Here, we investigated the effects of Zingiber mioga extracts (FSH-ZM) on the moisturization and depigmentation of skin as well as wrinkle formation in UVB-irradiated HRM-2 hairless mice. The mice were divided into six groups as follows: normal control (NC), UVB-irradiated control (C), positive control 1 (PC1, L-ascorbic acid 200 mg/kg b.w.), positive control 2 (PC2, Arbutin 200 mg/kg b.w.), Z100 (FSH-ZM 100 mg/kg b.w.), and Z200 (FSH-ZM 200 mg/kg b.w.). The experiment spanned a period of 6 weeks. We found that FSH-ZM led to an increase in the expression of hyaluronan synthase 2, fibrillin-1, and elastin mRNAs, and showed improved skin hydration in HRM-2 hairless mice compared to that in the UVB-irradiated control group. Furthermore, FSH-ZM also inhibited the expression of inflammatory cytokines and wrinkle forming factors generated by UVB and reduced the formation of wrinkles in the test group relative to that in the control group by increasing collagen synthesis. Moreover, we found that FSH-ZM decreased the expression of melanogenesis factors, which improved depigmentation in UVB-irradiated hairless mice. These results suggest that Zingiber mioga can potentially be utilized to develop products aimed at improving skin moisturization and depigmentation and reducing wrinkle formation.

The aim of this randomised, double blind controlled trial was to verify the efficacy of a herbal dentifrice on the reduction of plaque and gingivitis. Forty eight volunteers with established gingivitis were randomly assigned to either a test group (herbal dentifrice) or positive control group (dentifrice with triclosan and fluoride). The dentifrices were distributed in plain white tubes by an independent pharmacy, which revealed the contents of each tube only after the experimental period. Plaque and gingivitis assessments were carried out on baseline and after 28 days of product use. All examinations were conducted by the same calibrated investigator. Subjects were instructed to brush their teeth three times daily using their assigned dentifrice for 28 days. There was a significant reduction in plaque levels in both the test and control groups. However, there was no significant difference between the groups. A significant reduction in gingivitis was observed in both groups, although there was no significant difference between them. No adverse reactions were reported. The authors concluded that both dentifrices were effective in reducing plaque and gingivitis in subjects with established gingivitis.

  Objective: The effect of Zingiber ottensii Val. rhizome and Sauropus androgynus L. Merr leaves extract combination was investigated using histologic profile of adipose tissues in obese male rats induced by high-fat and carbohydrate diets.Methods: This was a preventive study, conducted for 42 days by simultaneous administration of diets and extracts administration. The subjects were divided into 8 groups. All groups except negative control group were fed with high-fat and carbohydrate diets. Orlistat, metformin, and curcumin were used as contrast.Result: The phytochemical screening of Z. ottensii Val. rhizome extract showed the presence of flavonoids, saponins, and triterpenoids, meanwhile S. androgynus L. Merr leaves extract presented flavonoids, tannins, saponins, steroids, and triterpenoids. The results showed tissues histological differences in all test group compared with positive control. The most effective combination dose for bangle and katuk leaves extract in protecting adipose tissue was 100 mg/Kg:100 mg/Kg body weight.Conclusion: The combination of black bangle and katuk leaves extract showed a protective role, demonstrated by adipose tissues histologic profile.

1. The vitamin C in the dietary of an institution was largely destroyed by the methods of cooking and distribution.2. Some 50 mg. of ascorbic acid per head per day were required to be added to the diet to produce an optimum excretion level.3. Large doses of ascorbic acid were given to a group of adolescents in the institution over a period of several months. A record was kept of the incidences of infectious diseases in this treated group and in the remainder (controls). The following conclusions were reached:(a) The incidences of common cold and tonsillitis were the same in the two groups.(b) The average duration of illness due to the common cold was the same in the two groups.(c) The duration of illness of tonsillitis was longer in the control group than in the test group.(d) Cases of rheumatic fever and pneumonia occurred in the control group but no case of either disease occurred in the test group.

