/g dry weight) concentrations were recorded in the methanolic extract of the leaves of N. juncea. The gas chromatography–mass spectrometry (GC–MS) analysis of the extracts showed that 1,8-cineole, 4aα-7α-7aα-nepetalactone, β-pinene, terpinen-4-ol, and α-terpineol were the major compounds, respectively. The best 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ferric-reducing antioxidant, cytotoxic, antifungal, and antibacterial activities were observed for the methanolic extract of the leaves. For the two latter activities, the best activity was revealed on Staphylococcus aureus, Bacillus cereus, and Candida albicans. The minimum inhibitory concentration (MIC) values for the antimicrobial of the methanolic extract from the leaves were in the range of 25–100 µg/mL, whereas the minimum bactericidal concentration (MBC) values were in the range of 50–200 µg/mL. The results reported herein show that, for the first time in the literature, N. juncea is a remarkable source of antioxidant, antifungal, and antibacterial compounds.

Alocasia longiloba, locally known as ‘Keladi Candik’, has been used traditionally to treat wounds, furuncle and joint inflammations. A. longiloba can be a new source of herbal medicine against hyperuricemia by inhibiting the activity of xanthine oxidase enzyme, the enzyme which is responsible for the development of hyperuricemia in human. Existing xanthine oxidase inhibitors (XOI drugs) show several side effects on gout patients. Therefore, an alternative herbal medicine from plants, with high therapeutic property and free of side effects, are greatly needed. This study was conducted to evaluate XO inhibitory activity, chemical composition, antioxidant activity and GC-MS profile of A. longiloba. Our results showed that ethanolic petiole extract exhibited the highest XO inhibitory activity (70.40 ± 0.05%) with IC50 value of 42.71 μg/mL, followed by ethanolic fruit extracts (61.44 ± 1.24%) with the IC50 value of 51.32 μg/mL. In a parallel study, the phytochemical analysis showed the presence of alkaloid, flavonoid, terpenoids, glycoside and saponin in petiole and fruit extracts, as well as higher total phenolic and flavonoid contents and strong scavenging activity on DPPH and ABTS antioxidant assay. The GC-MS analysis of fruit and petiole extracts revealed the presence of various compounds belonging to different chemical nature, among them are limonen-6-ol, α-DGlucopyranoside, paromomycin, aziridine, phenol, Heptatriacotanol, Phen-1,2,3-dimethyl and Betulin found in ethanolic fruit extract, and Phen-1,4-diol,2,3-dimethyl-, 1-Ethynyl-3,trans(1,1-dimethylethyl), Phenol,2,6-dimethoxy-4-(2-propenyl)- and 7-Methyl-Z-tetradecen-1-olacetate found in ethanolic petiole extract. Some compounds were documented as potent anti-inflammatory and arthritis related diseases by other researchers. In this study, the efficiency of solvents to extract bioactives was found to be ethanol > water, methanol > hexane > chloroform. Together, our results suggest the prospective utilization of fruit and petiole of A. longiloba to inhibit the activity of XO enzyme.

Calligonum comosum (Polygonaceae), an Egyptian desert plant, was extracted and fractionated using petroleum ether, methylene chloride, and ethyl acetate. The total methanolic extract and other fractions were tested for their anticancer activity using Ehrlich ascites, brine shrimp and antioxidant assays. Ethyl acetate fraction proved to be the most active in all assays. Eight compounds were isolated, purified, and identified from this fraction as (+)- catechin (1), dehydrodicatechin A (2), kaempferol-3-O-rhamnopyranoside (3), quercitrin (quercetin-3-O-rhamnopyranoside) (4), β-sitosterol-3-O-glucoside (5), isoquercitrin (quercetin- 3-O-glucopyranoside) (6), kaempferol-3-O-glucuronide (7), and mequilianin (quercetin-3- O-glucuronide) (8). All isolated compounds were tested for their cytotoxicity and antioxidant activity. Compound 2 showed the best cytotoxic and antioxidant activity.

Objective: Salvadora persica L. is a dense foliaceous evergreen shrub or small tree with diversified medicinal properties. The objective of this work was to do a comparative study on phytochemical composition between different plant parts of S. persica collected from the southern region of India. Methods: The phytochemical analysis of ethyl acetate fraction of ethanolic extracts from leaves, tender stems and tree bark of S. persica was done by gas chromatography-mass spectrometry/mass spectrometry (GC-MS/MS). Also, the anti-bacterial and anti-fungal activity of extracts was analyzed in vitro by Disc-diffusion method. Results: GC-MS/MS analysis of S. persica showed 29 phytocompounds. Among them, except for eugenol, caryophyllene, benzyl isothiocyanate, oleic acid, and fatty acid, the remaining 24 phytocompounds were newly reported in the present study. For the first time, a maximum amount of benzyl isothiocyanate (73.5%) was identified from tree bark extract of S. persica and this extract showed higher in vitro antimicrobial activity against gram-positive, gram-negative bacteria and fungi than leaves and tender stems. Conclusion: The study demonstrated that benzyl isothiocyanate could be the major antimicrobial component in S. persica.
