This study aims to determine whether mangosteen rind extract (in the form of ethanol extract/EE) can be used as an anti-diarrhea drug after compared with other anti-diarrhea substances in three experimental groups. This research is an in vitro experimental study using adult male guinea pigs weighing 400-600 gr through the standard method of Magnus with the Latin square controlled experiment design. The study was conducted at the Pharmacology and Toxicology Laboratory of the Faculty of Pharmacy, University of North Sumatra. The results showed that the contraction of ileum in Ach with Atp + Ach compared the difference in contraction of ileum Ach with EE + Ach showed the difference in difference between the two contractions of the ileum was significant, the contraction of ileum in His with Dip + His compared indifference in contraction of ileum His with EE + His showed a difference indifference. the two ileal contractions are significant, the ileal contraction in the bar with Papa + Bar compared to the difference between the ileum bar contraction with EE + Bar shows no difference in the difference between the two ileum contractions. The conclusion is that the Mangosteen Skin Ethanol Extract works similarly to Papaverine Hydrochloride which is an antidiarrheal drug used to relax smooth muscles so that it can also make blood vessels dilate by relaxing smooth muscles in the walls of blood vessels.

Contractility of all types of invertebrate and vertebrate muscle is dependent upon the actions and interactions of two divalent cations, viz., calcium (Ca2+) and magnesium (Mg2+) ions. The data presented and reviewed herein contrast the actions of several organic Ca2+ channel blockers with the natural, physiologic (inorganic) Ca2+ antagonist, Mg2+, on microvascular and macrovascular smooth muscles. Both direct in vivo studies on microscopic arteriolar and venular smooth muscles and in vitro studies on different types of blood vessels are presented. It is clear from the studies done so far that of all Ca2+ antagonists examined, only Mg2+ has the capability to inhibit myogenic, basal, and hormonal-induced vascular tone in all types of vascular smooth muscle. Data obtained with verapamil, nimopidine, nitrendipine, and nisoldipine on the microvasculature are suggestive of the probability that a heterogeneity of Ca2+ channels, and of Ca2+ binding sites, exists in different microvascular smooth muscles; although some appear to be voltage operated and others, receptor operated, they are probably heterogeneous in composition from one vascular region to another. Mg2+ appears to act on voltage-, receptor-, and leak-operated membrane channels in vascular smooth muscle. The organic Ca2+ channel blockers do not have this uniform capability; they demonstrate a selectivity when compared with Mg2+. Mg2+ appears to be a special kind of Ca2+ channel antagonist in vascular smooth muscle. At vascular membranes it can (i) block Ca2+ entry and exit, (ii) lower peripheral and cerebral vascular resistance, (iii) relieve cerebral, coronary, and peripheral vasospasm, and (iv) lower arterial blood pressure. At micromolar concentrations (i.e., 10–100 μM), Mg2+ can cause significant vasodilatation of intact arterioles and venules in all regional vasculatures so far examined. Although Mg2+ is three to five orders of magnitude less potent than the organic Ca2+ channel blockers, it possesses unique and potentially useful Ca2+ antagonistic properties.

A new prenylxanthone, garcimangostanol (1), was isolated from the EtOAc-soluble partition of the ethanol extract of the pericarp of Garcinia mangostana L., along with three known compounds, namely 8-deoxygartanin (2), 1-isomangostin (3), and garcinone C (4). The structure of compound 1 was elucidated on the basis of its 1D, 2D NMR and MS data. Compounds 1–4 exhibited either significant or moderate cytotoxicity against MCF-7, A549, Hep-G2 and CNE human cancer cell lines in vitro with IC50 values from 4.0 ± 0.3 to 23.6 ± 1.5 μM by MTT colorimetric assay.