BACKGROUND: Endometritis is a gynecological disease characterized by inflammation of the endometrial glands and stroma. Inflammatory stimuli or tissue injury induce inflammatory pain through the release of cytokines. Ascorbic acid (AA) is a water-soluble Vitamin that plays a role in inhibiting the production of proinflammatory cytokines and increases the expression of anti-inflammatory cytokines. AIM: The purpose of this study was to find out the association between administration of AA and inflammatory cytokines in experimental animals Rattus norvegicus with endometritis. METHODS: The research was conducted using virgin female R. norvegicus laboratory mice weighing 250–300 g and aged 11–12 weeks with an estrus cycle of 5–6 days. Mice with regular oestrous cycles were randomly divided into three groups: group 1 was given 200 L of water orally without Escherichia coli inoculation and represented a negative control. Groups 2 and 3 were inoculated (50 L/rat) E. coli intravaginally, 106 colony-forming unit/mL, Group 2 was not given AA and the other side Group 3 was assigned AA. The interleukin (IL)-10 and tumor necrosis factor (TNF)-α _cytokines examination was carried out by histopathological examination through a biopsy of the endometrial tissue. Hypothesis testing on the data was analyzed by the Kruskal Wallis test using Statistical Package for Social Sciences. RESULTS: Data from the current study revealed that the highest mean value of IL-10 was found in the negative control group (2.5) and the lowest value in the positive control group (1.3). Regarding TNF-α _the highest mean value (2.8) was found in the treatment group and the lowest mean value (2.1) was found in the treatment group. Using the Kruskal Wallis test, IL-10 and TNF-α _showed insignificant results (p = value 0.304 and 0.145 respectively). CONCLUSIONS: The administration of AA did not affect the decrease in TNF-α _or the upregulation of IL-10 as anti-inflammatory cytokines.

Hidrogen peroksida pada perawatan in-office bleaching meninggalkan residu yang dapat menghambat polimerisasi pada sistem resin bonding. Asam askorbat 10% sebagai antioksidan dapat mengeliminasi residu peroksida sebelum proses penumpatan. Kulit buah manggis (Garcinia mangostana) memiliki aktivitas antioksidan yang berpotensi untukmenggantikan peran asam askorbat. Penelitian ini bertujuan untuk membuktikan efek gel ekstrak kulit buah manggis dalam memperbaiki perlekatan komposit pada gigi pasca in-office bleaching. Penelitian ini merupakan penelitian eksperimental laboratorium. Sebanyak 25 gigi premolar pertama rahang atas dilakukan in-office bleaching danselanjutnya dibagi menjadi 5 kelompok terdiri dari kontrol negatif (tanpa agen antioksidan), kontrol positif (asam askorbat 10%), dan kelompok perlakuan gel ekstrak kulit buah manggis 10%, 20%, dan 40%. Perlekatan komposit ditentukan dengan uji geser, dilanjutkan pengamatan mikrostruktur permukaan email. Hasil penelitian menunjukkan nilai perlekatan komposit pada kelompok kontrol negatif dan kontrol positif berturut-turut sebesar 25,82±2,88 Mpa dan 37,68±1,06 Mpa. Pada kelompok perlakuan gel ekstrak kulit manggis 10%, 20%, dan 40% diperoleh nilai perlekatan berturut-turut sebesar26,54±2,68; 32,64±2,33; dan 57,34±1,51 Mpa. Nilai perlekatan komposit pada pemberian gel ekstrak kulit manggis 20% dan 40% berbeda signifikan dibandingkan dengan kontrol negatif maupun kontrol positif (p<0,05). Kenaikan konsentrasi gel ekstrak kulit manggis memberikan peningkatan jumlah fraktur email. Gel ekstrak kulit manggis 20% dan 40% dapat memperbaiki perlekatan komposit sehingga berpotensi sebagai agen antioksidan pada gigi pasca in-office bleaching. Effect of Mangosteen Gel on Shear Bond Strength Of Composite Post In-Office Bleaching. The residual peroxide after in-office bleaching inhibits the polymerization of the resin bonding systems. Ascorbic acid (10%) as anti-oxidant agent has a potential to remove the residual peroxide before restoration procedure. Mangosteen extracthas an anti-oxidant potential that can be used to remove the residual peroxide. The aim of the study is to prove the effect of mangosteen extract gel to improve shear bond strength of composite post in-office bleaching. The experimental laboratory design was implemented to this research. The study was carried out on 25 maxillary first premolars thathad been applied in-office bleaching procedure. The teeth were divided into five groups as follow: negative-control group (without anti-oxidant agent), positive-control group (10% ascorbic acid), and the treatment groups (10%, 20%, and 40% mangosteen extract gel). Shear bond strength of the specimens was tested and followed by microstructuretesting on email. The results show that negative-control and positive-control group scores were 25,82±2,88 Mpa and 37,68±1,06 Mpa, followed by the scores of 10%, 20%, and 40% mangosteen extract gel groups which were 26,54±2,68; 32,64±2,33; and 57,34±1,51 Mpa respectively. Significant higher shear bond strength values were observed in 20% and 40% mangosteen extract gel groups compared to the negative-control and positive-control group (p<0,05). The increasing concentration of mangosteen extract gel provided increasing number of email fractures. Mangosteen extract gel 20% and 40% improve the shear bond strength of composite, confirming its potential as an antioxidant agent for the teeth post in-office bleaching.

