Microbial cellulases have shown their potential application in various industries including pulp and paper, textile, laundry, biofuel production, food and feed industry, brewing, and agriculture. Due to the complexity of enzyme system and immense industrial potential, cellulases have been a potential candidate for research by both the academic and industrial research groups. Nowadays, significant attentions have been devoted to the current knowledge of cellulase production and the challenges in cellulase research especially in the direction of improving the process economics of various industries. Scientific and technological developments and the future prospects for application of cellulases in different industries are discussed in this paper.

Cellulase enzyme complex is comprised of three enzymes namely exo-glucanase, endo-glucanase and β-glucosidase which act synergistically to deconstruct cellulosic biomass in order to produce fermentable sugars. The enzymes are produced naturally by the living organisms such as bacteria, fungi and algae. The majority of microorganisms that live in extreme environments including hot/cold springs, rumen stomach, deep ocean trench, acidic/alkaline pH environment, have been regarded as appealing producers of cellulase. Cellulases produced by microorganisms have enormous applications in different industries such as agriculture, food and feed production, brewing, textile, laundry and biofuel production. Scientists as well as industry researchers consider cellulases as a prospective candidate for further studies due to the intricacy of the enzyme system and massive industrial potential. Scientific belief in its production and further studies challenges are receiving greater attention these days, notably in the intent of decreasing its production cost at the industrial scale. In this review, future possibilities of using cellulase for various industrial applications are also addressed.

ABSTRACT: Cellulase enzyme plays an important role in converting cellulosic biomass in high value products and therefore finds various applications in a number of industries such as pulp and paper, textile, laundry, biofuel production, food and feed industry, brewing and agriculture. Although there are various sources available for obtaining enzymes of industrial applications such as bacteria, fungi, actinomycetes, microalgae, animals and plants; microorganisms represent the most common source of enzymes because of their broad biochemical diversity, feasibility of mass culture and ease of genetic manipulation. Among the microorganisms, actinomycetes are increasingly becoming an important resource for the production of therapeutic molecules and industrially important enzymes. Considering this, the present investigation was undertaken to isolate a novel actinomycete strain from a sea sediment sample and explore its ability to produce the enzyme cellulase. After morphological and biochemical studies, this actinomycete strain was further characterized using its 16S rRNA gene sequence and a phylogenetic tree was constructed. The activity and stability of the extracted enzyme from this actinomycete was analyzed. Finally, the applications of this extracted enzyme were studied.

To date, Chlorella vulgaris is the most used species of microalgae in the food and feed additive industries, and also considered as a feasible cell factory for bioproducts. However, the lack of an efficient genetic engineering tool makes it difficult to improve the physiological characteristics of this species. Therefore, the development of new strategic approaches such as genome editing is trying to overcome this hurdle in many research groups. In this study, the possibility of editing the genome of C. vulgaris UTEX395 using clustered regularly interspaced short palindromic repeats (CRISPR)-associated protein 9 (Cas9) has been proven to target nitrate reductase (NR) and adenine phosphoribosyltransferase (APT). Genome-edited mutants, nr and apt, were generated by a DNA-mediated and/or ribonucleoprotein (RNP)-mediated CRISPR-Cas9 system, and isolated based on the negative selection against potassium chlorate or 2-fluoroadenine in place of antibiotics. The null mutation of edited genes was demonstrated by the expression level of the correspondent proteins or the mutation of transcripts, and through growth analysis under specific nutrient conditions. In conclusion, this study offers relevant empirical evidence of the possibility of genome editing in C. vulgaris UTEX395 by CRISPR-Cas9 and the practical methods. Additionally, among the generated mutants, nr can provide an easier screening strategy during DNA transformation than the use of antibiotics owing to their auxotrophic characteristics. These results will be a cornerstone for further advancement of the genetics of C. vulgaris.

N-3 polyunsaturated fatty acids (n-3 PUFAs), and especially eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are essential compounds for human health. They have been proven to act positively on a panel of diseases and have interesting anti-oxidative, anti-inflammatory or anti-cancer properties. For these reasons, they are receiving more and more attention in recent years, especially future food or feed development. EPA and DHA come mainly from marine sources like fish or seaweed. Unfortunately, due to global warming, these compounds are becoming scarce for humans because of overfishing and stock reduction. Although increasing in recent years, aquaculture appears insufficient to meet the increasing requirements of these healthy molecules for humans. One alternative resides in the cultivation of microalgae, the initial producers of EPA and DHA. They are also rich in biochemicals with interesting properties. After defining macro and microalgae, this review synthesizes the current knowledge on n-3 PUFAs regarding health benefits and the challenges surrounding their supply within the environmental context. Microalgae n-3 PUFA production is examined and its synthesis pathways are discussed. Finally, the use of EPA and DHA in food and feed is investigated. This work aims to define better the issues surrounding n-3 PUFA production and supply and the potential of microalgae as a sustainable source of compounds to enhance the food and feed of the future.

