Abstract The aim of this study was to evaluate the efficacy of two probiotic strains Bifidobacterium longum subsp. infantis CECT7210 (Laboratorios Ordesa S.L.) and Lactobacillus rhamnosus HN001 combined or not with a prebiotic mixture of oligofructose and inulin against Salmonella typhimurium. A total of 96 piglets (28 days) were distributed into 32 pens assigned to 5 treatments: one non-challenge (control diet, CTR+) and four challenged: control diet (CTR-), or supplemented with probiotic (>3x1010cfu/kg each, PRO), prebiotic (5%, PRE) or their combination (SYN). After one adaptation week, animals were inoculated with Salmonella. Feed intake, weight and clinical signs were recorded. On days 4 and 8 post-inoculation (PI), one animal per pen was euthanized and samples collected for Salmonella counts, fermentation products, ileal histomorphology and serum inflammatory markers. After the challenge, feed intake was decreased (P = 0.001) more markedly in the SYN group with a lower final weight (P = 0.009). PRE and SYN groups showed more liquid ileal consistency on day 8PI (P = 0.009). A higher percentage of animals receiving PRO became negative to Salmonella in feces at the end of the study (65% PRO vs 0% CTR-, P = 0.028). Animal receiving PRE did not reach countable levels in feces any time during the trial (vs 23.2% for CTR-group, P = 0.013). Histologically, on day 4PI a decrease in the ileal villous/crypts ratio was found in challenged animals (P < 0.001), being PRO able to improve the recovery from this damage at day 8PI (P = 0.008). Animals receiving PRE had higher number of intraepithelial lymphocytes (IEL) on day 8PI (P = 0.028). In conclusion, the probiotic strains were able to promote a faster clearance of the pathogen from the gut with a faster recovery of damaged histological architecture. The prebiotic reduced Salmonella colonization and stimulated the local immune response. However, these beneficial effects disappeared when they were administered in a synbiotic form.

We evaluated the potential of multi-strain probiotic (Bifidobacterium longum subsp. infantis CECT 7210 and Lactobacillus rhamnosus HN001) with or without galacto-oligosaccharides against enterotoxigenic Escherichia coli (ETEC) F4 infection in post-weaning pigs. Ninety-six piglets were distributed into 32 pens assigned to five treatments: one non-challenged (CTR+) and four challenged: control diet (CTR−), with probiotics (>3 × 1010 CFU/kg body weight each, PRO), prebiotic (5%, PRE), or their combination (SYN). After 1 week, animals were orally inoculated with ETEC F4. Feed intake, weight, and clinical signs were recorded. On days 4 and 8 post-inoculation (PI), one animal per pen was euthanized and samples from blood, digesta, and tissues collected. Microbiological counts, ETEC F4 real-time PCR (qPCR) quantification, fermentation products, serum biomarkers, ileal histomorphometry, and genotype for mucin 4 (MUC4) polymorphism were determined. Animals in the PRO group had similar enterobacteria and coliform numbers to the CTR+ group, and the ETEC F4 prevalence, the number of mitotic cells at day 4 PI, and villus height at day 8 PI were between that observed in the CTR+ and CTR− groups. The PRO group exhibited reduced pig major acute-phase protein (Pig-MAP) levels on day 4 PI. The PRE diet group presented similar reductions in ETEC F4 and Pig-MAP, but there was no effect on microbial groups. The SYN group showed reduced fecal enterobacteria and coliform counts after the adaptation week but, after the inoculation, the SYN group showed lower performance and more animals with high ETEC F4 counts at day 8 PI. SYN treatment modified the colonic fermentation differently depending on the MUC4 polymorphism. These results confirm the potential of the probiotic strains and the prebiotic to fight ETEC F4, but do not show any synergy when administered together, at least in this animal model.