This study aimed to observe dynamically the changes of x-ray, histomorphology appearance and serum inflammatory cytokines of cervical degenerative disease in rat models and to discuss the mechanism of cervical degeneration. Sixty Sprague Dawley rats were randomised into test ( n = 45) and control ( n = 15) groups, which were randomly subdivided into three groups corresponding to 1, 3 and 6 mo post operation. At the corresponding postoperative stage, cervical x-ray films were acquired, and intervertebral disc space and intervertebral foramen size were measured. Some serum inflammatory cytokines from all rats were quantitatively determined. Then, the morphological change in cervical intervertebral disc specimens stained with hematoxylin and eosin was observed. The results were analysed and compared among groups. Compared to the control group, the cervical x-ray and histomorphology appearance of rats in the test group showed varying degrees of degeneration. Furthermore, the serum IL-1β, TNF-α and IL-10 in the test group increased significantly at the corresponding postoperative stage ( P < 0.05, P < 0.01 and P < 0.001, respectively) compared to the control group. This model of cervical disc degeneration can accelerate imaging and histological degeneration, but it may be accompanied by changes in serum inflammatory cytokines levels.

Inflammatory diseases affecting the soft and hard tissues surrounding an implant represent a new challenge in contemporary implant dentistry. Among several methods proposed for the decontamination of titanium surfaces, the administration of topical 14% doxycycline gel seems to be a reliable option. In the present study, we evaluated the microbial effect of 14% doxycycline gel applied on titanium surfaces and exposed to human salivary microbes in anaerobic conditions. We also examined the composition of the exposed surfaces to assess the safe use of periodontal gel on titanium surfaces. Six anatase and six type 5 alloy titanium surfaces were used and divided into two groups: The test group and the positive control group. Both were cultured with human salivary samples in anaerobic conditions. On the test groups, 240 mg of periodontal gel was applied. The microbial assessment was performed with a colony-forming unit (CFU) count and matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) to identify the species. The surface integrity was assessed using scanning electron microscopy-energy dispersive X-ray spectrometry (SEM-EDS). The results demonstrated the microbial efficacy of the 14% doxycycline periodontal gel and its safe use on titanium surfaces. However, the SEM observations revealed the permanence of the gel on the titanium surfaces due to the physical composition of the gel. This permanence needs to be further investigated in vivo and a final polishing protocol on the titanium surface is recommended.