ABSTRACTBackground Staphylococcus aureus is a pathogenic bacteria that can cause infection. Biduri leaf has antibacterial compound such as tannin, flavonoid, saponin, and polyphenol.Objective This study aimed to know the difference of growth inhibition zone of Staphylococcus aureus at the various concentration of ethanol extract of biduri leaf and to determine an effective concentration.Methods This study was the true experiment with posttest only control design, used Kirby-Bauer disk diffusion method with five concentrations (20%, 40%, 60%, 80%,100%), positive control (chloramphenicol 30 µg) and negative control (ethanol 96%).Result The result of this study showed the average of inhibition zone diameter of each concentration consecutively 26,2 mm, 28,3 mm, 29,7 mm, 31 mm and 31,5 mm. One Way Anova statistic analysis showed that the value of p is 0.000 so there is the difference of growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of a biduri leaf.Conclusion The conclusion of this study is there are differences in growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of biduri leaf and an effective concentration that can inhibit of Staphylococcus aureus is concentration 20% with mean inhibition zone is 26,2 mm. Keywords: ethanol extract of biduri leaf; Staphylococcus aureus; inhibition zone

1. Three sheep fitted with duodenal re-entrant cannulas and three with large rumen fistulas were given red clover, perennial ryegrass and a 2:1 (w/w) mixture of grass and clover in two Latin square arrangements. Measurements were made of voluntary intake, digestibility, flow of nutrients into the duodenum and the flow of Cr-EDTA marker through the reticulo-rumen.2. Organic matter (OM) digestibility was similar for the three feeds but the voluntary intake decreased in the order mixture > red clover > perennial ryegrass. There was an increase in the rate of marker flow from the rumen and a decrease in retention time of the same order. Rumen volume did not change significantly.3. There was a reduction in the mean particle size of rumen contents in the order perennial ryegrass > clover > mixture. The in vitro digestibility of particles decreased with size; the reduction being more rapid for clover than perennial ryegrass.4. The proportion of ingested digestible OM appearing at the duodenum increased from 18.4% to 26.7% to 30.0% for perennial ryegrass, clover and the mixture respectively.5. A higher proportion of digestible cellulose and hemicellulose disappeared over the stomach for the perennial ryegrass feed compared to the clover and the mixture but over 96% of water soluble carbohydrates and starch disappeared over the stomach for all three feeds.6. The apparent digestibility of nitrogen was similar for all three feeds but the proportion of undigested feed N appearing at the duodenum was calculated to be greater by a factor of 1.71 and 2.52 for clover and mixture feeds compared to grass.7. It was concluded that the higher nutritive value of red clover compared to perennial ryegrass was due to an increased rate of flow of nutrients througth the reticulo-rumen and an increase in the proportion of digestible OM digested post ruminally.

Infection disease is a threat for human life. Research on preventing infection disease is very important. The aim this research was to improve the preventing of infection disease. This research was the basic on identification of MSA activity. The in vitro result of MSA which was isolated from mangosteen (Garcinia mangostana L.) and the extract of the fruit skin byusing Saccharomyces cerevisiae. The result showed that there were 11 isolates of potential Lactobacillus, that have variation in MSA activities. Four isolates of them showed that the higher activities between 9.6-11.8 were TM1, BST1, BST2, and BST5. The MSA activities showed by reducing coagulation after adding D-Mannose. The highest activities showedby isolate TM1, and the lowest showed by isolate BST7. By using this method it is clearly found that the skin fruit not contain MSAkeywords : mannose-specific adhesin (msa ), probiotic, Saccharomyces cerevisiae.

Apoptosis or programmed cell death serves to regulate physiological and pathological conditions. Garcinia mangostana Linn. is one of the medical herbs that is widely used to suppress human tongue cancer growth by inducing apoptosis. The research aimed to analyze the effect of ethanol extract of Garcinia mangostana Linn. on tongue cancer cells apoptosis Supri's Clone-1 (SP-C1). The type of the research was an experimental laboratory. Tongue cancer cells SP-C1 were treated by various ethanol extract concentrations (0, 300, 400, 500, 600, 700 pg/ml) of Garcinia mangostana Linn. to detect the apoptosis, which was done by acridine orange and ethidium bromide coloring tests. The number of tongue cancer cells SP-C1 2x104 cells/dish. Observation on cells apoptosis was observed by a fluorescent microscope with 40 x magnification. The data were analyzed using one-way ANOVA and was followed by Post Hoc test (Tukey-test) with 95% significance level. The result showed that tongue cancer cells SP-C1 apoptosis treated by various ethanol extracts of Garcinia mangostana Linn. increase significantly. The highest effect of apoptosis was detected at the 700 pg/ml concentration that has an effect on the percentage of tongue cancer cells apoptosis by 65%.

