The aim of our scientific work was to study the effect of exogenous melatonin on the indicators of fibrinolytic processes in the heart tissues and to analyze the changes in fibrinolytic activity occurring in the heart tissues of enucleated hyper- and hypothyroid white rats. The experiments were carried out on white nonlinear male rats weighing 0.12-0.14 kg. 4 experimental groups of animals were formed. The control group consisted of 11 normothyroid rats, which were injected with a melatonin solution in appropriate volumes. The animals were euthanized under light ether anesthesia. Samples of the examined heart tissue were ground in a glass homogenizer with borate buffer (pH 9.0). The homogenate was used in biochemical analysis. Determination of the total, enzymatic and non-enzymatic fibrinolytic activity (TFA, EFA, NFA, respectively) in the heart tissues was carried out by the method of azofibrin lysis (LLC "Simko", Ukraine). Statistical processing of the results was carried out by the method of variation statistics using the Student's test. Experimental studies on nonlinear male white rats have shown that the introduction of exogenous melatonin, as well as modeling the conditions of endogenous melatonin overproduction by blinding (enucleation) cause an increase in the intensity of enzymatic and non-enzymatic fibrinolysis in heart tissue in normotyroid animals. At other hand, enucleation causes an increase in the intensity of fibrinolysis in the tissues of the heart in hypothyroid rats, but not in hyperthyroid.

Introduction. Toxic hepatitis (TH) is a complex and multifaceted disease, the development of which is mediated by a complex of biochemical and molecular genetic interactions. The current understanding of the pathogenesis of TH and, as a consequence, its treatment is based on standardization of the phenotype of the disease, often without taking into account metabolic disorders within the cells. Material and methods. experimental studies were performed on white outbred male rats weighing 200-220 g. A 50% solution of TCM was used as a toxicant. Biochemical studies were performed on a laboratory medical photometer “Stat Fax 3300” using clinical test kits and control materials manufactured by Vector-Best LLC. Liver tissue for histological examination was subjected to the standard histological procedure and paraffin embedding. Sections 5-7 μm thick were stained with hematoxylin-eosin. Gene expression analysis was performed using real-time PCR amplification on a RotorGene instrument (QIAGEN). Statistical processing of experimental data was performed using the Pearson correlation coefficient and one-way analysis of variance (ANOVA). The results were considered reliable at p <0.05. Results. As a result of the analysis of the correlation of the expression of the studied genes and the level of biochemical parameters, it was found that the correlation of the expression of the Nfe2l2 and Gstm1 genes was r = 0.812 (p = 0.0001). The dynamics of gene expression of Chek, Gstm1, Gstp1, Nfe2l2, had a negative correlation with the level of AST activity in blood serum. And the expression of the genes Chek, Gclc, Gstm1, Nfe2l2, Ripk, Sod1 with an index of ALT activity in the blood serum. After 72 hours, the expression of almost all of the studied genes became multidirectional. And the correlation between indices is often not determined. An analysis of the relationship between the level of cytolysis enzymes and the correlation level of the studied genes showed that after 72 hours the correlation was observed in the Gstm1, Hmox, and Sod1 genes with the levels of AST and ALT.

The aim of this research was to know whether beetroot (Beta Vulgaris) extract could protect spermatogenesis by maintaining spermatogenic and sertoli cell count  in rats (Rattus novergicus) induced with CCl4. Rats were given beetroot extract daily, for 14 days and 3ml/kg BW CCl4 intraperitoneally one hour after last treatment. This study used twenty rats which were devided equally into 5 groups. K(-), the negative control group was not induced with CCL4 and only given 1% CMC-Na suspension. K(+), the positive control group was induced with CCl­4 and given 1% CMC-Na suspension. P1, P2 and P3 were given beet root extract with doses of 200, 400, and 800 mg/kg BW daily before feeding. All of the beetroot treatment were given orally (2 ml). After 24 hours CCl4 induction, rats were sacrificed and testis were collected to make histology slides. The observations showed significantly different (p<0,05) in all of variables. Result showed significant differences in spermatogenic and sertoli cells between K(-) and K(+) groups, K(+) with P2 and P3 group, and showed insignificant difference between P2 and P3 group in spermatogenic and sertoli cells. The result of this research showed that beetroot extract could protect the spermatogenic and sertoli cells in male rats induced with CCl4.

