Objective: Study described the screening potential antidiabetic activity of kabau seed extract and fraction. Methods: The powdered crude drugs weighing 1349.32 grams were extracted with a solvent with solvents with escalating polarity by using soxhletation. The solvents used were n-hexane, ethyl acetate, and 96% ethanol. Screening activity using three variations on doses on the three extracts using the glucose test tolerance method, then the alloxan induction and high-fat feed induction testing methods using selected doses, decreasing blood glucose levels using the GOD PAP enzyme and decreasing MDA levels and increased levels of the enzyme SOD. Extracts that have potential antidiabetic activity are fractionated using liquid-solid fractionation; then the fraction is screened for antidiabetic activity using the glucose test tolerance method. Results: Screening for antidiabetic activity on the three extracts using the glucose test tolerance method showed that the ethanol extract at a dose of 250 mg/kg BW. The three extracts were then screened for the next mechanism using the alloxan induction method and high-fat feed induction, the decrease in blood sugar levels by the GOD-PAP method showed a good decrease in the ethanol extract by 202.94±2 mg/dl, the three extracts showed good less significant, in the SOD enzyme method, the ethanol extract gave a good value such as the positive control value. Testing on fraction can decrease in blood sugar; the results showed that the ethanol extract and methanol fraction gave a small AUC 0-150 (32695,3 and 33167,71), where the value was close to the result of the glibenclamide 30238,48. Conclusion: The antidiabetic activity of the extract showed that the ethanol extract was better with the glucose test tolerance method, with alloxan induction animal models and high-fat feed induction. In the methanol fraction derived from 96% ethanol extract, it provides a good reduction in blood sugar levels in the screening method with a glucose test tolerance

Diabetes Mellitus (DM) is a collection of metabolic symptoms that arise in a person caused by an increase in blood glucose due to damage to insulin secretion or resistance to insulin or both. Diabetes mellitus medication can be either parenteral or oral which can cause side effects in long-term use such as mild diarrhea, bloating, fatigue acidosis, even hypoglycemic renal impairment. To prevent it can use alternative medicine using Dayak onion (Eleutherine palmifolia (L) Merr.). One of the active substances contained in the Dayak onion is the flavonoids. Flavonoids have antidiabetic activity through their functions as antioxidants. The study aims to determine the activity of Dayak scallion Extract (Eleutherine palmifolia (L) Merr.) To decrease blood sugar levels (Mus muscullus), which is glucose-induced. The research was conducted with research draft pre and post test with control group design. The mice used amounted to 25 male white mice that were divided into 5 groups i.e. negative control (CMC Na 0.5%), control positive (Glibenklamid 3 mg/kgBB), Dayak onion-leaf ethanol extract at a dose of 50 mg/kgBB, 100 mg/kgBB and 200 mg/kgBB. Observations of the decrease in blood sugar levels of each test group were conducted on the same day, 30 minutes, 60 and 90 after hyperglycemia. Results showed that Dayak onion leaves ethanol extract had activity as a lowering of blood sugar levels in glucose-induced white mice

Diabetes mellitus constitutes a metabolic disorder indicated by a decrease in insulin secretion, insulin insensitivity, or a combination of both. Golden sea cucumber (Stichopus hermanii) is a natural resource of Indonesian waters with benefit for reducing blood sugar levels. This research aimed to determine the anti-hyperglycemic mechanism of golden sea cucumber. This study used a experimental laboratory research with Post-test Only Control Group. The sample of this study consisted of 25 male rats aged 60-70 days, divided into 5 groups: K(-), K(+), P1, P2, and P3 randomly. Rats from K(+), P1, P2, and P3 groups were injected with streptozotocin (STZ) with a dose of 50 mg/kg BW. Treatment was administered for 21 days. K(-) and K(+) groups were fed normally while P1, P2 and P3 groups were given golden sea cucumber 80% ethanol extract with a dose of 4,25 mg/kgBW/day, 8,50 mg/kgBW/day and 12,75 mg/kgBW/day respectively. On the 22nd day, fasting blood sugar level was measured and the number of pancreatic islet was counted with histopathological examination. The administration of golden sea cucumber (Stichopus hermanii) extract on rats with hyperglycemia does not reduce blood sugar level, but the extract administration increases the number of pancreatic islet. Golden sea cucumber (Stichopus hermanni) extract may improve the number of pancreatic islet and prevent the progressive damage of pancreatic islet.C

Obesity has recently risen and become a serious health concern in Korea according to the westernized diet and altered lifestyle. Hence, there is a growing interest in the supplementation of phytochemicals to find a safe and effective functional ingredient to treat obesity. Spergularia marina Griseb (SM) has traditionally been used as a natural herb against chronic diseases in Korea. In this study, we investigated the antiobesity effects of SM in vitro and in vivo. SM ethanol extract (SME) inhibited proliferation and differentiation in murine adipocytes and primary porcine pre-adipocytes in a dose-dependent manner. In the in vivo study, supplementation of SM powder (SMP) remarkably attenuated fat accumulation in HFD-induced obese rats. In addition, SMP supplementation improved lipid profiles in the serum and tissues of high-fat induced obese rats. Collectively, these data indicated that SME exhibited antiobesity effects by modulating adipogenesis and lipolysis. Furthermore, SMP could be developed as an obesity-induced metabolic syndrome treatment.