Cellulose hydrolysis has many industrial applications such as biofuel production, food, paper and textile manufacture. Here, we present a novel approach to cellulose hydrolysis using a consortium of motile bacteria, Paenibacillus vortex, that can swarm on solid medium carrying a non-motile recombinant E. coli cargo strain expressing the β-glucosidase and cellulase genes that facilitate the hydrolysis of cellulose. These two species cooperate; the relationship is mutually beneficial: the E. coli is dispersed over long distances, while the P. vortex bacteria gain from the supply of cellulose degradation products. This enables the use of such consortia in this area of biotechnology.

Composites have tremendous applicability due to their excellent capabilities. The performance of composites mainly depends on the reinforcing material applied. A Graphene nanoparticle (GNP) is successful as an efficient reinforcing material due to its versatile as well as superior properties. Even at very low content, graphene can dramatically improve the properties of polymer and metal matrix composites. In this paper the effects of GNP on composites based on epoxy resin were analyzed. Different contents of GNP (0 – 4.5 vol. %) were added to the epoxy resin. The GNP/epoxy composite was fabricated under room temperature. Mechanical tests result such as tensile, flexural and hardness test show enhancements of the mechanical properties of the GNP/epoxy composite. The experimental results clearly show an improvement in Young’s modulus, tensile strength, and hardness as compared to pure epoxy. The results of this research are strong evidence for GNP/epoxy composites being a potential candidate for use in a variety of industrial applications, especially for automobile parts, aircraft components, and electronic parts such as super capacitors, transistors, etc.

While complex lipids of seaweeds are known to display important phytochemical properties, their full potential is yet to be explored. This review summarizes the findings of a systematic survey of scientific publications spanning over the years 2000 to January 2021 retrieved from Web of Science (WoS) and Scopus databases to map the state of the art and identify knowledge gaps on the relationship between the complex lipids of seaweeds and their reported bioactivities. Eligible publications (270 in total) were classified in five categories according to the type of studies using seaweeds as raw biomass (category 1); studies using organic extracts (category 2); studies using organic extracts with identified complex lipids (category 3); studies of extracts enriched in isolated groups or classes of complex lipids (category 4); and studies of isolated complex lipids molecular species (category 5), organized by seaweed phyla and reported bioactivities. Studies that identified the molecular composition of these bioactive compounds in detail (29 in total) were selected and described according to their bioactivities (antitumor, anti-inflammatory, antimicrobial, and others). Overall, to date, the value for seaweeds in terms of health and wellness effects were found to be mostly based on empirical knowledge. Although lipids from seaweeds are little explored, the published work showed the potential of lipid extracts, fractions, and complex lipids from seaweeds as functional ingredients for the food and feed, cosmeceutical, and pharmaceutical industries. This knowledge will boost the use of the chemical diversity of seaweeds for innovative value-added products and new biotechnological applications.

Cellulose is the most abundant polymer in plants and the microbial conversion of cellulose is a subject of active research. Currently, cellulase is commonly used in many industrial applications, especially in animal feed, textile, waste water, brewing and wine making. A challenging strategy for the efficient utilization of this renewable resource is to use it as a base material for the production of desired metabolites. This chapter therefore focuses on exploring the cellulase producing bacteria and optimizing the parameters for the enzyme cellulase under varied conditions. Cellulolytic bacteria can be exploited for cellulase production which serves wide applications in industries, pharmaceuticals and further, use of these CDB as bio-inoculants can be incorporated to enhance organic matter decomposition in soil to increase soil fertility and to minimize the fertilizer application. It finds wide applications in reducing the environmental pollution and promote sustainable agriculture.

Fruits and vegetables are an important supplier of biological active substances, such as vitamins and secondary metabolites (SM) for human nutrition, but also for further industrial applications. Pepper (Capsicum annuum) is grown and consumed all over the world as a fresh vegetable or dried as a spice. It is also used as a coloring agent, as well for medical purposes. Pepper fruits are considered as an attractive source of health-related compounds, but other organs like the leaves and stem also contain considerable amounts of antioxidants, e.g., phenolic compounds. This indicates potential for valorization of residual biomass from horticultural production by using innovative bioeconomic concepts. Herein, we present an overview about the biosynthesis of phenolic compounds, with a special focus on flavonoids and their regulation in pepper, the current knowledge of amounts and distribution of these valuable substances, as well as possible strategies for: (1) increasing flavonoid contents in pepper, (2) improving the nutritional value of fruits, and (3) new concepts for utilization of residual biomass from horticultural production.