Introduction. In recent years, magnetic nanoparticles, which can simultaneously have a therapeutic effect on the pathological focus, are used to magnify contrast enhancement and increase diagnostic sensitivity during magnetic resonance therapy (MRT). The last is carried out by the effective capture of neutrons, which among all the chemical elements is most pronounced in gadolinium. The use of gadolinium nanoparticles encapsulated in a polymeric matrix allows increasing the bioavailability of nanoparticles, reduces the possible toxicity of drugs. Aim. Evaluation of impact of new nanocomposite magnetically active metal complex gadolinium system on the morphofunctional state of the nervous tissue, liver, and kidney of rats. Material and methods. Experimental studies of biological effects of gadolinium-arabinogalactan nanocomposite (Gd-AG) were carried out on rats that were injected intraperitoneally for 10 days at the dose of 500 μg/kg in 0.5 ml of saline. Animals were sacrificed by decapitation under light ether anesthesia the next day after the end of exposure. To perform pathological studies, frontal sections of the temporal-parietal zone of the sensorimotor cortex, liver and kidney tissues were stained on ordinary histological glass slides with hematoxylin and eosin for viewing microscopic picture. The immunohistochemical method was used to determine the activity of the bcl-2, caspase-3 and hsp70 modulatory protein in apoptosis of white rats in brain neurons and to study the biological response of the organism at the subcellular level. Results. Histological analysis of tissues revealed a pronounced compensatory response of liver, a violation of the functional activity of kidneys. A decrease in the total number of normal neurons per unit area in brain tissue and an increase in the number of acts of neuronophagy indicate the initial stage of neurodegenerative process. Evaluation of the intracellular metabolism of neurons has not established the presence of signs characteristic of apoptotic process. Conclusion. The subacute effect of Gd-AG in a dose of 500 µg/kg causes a disturbance of morphofunctional state of liver, kidneys and nervous tissue, as well as modulation of cellular proteomics.

This study aims to determine the content of flavonoid and the effect of ethanol extract of moringa seeds on the regeneration of pancreatic β cells in male white rats streptozotocin induced diabetes. This study method used has total flavonoid equivalent quercetin by spectrophotometry uv-vis and to regeneration of pancreatic β cells in male white rats used 30 test animals,namely male white rats divided into 6 groups, each group consisted of 5 male white rats with details of group I as normal control, Group II as negative control given 0.5% Na-CMC suspension, Group III as positive control given glibenclamide suspension and in Groups IV, V, and VI were given with each dose of 100 mg/kg BW, 200 mg/kg BW and 400 mg/kg BB. Histopathological damage picture of the pancreas was observed by staining HE using a 400x magnification olympus Cx21 microscope. The results showed that the ethanol extract of moringa seeds contained secondary metabolites, namely flavonoids, alkaloids, saponins and tannins. The results showed has total flavonoid equivalent quercetin of moringa seeds is 1,26% and regeneration of pancreatic β cells in male white rats streptozotocin induced diabetes of Moringa seed ethanol extract at a dose of 400 mg/kg BB can have an effect on the regeneration of β cells in the pancreas of white diabetic male rats.