Several cases of animal and human salmonellosis caused by the Salmonella serotype Typhimurium have been reported. In animals, subclinical infection favors pathogen dissemination through feces. In this context, the domestic pigeon (Columba livia) with an asymptomatic condition may play an important role in the transmission of salmonellosis, through the elimination of contaminated feces in commercial aviaries or in poultry feed facilities, causing economic losses to the poultry industry and presenting a risk to public health. This study aimed to evaluate the mortality, clinical signs and the presence of Salmonella Typhimurium in the feces and organs of chicks previously inoculated with bacteria isolated from a pigeon. One-day-old chicks were distributed in two experimental groups (G1 and G2) of 32 birds each, and a control group of six birds. Two inocula of 0.4 and 0.7 mL with 105 and 106 colony forming units were used in G1 and G2 birds, respectively. At 1, 4, 7 and 14 days post-inoculation (dpi) fecal samples were pooled from each cage and individual cloacal swabs were collected. At 14 dpi, all chicks were euthanized and samples were collected from the liver, spleen, lung, cecum and intestine for microbiological analysis. Mortality was only observed among G2 birds (6.25%). Most birds presented clinical signs of diarrhea at 4 dpi and no symptom as observed at 14 dpi. The results from cloacal swabs demonstrated bacterial elimination in 68.8% and 53.1% of G2 and G1 birds, respectively at 1 dpi. Additionally, fecal samples had elevated bacterial shedding in all four periods of observation , with a higher excretion at 4 dpi (62.5%) for both groups. Among G2 birds, 74.2% were positive for the pathogen in the intestine; G1 birds presented the lowest rate of lung infection (29%), and both groups had more than 50% positivity for liver and caeca. The results revealed that infected chicks with a Salmonella Typhimurium strains isolated from pigeons may host the pathogen in several organs, and simultaneously present diarrheic disorders with significant levels of bacterial excretion in feces.

Abstract Lactating sows (N=140, York x Landrace) were used to evaluate the effects of a liquid prototype (LIQP) and dry (XPC®; Diamond V) Saccharomyces cerevisiae feed additives on sow and litter performance. Sows were fed a common gestation diet (0.55% SID-Lysine) until d112 of pregnancy and then allotted to lactation treatments: 1) Control diet (CON; 1.00% SID-Lysine), 2) CON +15 mL of LIQP from d112 to weaning (LIQ), 3) CON +0.20% of XPC from d112 to weaning (DRY), and 4) DRY +15 mL of LIQP from d112 to d7 post-farrowing (D+L). The LIQP was given once daily using an oral dose gun and XPC was included in the feed. Immunoglobulin concentrations were estimated on colostrum samples using Brix refractometer and piglet d 1 serum immunocrit ratio and plasma IgA and IgG. Daily sow water intake and daily feed intake (DFI) from d112 of gestation to d7 post-farrowing and weekly feed intake (ADFI) were recorded. There were no treatment effects on sow BW, backfat, or loin depth (P >0.05) although sows from LIQ group had numerically smaller BW reductions compared to CON sows (4.9% vs 7.2%, P=0.19). LIQ sows had greater DFI and CON lower DFI during week 1 of lactation (P=0.04) as well as ADFI for weeks 2, 3, and overall lactation period (P< 0.01) with DRY and D+L sows being intermediate. Water intake, immunoglobulins, and litter performance did not differ among treatments (P >0.05) although pigs from LIQ sows, compared to CON, had numerically increased weaning (6.14 vs 5.82kg, P >0.05) and litter weights (63.2 vs 60.1kg, P >0.05). LIQ sows had 0.7 d reduced wean-to-estrus interval (P< 0.001) and tended to have greater conception rates (P=0.07). In conclusion, LIQ supplementation of lactating sows improves feed intake, allowing sows to keep body reserves, and have better subsequent rebreeding performance.

