Introduction: One strategy for molecular cancer therapy is to know the key mechanism of cytotoxic compounds that can kill cancer cells. Ramie (Boehmeria nivea L. Gaud.) leaves contain active compounds that have important effects on cancer chemoprevention. Objective: To obtain the active fraction of a Ramie leaf extract in inhibiting the proliferation of MCF-7 breast cancer cell lines and to determine the mechanism of apoptosis induction using MCF-7 and Saccharomyces cerevisiae strains 1140, 1353, and 1138. Method: Fractions were prepared using n-hexane, dichloromethane (CH2Cl2), ethyl acetate, and n-butanol as solvents. All fractions were tested qualitatively through phytochemical. The MTT-based cytotoxicity assay used MCF-7 (in vitro) to obtain the IC50 value, whereas the model system that targets the enzymatic (topoisomerase) used a yeast-based bioassay to obtain the IC12 value. Apoptotic induction of the active fraction in MCF-7 was performed using flow cytometry and qPCR (2-ΔΔCt method). Results: The phytochemical analysis indicated that the extract fraction consisted of alkaloids and steroids. The smallest IC50 value was obtained from the CH2Cl2 fraction as 3.79 g/mL, potentially act as an anticancer. A higher percentage of apoptosis than that of necrotizing cells and live cells was observed through flow cytometry. The CH2Cl2 fraction with an IC12 value < 8000 in strains 1140, 1353, and 1138 consistently showed the mechanism of apoptosis induction as topoisomerase I and II inhibitors. Also, another mechanism could be through the intrinsic pathway, indicated by the highest expression level in p53. Conclusions: The CH2Cl2 fraction of Ramie leaves can inhibits the proliferation of MCF-7 cells in the active and strong categories. The CH2Cl2 fraction induces apoptosis by increasing p53 gene expression and inhibiting topoisomerase I and II. Thus, it showed potential as an anticancer drug candidate.

Background: Otomycosis affects about 9% of patients with otitis externa. One of the predisposing factor is impacted cerumen.  Earphone usage causes cerumen impaction. In the light of the recent COVID-19 pandemic, people are housebound due to prolonged lockdown. Hence people are more technology dependent as working from home and studying online has become the norm. Therefore, usage of earphones has proportionately increased, and the impacted cerumen cases have increased. Methods: This cross-sectional study aimed to analyze data from questionnaire to assess the prevalence of otomycosis among patients with impacted cerumen due to excessive earphone usage, establish associations between otomycosis and symptoms such as pain and hearing loss as well as the correlation between fungal growth and the long hours of earphones usage. The relationship between the age of participants and the usage of earphones during the pandemic was also explored. The data was collected from 100 individuals aged 14 to 51 years who sought treatment for symptomatic impacted wax at an ENT clinic in Malaysia. Results: The mean age of participants was 25.9 years. There was a 31% prevalence of otomycosis among these patients. Otalgia was present in 60% of patients with otomycosis (t value 2.94, coefficient 0.27). Approximately 37% of patients had a large air-bone gap indicating conductive hearing loss. There was an insignificant correlation between otomycosis and the longer hours of earphones usage (t value 1.51, coefficient 0.00015). No correlation was found between age of participants and the total hours of earphone usage (t value 0.63, coefficient 0.0012). Conclusion: This study offers initial evidence that earphone usage could be a predisposing factor in developing otomycosis. However, there was no evidence of longer hours of earphones usage increasing the chances of developing otomycosis. Additionally, symptoms like otalgia and conductive hearing loss could be present in both earwax impaction and otomycosis.