Цель - исследовать интенсивность экспрессии белка альфа-синуклеина клетками Панета у крыс в норме и условиях длительного бактериофагального инфицирования микробиоты. Материал и методы. Экспериментальные исследования проведены на 12 половозрелых крысах-самцах линии Вистар массой 280-320 г. Крысам основной группы (n=7) еженедельно на протяжении 12 нед ректально вводили 0,5 мл раствора, содержащего смесь бактериофагов против 14 патогенов человека (Microgen, Россия). Однократная доза вводимой бактериофагальной смеси содержала 0,5×10ед./мл каждого фага. Животные контрольной группы (n=5) получали по аналогичной схеме 0,5 мл стерильного изотонического раствора натрия хлорида. После интракардиальной перфузии отбирали фрагменты проксимального отдела подвздошной кишки на уровне 1-3 см выше илеоцекального соединения. Серийные криостатные срезы кишечника использовали для окрашивания гематоксилином - эозином и выявления иммунопозитивного альфа-синуклеина при помощи моноклональных мышиных антител (Sigma-Aldrich, USA) и коммерческого набора, содержащего авидин-биотин-пероксидазный комплекс (АBC Elite; Vector Laboratories, Burlingame, CA). Результаты. Длительное бактериофагальное инфицирование приводило к значимому снижению относительно контроля количества клеток, иммунопозитивных к альфа-синуклеину. В клетках Панета значимо снижались площади, занимаемые иммунореактивным продуктом к альфа-синуклеину и ацидофильными гранулами. Выводы. В апикальных частях клеток Панета, в области локализации ацидофильных гранул детектируется иммунопозитивный альфа-синуклеин. Дисбиоз кишечника, вызываемый бактериофагальным инфицированием микробиоты, приводит к гранулярному истощению клеток Панета и снижению экспрессии в них иммунореактивного альфа-синуклеина, что свидетельствует о его вовлеченности в механизмы экскреции. Objective - to investigate the intensity of the alpha - synuclein expression by Paneth cells of rat in normal conditions and prolonged bacteriophagal infection of gut microbiota. Material and methods. Experimental studies were performed on 12 adult Wistar male rats weighting 280-320 g. The rats of the main group (n=7) received rectally a 0,5 ml of solution containing a mixture of bacteriophages directed against 14 human pathogens (Microgen, Russia). The solution was introduced weekly for a period of 12 weeks. Each dose of bacteriophagal mixture contained 0,5×10 units/ml of each phage. Animals of the control group (n=5) received 0,5 ml of sterile physiological saline according to the same scheme. After transcardial perfusion, specimens of proximal portion of ileum 1-2 cm upstream the ileocecal junction were obtained. Serial cryostat sections were used for hematoxylin and eosin staining and for detection of immunopositive alpha-synuclein by monoclonal mouse antibodies (Sigma-Aldrich, USA) and commercially available kit containing avidin-biotin-peroxidase complex (АBC Elite; Vector Laboratories, Burlingame, CA). Results. Prolonged bacteriophage infection led to a significant decrease in the number of alpha-synuclein immunopositive cells compared with control. The area of Paneth cells occupied by the alpha synuclein-immunoreactive product and acidophilic granules significantly reduced. Conclusions. Immunopositive alpha-synuclein was detected in the apical parts of Paneth cells, in the area of acidophilic granules localization. The intestinal dysbiosis caused by bacteriophage infection of microbiota led to granular depletion of Paneth cells and a decrease in the expression of immunoreactive alphasynuclein in them, which indicates its involvement in excretion mechanisms.