Abstract This study evaluated the effects of a liquid prototype (LIQP) and dry (XPC®; Diamond V) Saccharomyces cerevisiae feed additives on blood and uterine cytokine profiles in sows (n = 40). On d 112 of pregnancy sows were allotted to dietary treatments: 1) Control diet (CON), 2) CON +15 mL of LIQP (LIQ), 3) CON +0.20% of XPC (DRY), and 4) DRY +15 mL of LIQP until d 7 post-farrowing (D+L). Interleukin (IL)-1β, IL-4, IL-6, IL-8, IL-10, tumor necrosis factor-α (TNF-α), and interferon-γ (IFN-γ) were quantified from serum samples collected on d 112 of gestation, d 2 and 6 post-farrowing, and from uterine fluid collected on d 2, 4, and 6 post-farrowing. Serum C-Reactive protein (CRP) and haptoglobin concentrations were evaluated. No interactions between treatments and day of collection were observed (P > 0.13). LIQ and D+L sows had the greatest serum IL-10 concentration (P < 0.001) and sows fed CON tended to have lower concentration of IL-8 (P < 0.06) vs. other treatments. Serum CRP concentrations were greatest on d 2 (P < 0.001), serum IL-10 (P < 0.04) and IL-4 (P < 0.07) linearly decreased while serum haptoglobin (P < 0.02) and INF-γ (P < 0.001) linearly increased post-farrowing. In the uterine fluid, LIQ and D+L sows had greater INF-γ (P = 0.04) concentrations and CON tended to have the least concentration of TNF-α (P = 0.08). Uterine fluid IL-1 tended to linearly increase (P < 0.07) and IL-6 linearly decrease (P < 0.01) post-farrowing. No strong correlations were detected between cytokines in the serum and uterine fluid within day. LIQ sows had the greatest daily feed intake and CON the least during the first week of lactation (P = 0.04). Providing LIQP post-farrowing to sows modified immune response increasing both pro- and anti-inflammatory cytokines in serum and uterine fluid in an independent manner, allowing animals a quicker recovery and increased feed intake.

Abstract Background The search for alternatives to antibiotics in pig production has increased the interest in natural resources with antimicrobial properties, such as medium-chain fatty acids (MCFA) as in-feed additives. This study evaluated the potential of a novel blend of MCFA salts (DIC) from distilled coconut oil with a lauric acid content to reduce enteropathogens and control intestinal diseases around weaning. Two experimental disease models were implemented in early-weaned piglets, consisting of two oral challenges: Salmonella Typhimurium (1.2 × 108 CFU) or enterotoxigenic Escherichia coli (ETEC) F4 (1.5 × 109 CFU). The parameters assessed were: animal performance, clinical signs, pathogen excretion, intestinal fermentation, immune-inflammatory response, and intestinal morphology. Results The Salmonella challenge promoted an acute course of diarrhea, with most of the parameters responding to the challenge, whereas the ETEC F4 challenge promoted a mild clinical course. A consistent antipathogenic effect of DIC was observed in both trials in the hindgut, with reductions in Salmonella spp. plate counts in the cecum (P = 0.03) on d 8 post-inoculation (PI) (Salmonella trial), and of enterobacteria and total coliform counts in the ileum and colon (P < 0.10) on d 8 PI (ETEC F4 trial). When analyzing the entire colonic microbiota (16S rRNA gene sequencing), this additive tended (P = 0.13) to reduce the Firmicutes/Bacteroidetes ratio and enriched Fibrobacteres after the Salmonella challenge. In the ETEC F4 challenge, DIC prompted structural changes in the ecosystem with increases in Dialister, and a trend (P = 0.14) to increase the Veillonellaceae family. Other parameters such as the intestinal fermentation products or serum pro-inflammatory mediators were not modified by DIC supplementation, nor were the histological parameters. Only the intraepithelial lymphocyte (IEL) counts were lowered by DIC in animals challenged with Salmonella (P = 0.07). With ETEC F4, the IEL counts were higher with DIC on d 8 PI (P = 0.08). Conclusions This study confirms the potential activity of this MCFA salts mixture to reduce intestinal colonization by opportunistic pathogens such as Salmonella or E. coli and its ability to modulate colonic microbiota. These changes could explain to some extent the local immune cell response at the ileal level.