Objective. Ascorbic acid (AA) and controlled inflammatory stimuli are postulated to possess the ability to independently exert positive effects on a variety of proliferative, pluripotency, and differentiation attributes of gingival mesenchymal stem/progenitor cells (G-MSCs). The current study’s objective was to explore and compare for the first time the impact of the major inflammatory cytokines (IL-1β/TNF-α/IFN-γ), AA, or their combination on multipotency/pluripotency, proliferative, and differentiation characteristics of G-MSCs. Design. Human G-MSCs (n=5) were isolated and cultured in basic medium (control group), in basic medium with major inflammatory cytokines; 1 ng/ml IL-1β, 10 ng/ml TNF-α, and 100 ng/ml IFN-γ (inflammatory group), in basic medium with 250 μmol/l AA (AA group) and in inflammatory medium supplemented by AA (inflammatory/AA group). All media were renewed three times per week. In stimulated G-MSCs intracellular β-catenin at 1 hour, pluripotency gene expression at 1, 3, and 5 days, as well as colony-forming units (CFUs) ability and cellular proliferation over 14 days were examined. Following a five-days stimulation in the designated groups, multilineage differentiation was assessed via qualitative and quantitative histochemistry as well as mRNA expression. Results. β-Catenin significantly decreased intracellularly in all experimental groups (p=0.002, Friedman). AA group exhibited significantly higher cellular counts on days 3, 6, 7, and 13 (p<0.05) and the highest CFUs at 14 days [median-CFUs (Q25/Q75); 40 (15/50), p=0.043]. Significantly higher Nanog expression was noted in AA group [median gene-copies/PGK1 (Q25/Q75); 0.0006 (0.0002/0.0007), p<0.01, Wilcoxon-signed-rank]. Significant multilineage differentiation abilities, especially into osteogenic and chondrogenic directions, were further evident in the AA group. Conclusions. AA stimulation enhances G-MSCs’ stemness, proliferation, and differentiation properties, effects which are associated with a Wnt/β-catenin signaling pathway activation. Apart from initially boosting cellular metabolism as well as Sox2 and Oct4A pluripotency marker expression, inflammation appeared to attenuate these AA-induced positive effects. Current results reveal that for AA to exert its beneficial effects on G-MSCs’ cellular attributes, it requires to act in an inflammation-free microenvironment.

Background: Ayurvedic literature indicated use of Bryonia laciniosa (BL) for metabolic abnormalities. However only one study evaluated Bryonia laciniosa seed ethanolic extract in streptozotocin-induced diabetes in rats and was found to have anti-hyperglycemic and anti-hyperlipidemic effects. The authors looked at the renal function test in addition and found that it prevents deterioration of renal function in rats. Hence it was interesting to find if Bryonia laciniosa had any protective action in streptozotocin induced diabetic nephropathy in rats.Methods: Streptozotocin induced diabetic nephropathy model in wistar rats was standardised in phase 1. In phase 2 animals were divided into 4 groups and diabetic nephropathy was induce with STZ. Vehicle Control group animals received CMC 0.5%, Positive control group received enalapril 5 mg/kg, Test group 1 received ethanolic extract of Bryonia laciniosa (EEBL) 250 mg/kg and Test group 2 received (EEBL) 500 mg/kg from 3rd day onwards till 6 weeks. Renal function, morphology and histopthological changes were evaluated.Results: There was a significant reduction (p <0.001) in the serum creatinine and BUN in both low and high dose EEBL groups. High dose of EEBL lowered raised MDA values significantly (p<0.001) and raised the GSH values (p<0.001). On histopathology, the high dose EEBL reversed nephropathic changes which were equivalent to enalapril.Conclusions: Bryonia lacinosa in high doses 500 mg/kg has potential to reverse diabetic nephropathic changes induced in wistar rats.

Lactobacillus plantarum (LAB) is a group of lactic acid bacteria that can produce exopolysaccharide (EPS). The objective of this study was to determine the effect of adding selenium chloride oxide (SeOCl2) to the increased immunomodulatory potential of EPS L. plantarum Y-1.2. The experiment was conducted in vivo using macrophage cells of peritoneal fluid of mice induced by Staphylococcus aureus. EPS was obtained by precipitation at 4 °C. Identification test using HPLC showed that crude EPS of L. plantarum Y-1.2 containing glucose monomers for value retention times 4.198 and 4.204, crude EPS approaches the retention time of glucose BP 4.234. Testing of immuno-modulatory divided into six treatment groups. Group 1-3 as the control group, a positive control (Stimuno), negative control (crude EPS 50 mg/kg BW in mice), normal control (distilled water). Group 4-6 as the test group was given crude EPS 50 mg/kg BW in mice and selenium oxide chloride doses of 0.13 µg; 0.26 µg; 0.52 µg. The test results showed a dosage of 0.52 µg selenium chloride oxide to crude EPS of 50 mg/kg BW in mice could increase the activity and phagocytic capacity amounted to 28.06% and 28.39%