Submicroscopic studies of the respiratory part of the lungs after 2 and 6 hours after the experimental acute lung injury with hydrochloric acid established adaptive-compensatory and destructive changes in the components of the air-blood barrier.The aim of the study – to learn submicroscopic changes in the components of the air-blood barrier of the lungs in the early period after acute lung injury.Materials and Methods. The experiments were carried out on 30 white mature non-linear male rats weighing 200–220 g. The animals were divided into 3 groups: 1 – control group, 2 – hydrochloric acid damage after 2 hours, 3 – hydrochloric acid damage after 6 hour.Results and Discussion. In an experiment on mature white rats, a study was made of the submicroscopic state of the components of the air-blood barrier in the early periods after acute lung injury. It has been established that adaptive-compensatory and initial destructive changes of the alveolar epithelium and the walls of the hemocapillary take place at 2 o'clock in the experiment. The cytoplasm of respiratory epitheliocytes during this period of the experiment was focal-edematous and enlightened, organelles were destructively altered. For alveolocytes of type I, there was a significant swelling and clarification of the cytoplasm. During this period of the experiment, an increased number of actively phagocytizing macrophages appeared, which acquired a rounded shape, clearly contoured membranes of the cariolema, their invaginations were determined, and in the karyoplasm euchromatin predominated. In alveolocytes of type II, after 6 hours, the progression of destructive changes was established. For which there were peculiarity hypertrophied nuclei with deep invagination of the cariolema, in which there were few nuclear pores, locally expanded perinuclear space. In the edematous cytoplasm, organelles were found to be destructively altered.Conclusions. Acute damage to the lungs leads to a disruption of the ultrastructural organization of the air-blood barrier. Established adaptive-compensatory processes and signs of destructive changes in the alveolar epithelium and the walls of hemocapillaries, which leads to deterioration of gas-exchange processes in the lungs.

The research purpose was to analysis effect of giving mahkota dewa fruits (Phaleria macrocarpa) extract to epithelialization in incision wound of white rats (Rattus norvegicus). The method was randomized posted-only control group design. There were 30 male rats (Rattus norvegicus) grouped on control and treatment group. Control group divided into three groups (KK1, KK2, KK3) and also treatment group divided into three groups (KP1, KP2, KP3). Control group just given CMC 1% peroral without mahkota dewa fruits extract, the treatment group given mahkota dewa fruits extract 22.5 mg/kg body weight. The data was analyzed by Kruskall Wallis. Based on Kruskall Wallis test, obtained result that there was a significant difference (p<0.05) epithelialization variable with p value p = 0.000 between control and treatment group. And based on One-way Anova test, obtained result that there was a significant difference (p<0.05) with p value p =0.000 between control and treatment group. The conclusion of this research was giving mahkota dewa fruits (Phaleria macrocarpa) extract can increase epithelialization in incision wound of white rats (Rattus norvegicus). Keywords : Mahkota Dewa Fruits (Phaleria Macrocarpa) Extract, Epithelialization, Incision Wound

Sodium-containing preparations have antimicrobial, anti-inflammatory and wound-healing activity, which determines their use in veterinary medicine. This effect is due to a wide range of physiological effects of sodium ions on animals metabolism. The article presents the results of experimental studies of the irritant effect of sodium-containing drugs: known – 2 % glycetinate, its analogues – 2 % ovocid, 3 % ovocid, 1.5 % potassium-sodium drug (Na / K, 1.5%). Studies of irritant and skin-resorptive action of drugs were performed by immersing the tail of animals in a test tube with the substance on white rats weighing 250–270 g. Four groups of rats were formed with 3 animals each: rats of I group (control) tails were immersed in test tubes with 2 % glycetinate, animals of II group – in test tubes with 2% oocide, III group – in test tubes with 3 % oocide, and in IV group rats – with Na / K, 1.5% drug. The exposure time was 4 hours. The animals were subjected to clinical observations, assessing their general condition and the reaction of the tail’s skin. The effect of drugs on the morphological and biochemical parameters of the rats blood was studied. The study of the local irritant effect of the studied drugs in rats from I, II and III groups showed a moderate local irritant reaction, as evidenced by the appearance of redness and thickening of the skin, and in group IV animals appeared only slight redness on the skin of the tails. This indicates a weaker local irritant effect of potassium-sodium-containing drug (1.5% K, Na), compared with other studied drugs. According to hematological studies, in the rats’s blood of groups III and IV, a reliable increase, within normal limits, the number of leukocytes and a decrease in hemoglobin content, compared with those of animals of group I. It was found that at 4 hours of the experiment under the action of the studied potassium-sodium drugs increases the permeability of Sodium and Potassium through the rats’s skin into the blood. In particular, in serum of animals from groups II and IV there was a reliable increase of Potassium content by 2.1 times, respectively, and a significant increase in Sodium content in the blood of animals from groups III and IV, compared with control group. In animals of groups II and IV there was a reliable increase of serum aspartate aminotransferase activity, and in rats of group III – increase of serum alanine aminotransferase, alkaline phosphatase activity and in triacylglycerol’s content, compared with rats of group I, which indicates the effect of 2 % glycecin analogues on metabolic processes in animals.