This study aimed to evaluate the liver, kidney damage caused by S. typhimurium and to estimate the oxidative damage in association with this bacteria. A highly virulent isolates of S. typhimurium were obtained from the department of internal and preventive medicine/ College of Veterinary Medicine/ University of Baghdad. A twenty five local rabbits of both genders with age range (2-4 months) weeks old were used for this study, the rabbits were divided randomly into five groups each group contains 5 rabbits :- group 1: drenched orally with 5 ml of normal saline and consider as control group, group 2: were drenched orally with (5 ml) suspension which contain (5��109 CFU) of Salmonella typhimurium and regarded as infected group, group 3 were drenched orally with (5 ml) suspension which have (5��109 CFU) of Salmonella typhimurium then treated with a single dose of gentamicin alone at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation), group 4 were drenched (5 ml) suspension having (5��109 CFU) of Salmonella typhimurium then treated with a single dose of Ca-EDTA alone at 40mg/kg orally after presence of signs (after 24hrs. post inoculation) and group 5 were drenched (5 ml) suspension that contain (5��109 CFU) of Salmonella typhimurium then treated with a single dose of combined gentamicin at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation) and Ca-EDTA 40mg/kg after presence of signs (after 24hrs. post inoculation).The results of biochemical profile showed a significant increase (p less than 0.05) in ALT, creatinine and urea levels in infected group as compared with control group, while, the treated groups especially group 5 showed a significant improvement in ALT, Urea and creatinine levels which returned to relative normal levels as compared with infected group after 96hrs. post treatment. Also, the results of oxidative stress showed a significant increase in the levels of MDA in G2, G3, G4 and G5 after 48 hrs. post treatment, while the level of GSH showed a significant decrease in the level at 48hrs., both were returned to relative normal levels after 96hrs.post treatment especially in group 5.In conclusion, S. typhimurium can causing liver and kidney damage which is manifested by increase ALT, Urea and Creatinine. Also, MDA and GSH is increased due to salmonellosis.

ABSTRACT   EFFECT OF UTILIZATION OF VIRGIN COCONUT OIL (VCO) IN THE DIET ON PIG PERFORMANCE. Utilization of fats and oils in pig diets is of great importance due to their high energy value. VCO is obtained by cold press processing of the kernel from the coconut fruit. Utilization of virgin coconut oil (VCO) in the diets on pig performance.  The present study was designed to elaborate the effect of utilization of VCO in the diets on energy and protein digestibility of growing pigs. The experiment was conducted using 20 castrated male pigs aged 1.5 - 2.0 months weighing 12,0±2,0 kg. The data were analyzed according to the linear model procedure for ANOVA appropriate for Randomized Block Design with 5 treatments and 4 replications. Treatments were formulated as follow: R0 = 100% control diet + 0% VCO; R1 = 99.5% control diet + 1.0% VCO; R2 = 98.0% control diet + 2.0% VCO; R3 = 97.0% control diet + 3.0% VCO; and R4 = 96.0% control diet + 4.0% VCO. Parameters measured were: daily feed intake, daily gain, daily water consumption. The results showed that the utilization of VCO up to 4% in the diets had no significant effect (P > 0.05) on daily feed intake, daily gain, daily water consumption of pigs. It can be concluded that the addition of VCO up to 4.0% in the diets has no significant meaning on pig performance.   Key words: Virgin coconut oil (VCO), Performance, Growing pigs

Abstract Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically-important disease and ingestion of soy isoflavones (ISF) may benefit PRRSV-infected pigs due to demonstrated anti-inflammatory and anti-viral properties. The objective of this study was to quantify long-term effects of ISF consumption on fecal microbiome characteristics under disease challenge. In total, 96 weaned barrows were group-housed in a BSL-2 containment facility and allotted to 1 of 3 experimental treatments that were maintained throughout the wean-to-finish study: non-infected pigs receiving an ISF-devoid control diet (NC, n=24), and infected pigs receiving either the control diet (PC, n=36) or that supplemented with total ISF in excess of 1,600 mg/kg (ISF, n=36) (Table 1). Following a 7-day adaptation, pigs were inoculated intranasally with either a sham-control (PBS) or live PRRSV (1×105 TCID50/mL, strain NADC20). Fecal samples were collected from 48 individual pigs at pre-infection (-2 days post-inoculation, DPI), peak-infection (10 DPI), and post-infection (144 DPI) time-points and extracted DNA was used for 16S bacterial rRNA sequencing. Differences in bacterial communities among diet groups were evaluated using UniFrac distance matrices (weighted and unweighted) in QIIME. All other data were analyzed by one-way ANOVA performed on transformed data using R. Across all time-points, only minimal differences were observed due to ISF alone. At 10 DPI, PRRSV infection reduced Prevotella 9 genera abundance from approximately 20% to less than 10%, but the specific function of this variety in pigs is unclear. The most notable finding was decreased relative abundance of Actinobacteria at 144 DPI between non-infected and infected treatments (P &lt; 0.05), which is consistent with various dysbioses observed in other disease models. Our findings indicate that differences present were mainly due to PRRSV infection and not strongly influenced by ISF ingestion, which implies previously observed performance benefits conferred by dietary ISF are not likely due to changes in microbiome composition.