The present study was designed to estimate the detailed antidiabetic activity ofPterospermum acerifolium(L.) Willd flowers. In vitroalpha amylase inhibition study was carried out on 50% ethanol extract of flowers (PAFEE) and its various fractions. The active ethyl acetate fraction (PAFEF) was subfractionated into three subfractions (PAFE1, PAFE2, and PAFE3) and subjected to acute toxicity studies followed by antidiabetic screeningin vivoby streptozotocin-nicotinamide induced type II diabetes. Diabetic animals treated with PAFE2 (30 mg/kg) reduced the levels of fasting blood glucose, significantly (P<0.001) compared to that of diabetic control animals. Histological studies on drug treated groups did not show remarkable positive changes inβ-cells. PAFE2 showed32.6±1.93% glucose uptake over control and, in the presence of PI3K inhibitor wortmannin, declined to13.7±2.51%. HPLC analysis of PAFE2 reveals the presence of quercetin and apigenin as major constituents and both are inhibiting the glycogen phosphorylase enzyme in molecular modelling studies. The study evidenced strongly that the probable glucose lowering mechanism of action of active subfraction PAFE2 is by increasing the glucose uptake in peripheral tissues and by inhibition of gluconeogenesis.

Objective: The present study was to explore the phytochemical analysis and antidiabetic potential of the root of Commiphora caudata in high-fat diet (HFD) streptozotocin-induced diabetic rats. Methods: The ethanolic root extract of C. caudata at a dose of 400 mg/kg and 200 mg/kg was administered to diabetic rats. Glibenclamide (5 mg/kg) was used as standard drug. Results: The data were statistically assessed using one-way ANOVA followed by Dennett’s multiple comparison tests. To unfold the mechanism, we studied all the biochemical parameters glucose, total cholesterol, triglycerides (TG), high-density lipoproteins (HDL), low-density lipoproteins (LDL), and very LDL (VLDL) and histopathological examination of the pancreatic tissue section. The ethanolic extracts of root of C. caudata showed significant reduce of the level of cholesterol, TG, LDL, VLDL, and significant increase in the serum level of HDL at 400 mg/kg rather than 200 mg/kg. Conclusion: Further studies should look into the characterization and isolation of the constituents to know the exact mechanism of hypoglycemic activity. Statistical analyses of this screening method confirm that the proposed method is appropriate and it can be expected to improve basic idea to the researcher who is working in area-like antidiabetic activity.

Objective: The early stage of diabetes mellitus type 2 is associated with postprandial hyperglycemia. The therapeutic approach involved in the treatment of type 2 diabetes mellitus is the use of agents that can decrease postprandial hyperglycemia by inhibiting carbohydrate digesting enzymes. In an effort of identifying herbal drugs which may become useful in the prevention or mitigation of diabetes, the antidiabetic activity of Ficus semicordata (FS) and its constituents were studied. The present study was undertaken in part to identify the potent antihyperglycemic fraction from the ethanol extract of the plant, using bioassay guided evaluation. Methods: The ethanol extract of Ficus semicordata were fractionated to obtain chloroform, ethyl acetate, n-butanol and ethanol extracts which were tested for alpha-amylase, alpha-glucosidase inhibitory, properties. Further fractionation of the more active ethanol fraction yielded isolates FS-1 and FS-2 which were tested for in vivo antidiabetic activity using Streptozotocin (STZ)-induced diabetic rats. Results: Ethanol extract from leaves of the plant showed notable alpha-amylase (IC50 = 3.352µg/ml and alpha-glycosidase inhibitory activity (IC50= 3.448µg/ml) as compared to standard acarbose (IC50 = 3.175µg/ml. Subfraction FS-1 and FS-2 which were tested for in vivo antidiabetic activity using acute STZ-induced diabetic rats significantly (*p<0.05, **p<0.01, *** p<0.001) reduced blood glucose level. Conclusion: The Ficus semicordata plant extracts and the fractionated components could be used as a natural antidiabetic after comprehensive in vitro and in vivo biological studies.