ABSTRACTSawilangit leaves (Vernonia Cinerea (L.) Less) is a plant that contains flavonoids as an antipyretic. The purpose of this study was to determine the effectiveness of flavonoid compounds from ethanol extract of sawilangit (Vernonia cinerea (L.) Less) leaves as an antipyretic in male white rats Wistar strain (Rattus norvegicus) induced by DPT vaccine. A total of 15 male rats were divided into 5 treatment groups, namely the negative control group (CMC 1%), the positive control group (paracetamol), and the ethanol extract group of sawilangit leaves with a dose of 200 mg, a dose of 400 mg, a dose of 800 mg. Each mouse was induced intramuscularly with a DPT vaccine at a dose of 0.3 ml. Temperature measurements were carried out before and after induction, then every 30 minutes after giving the test material from the 30th minute to the 180th minute. Data were analyzed statistically with One Way ANOVA and continued with LSD test to see significant differences between treatments. The results showed that the ethanol extract of sawilangit leaves had an antipyretic effect on male white rats Wistar strain..Key words: Antipyretics, Vernonia Cinerea, male white rats ABSTRAKDaun Sawilangit  (Vernonia Cinerea (L.) Less) adalah salah satu tanaman yang mengandung zat Flavonoid sebagai antipiratik. Tujuan penelitian ini untuk mengetahui adanya efektivitas senyawa flavonoid dari ekstrak etanol daun sawilangit (Vernonia cinerea (L.) Less) sebagai antipiretik pada tikus putih jantan galur wistar (Rattus norvegicus) yang diinduksi vaksin DPT. Sebanyak 15 ekor tikus putih jantan dibagi 5 kelompok perlakuan yaitu kelompok kontrol negatif (CMC 1%), kelompok kontrol positif (parasetamol), dan kelompok ekstrak etanol daun sawilangit dosis 3,6 mg, dosis 7,2 mg, dosis 14,4 mg. setiap tikus diinduksi vaksin DPT dosis 0,3 ml secara intramuscular. Pengukuran suhu dilakuan sebelum dan sesudah induksi, kemudian setiap 30 menit  setelah pemberian bahan uji dari menit ke-30 sampai menit ke-180. Data dianalisis secara statistika dengan One Way ANOVA dan dilanjutkan uji LSD untuk melihat beda nyata antar perlakuan. Hasil penelitian menunjukan bahwa Ekstrak etanol daun sawilangit mempunyai efek antipiretik terhadap tikus putih jantan galur wistar. Kata kunci: Antipiretik, Vernonia Cinerea, tikus putih jantan

AbstractBackground : Diabetes mellitus is the most common metabolic syndrome in the whole world with an incidence of 1-8%. This disease arises when insufficient insulin is produced or insulin cannot function properly.The ability of coriander juice water extract is expected to reduce blood glucose levels because the action is similar to insulin and can stimulate insulin release. Purpose : Knowing effectiveness coriander extract (Coriandrum sativum L.)to decline blood sugar levels in male white rats (Rattus norvegicus L.) wistar strain induced by alloxan. Method : This is a pure experimental study (true-experiment) using a pre and post test design with control group design using subjects namely white rats (Rattus norvegicus L.) male Wistar strain aged 10-12 weeks with a weight of 150-200 grams. By looking at changes in blood sugar levels during administration of alloxan and administration of coriander extract. Results : There were significant changes in some groups induced alloxan (p <0.05), but the effect of coriander extract was not very significant (p> 0.05). Conclusion : Coriander extract can control blood sugar levels to be stable or normal, but it takes longer and more research.  Keywords : coriander extract, blood sugar levels, alloxan