Objective: to determine the effect of mangosteen peel extract (Garcinia mangostana Linn) on ileal peristalsis separate male guinea pigs as diarrhea. Results: t-independent test, the difference: Contraction of ileum Acethlcholin with Atropine + Acethylcholin is 6.333 compared to Acethylcholin with extract + Acethylcholin is 8,500 with ANOVA test P value meaning the difference between the two contractions of the ileum is significant. The t-independent test results are the difference: Contraction of ileum Histamine with Diphenhidramine + Histamine is 8.333 compared to Histamine with extract + Histamine is 10.833 with ANOVA test P value meaning the difference between the two contractions of the ileum is significant. The results of the t-independent test are the difference: Contraction of the Barrium ileum with Papaverine + Barrium is 2,500 compared to the contraction of Barrium ileum with extract + Barrium is 2,500 with ANOVA test P value meaning that there is no difference between the two contractions of the ileum. From the three experimental groups, it was concluded that the extract worked similar to Papaverine. Cecilia's study, fruit skin extract can be used as diarrhea in Diare Effect of Mangosteen Ethanol Extract of Oleum ricini-induced Webster Swiss Mice.

We compared in vivo and in vitro dry matter (DM) and neutral detergent fiber (NDF) digestibility in donkeys using feces as microbial inoculum. Four donkeys were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments. The animals were fed two types of hay, with or without flaked barley. For the in vivo procedure, total feces were collected for 6 days from each donkey; digestibility was calculated as the difference between ingested and excreted DM and NDF. For the in vitro procedure, donkey feces were buffered and used as microbial inoculum in an Ankom DaisyII Incubator; digestibility was estimated after 60 h of incubation. In vivo results showed that the addition of barley to hays did not change the digestibility values. In vivo estimates were higher than in vitro ones. The equations used to predict in vivo estimates from in vitro data were not reliable (R2 = 0.47 and 0.21; P = 0.003 and 0.078 for NDF and DM digestibility, respectively). Further studies need to evaluate different sample size and digestion times.

Mangosteen rind has a benefical compounds liked xanthone which consisting of mangostin, mangosterol, mangostinon A and B, trapezifolixanthone, tovophyllin B, alpha and beta mangostin, garcinon B, mangostanol, gartanin, and flavonoid epikatekin that known contain an antibacterial and antidiare agent. Escherichia coliis an opportunistic bacterium that is commonly found in the human colon as a normal flora. Its unique because it can cause primary infection in the intestine such as diarrhea in children, as well as its ability to cause infection in other body tissues outside the intestine.This research was done from December 2016 to March 2017. The methods that used in this research were well diffusion test to know the inhibitory power by measuring the clear zone formed. measure the clear zone of mangosteen rind against Escherichia coli. Then to testing extract compounds, there was used phytochemicals screening test. Data there has been found will be analyzed through analysis of variance (ANOVA). In the inhibitory test of ethanol extract from mangosteen rind (G. mangostana L.), it was so effective to inhibit the growth of Escherichia coli through in-vitro test with a resistivity of 1,58 cm and categorized as a strong inhibitory, which MIC from mangosteen rind against Escherichia coli was 3,9% and the compounds that contain in ethanol extract from mangosteen rind (G. mangostana L.) were alkaloid, phenolik, flavonoid, saponin and terpenoid. G. mangostana leaves contained xanthone substances, it could be used as antioxidants. Ethanolic extract of G. mangostana leaves were formulated into nanoparticles compound which were made available into nano size through ionic gelation method. G. mangostana leaves were extracted by using maceration method with 50% ethanol as solvent, then were continued with evaporation until thick extract were formed. The nanoparticle formulas were made by mixing G. mangostana extract in ethanol, a solution of chitosan in glacial acetic acid as a polymer, and pectin as a cross-linker solution. There were three formulas (1,2,3) used, with the difference in pectin concentration which were 0.005; 0.01; and 0.02. Characterization of the three formulas include transmittance values were used spectrophotometric method and particle sizes by using particle size analysis (PSA). The results of this research showed that the transmittance of formulations 1, 2 and 3 were 95.9%; 97.7% and 97.3%. The highest transmittance value was formula 3, then were analysed its particle size and zeta potential were 2280 nm and -20.6 mV.