Introduction: Curcumin is a polyphenol that has pharmacological activity that can inhibit tumor cell growth and induce apoptosis through various mechanisms. However, the specific mechanism of curcumin cytotoxicity remains controversial because of many anti- and pro-apoptotic signaling pathways in various cell types. This study aims to examine the relationship among curcumin on RASSF1A, Bax protein levels, and caspase-3 activity in supporting the apoptotic mechanism in CSA03 breast cancer cells. Method: Curcumin administration to cancer cells is based on differences in dosage with 24-hour incubation. Cytotoxicity after curcumin administration was determined using MTS. RASSF1A and Bax protein levels were tested through ELISA. Caspase-3 activity was used to determine apoptosis and was tested using flow cytometry. Results: The results indicated that curcumin had a cytotoxicity effect of 40.85 µg/mL. At concentrations of 40 µg/mL and 50 µg/mL, curcumin increases levels of protein RASSF1A (∆ = 26.53% and 47.35%, respectively), Bax (∆ = 48.79% and 386.15%), and caspase-3 (∆ = 1,678.51% and 1,871.889%) significantly. Conclusions: Curcumin exhibits anti-proliferative activity and apoptotic (Caspase-3) effects through activation of RASSF1A and Bax.

Covid-19 is a new infectious viral illness. The first one appeared in Wuhan, and within two months, it became a pandemic. Medical diagnosis is confirmed by fever, cough, shortness of breath, combined with neutrophil ratio lymphocyte analysis and chest x-ray or chest C.T. radiology imaging, with a ground-glass appearance. C.T. scans are not widely available in hospitals in Indonesia. Many hospitals only own x-ray for covid-19 as radiologic diagnostic imaging. With digital imaging capabilities, Due to the similarity of applications such as the radiological workstation, Adobe Photoshop Express will improve the capacity to diagnose Covid-19 from a chest x-ray. Adobe Photoshop Express has outstanding digital processing capabilities to enhance the presentation of images so that the efficiency of diagnosing plain x-ray thorax image cases with Covid-19 becomes easier and more manageable.

COVID - 19 is an issue ravaging the whole world. Numerous deaths have been recorded particularly elderly individuals and the most vulnerable. In Nigeria, the case isn't extraordinary. The aim of the study was to quantify the number of cases reported in Nigeria. To this effect, the global literature cited in the Nigeria Centre for Disease Control (NCDC) website, the WHO COVID-19 database, other expert-referred scientific articles, and bibliographic databases were used. The results show that as at the sixteenth of May 2020, 288 new affirmed cases and 3 deaths were recorded in Nigeria. Till date, 5445 cases have been affirmed, 778 cases have been released and 171 deaths have been recorded in 34 states and the Federal Capital Territory, While Lagos State is for the most part influenced with 36 deaths in comparison with the cases revealed by the World Health Organization (4,425,485 cases affirmed, 89,269 new cases and 302,059 deaths). The death rate in Nigeria is about 0.05% of the global deaths. The explanation could be because of high temperature (> 30 oC), the intake of garlic, ginger, honey, and heated water by the individuals, and to stop the spread, the Nigerian government has implemented the utilization of nose cover, social distancing, and semi-lockdown of the towns and urban communities. Like HIV and Laser fever this pandemic will be an issue of the past when the adequate vaccine is made available.

Pterygium is a degenerative and invasive disorder of the subconjunctival tissue that can reach the cornea area which is become a worldwide health problem. UV exposure is the biggest risk factor for pterygium. A profession as a fisherman is one of the occupations that are often related to the appearance of pterygium, because of long-term exposure to UV rays. This study aimed to determine the correlation between age and duration as a fisherman with the degree of pterygium in Kedung Cowek village, Bulak Surabaya district, East Java - Indonesia. The research design was an analytic observational using consecutive sampling during March - July 2011; with the total number of respondents is 42 fishermen who met the inclusion criteria. The results of the statistical analysis test showed that there was a strong and significant relationship between the age of the fishermen (p = 0.007; coefficient correlation = 502) and the duration of working as a fisherman (p = 0.029; coefficient correlation = 409) with the severity of the pterygium. Based on these results it can be concluded that the age and duration of work as fishermen are related to the severity of the pterygium in fishermen in the Kedung Cowek sub-district, Surabaya, East Java-Indonesia.