Abstract A total of 109 sows (Line 241; DNA, Columbus, NE) were used in a study to evaluate the effect of increasing phytase concentration in lactation diets on sow and litter performance. On d 107 of gestation, sows were blocked by body weight and parity and allotted to 1 of 3 dietary treatments of increasing phytase concentration (0, 1,000, or 3,000 FYT/kg; Ronozyme HiPhos 2700; DSM Nutritional Products, Inc., Parsippany, NJ). The control diet contained no phytase and was formulated to contain 0.50% standardized total tract digestible phosphorus (STTD P; 0.45% available P) and 0.62% STTD calcium (0.90% total Ca). The same STTD P and Ca concentrations were formulated for the phytase diets considering a release of 0.132 STTD P and 0.094 STTD Ca in both diets. Diets were fed from d 107 of gestation until weaning (d 18 ± 2). Litters were cross-fostered within treatment until 48 h post-farrowing to equalize litter size. Linear and quadratic response to phytase concentration was evaluated using the lmer function in R. There was no evidence for difference in sow body weight change, farrowing performance, wean-to-estrus interval, or litter size among dietary treatments. Sow average daily feed intake from farrowing to weaning tended to increase (linear, P=0.093) as phytase increased. Although not significant (linear, P =0.226), farrowing duration decreased as phytase increased. Litter weaning weight increased (quadratic, P=0.039) and overall litter gain increased (quadratic, P=0.047) with 1,000 FYT of phytase. In summary, sow feed intake tended to increase linearly with increasing phytase; however, feeding 1,000 FYT/kg maximized overall litter gain and weaning weight. This small-scale study suggests sow and litter performance benefits due to high inclusions of dietary phytase; however, a commercial trial with more sows is warranted to confirm these results.

Summary Objective: In the present study various tissues of pigs were investigated for the presence of histopathologic lesions after an experimental infection with Haemophilus (H.) parasuis serovar 5. Material and methods: Conventional pigs (n = 36) were divided into a control group B (n = 9) and a challenge group A (n = 27), which was infected intratracheally. Pigs that did not die prior to study termination were euthanized on day 14 post inoculation. Postmortem samples of the lung, heart, liver, kidney, spleen, left tarsal joint capsule and brain were collected. Results: All but one pig with detectable histopathologic lesions (n = 11) showed typical macroscopic changes. Histopatho logic examination of all tissue samples identified pyelitis (n = 10), synovitis (n = 7) and meningitis (n = 7) and all those animals were euthanized prior to study termination. No histopathologic lesions were found in pigs of the control group. The correlations between pyelitis and meningitis, pyelitis and synovitis and synovitis and meningitis were significant (p < 0.001). No significant correlation could be observed between the histopathologic and the clinical examination of the joints. The investigation of samples from the joints by PCR was not significantly correlated with the observed synovitis. The clinical observation of neurologic signs was significantly correlated with meningitis (p = 0.03). A significant correlation (p < 0.001) could be detected between meningitis and the detection of H. parasuis by PCR in brain samples. Conclusions: H. parasuis constantly causes clinical signs and pathologic lesions as soon as it infects the brain while it can infect the joints without causing histopathologic lesions. Pigs with histopathologic lesions do not always show typical clinical signs. Only few studies described the finding of kidney lesions in pigs with Glässer’s disease and this is the first study to describe a pyelitis in pigs experimentally infected with H. parasuis. The observed pyelitis mainly occurred in acute cases.
