Background. The problem of non-drug use of opioid drugs occupies a significant place among the current problems of world medicine. Objective. Тo study the hematological parameters of coagulation hemostasis in the norm and the dynamics of their changes at different times of opioid exposure. Methods. The experimental study was performed on sexually mature, outbred male rats in the number of 80 animals, weighing 160-270 g, aged 4.5-7.5 months. Animals were injected intramuscularly with “Nalbuphine” once daily for one day (10-11 hours in the morning) for 98 days. The initial dose of nalbuphine during the first 2 weeks was 0.212 mg / kg, the next 2 (II - IV weeks) - 0.225 mg / kg, the next (IV - VI weeks) - 0.252 mg / kg, the next (VI - VIII weeks) ) - 0.260 mg / kg, the next (VIII - X weeks) - 0.283 mg / kg, the next (X - XII weeks) - 0.3 mg / kg, and during (XII - XIV weeks) - 0.454 mg / kg. Thus, the conditions for chronic opioid exposure were created. Animals are divided into 3 groups. The 1-st group of animals received Nalbuphine for 98 days, with subsequent collection of material (end of 2 weeks, 4 weeks, 6 weeks, 8 weeks, 10 weeks, 12 weeks and 14 weeks of experimental opioid exposure); The 2-d was the control group, which for 98 days received injections of saline intramuscularly in one period of time (10 - 11 o'clock in the morning). Blood sampling and study of hematological parameters of blood (platelet count, prothrombin time, prothrombin index, time of recalcification of stabilized blood, total fibrinogen, determination of hemoglobin, hematocrit) were performed according to conventional methods. Software R v 4.0.3 and RStudio v 1.2.5042 were used for statistical calculations and graphing. MSOffice Excel 2010 spreadsheets were used to generate the final tables and store the data. Results. The key to the dynamics of changes in the blood parameters of experimental animals was week 6 of the experiment, as most indicators had the highest dynamics up to 6 weeks including further indicators of stability, which was higher (fibrinogen and prothrombin index) or less (prothrombin time, recalcification time and hemoglobin) indicators of the control group. The blood hematocrit of the experimental animals decreased evenly at all study terms to a minimum value at 14 week, and the number of platelets evenly all times increased to a maximum value at the last term of the experiment. This trend in all indicators was confirmed statistically. Conclusion. Our research has made it possible to study first and then observe the dynamics of changes in coagulation hemostasis and the degree of oxygenation of blood in acute, subchronic and chronic periods of experimental opioid exposure with subsequent statistical comparison.

BACKGROUND: Moringa stenopetala is used as nourishments, and treatment of various diseases. However, there is no much information on its safety. Hence, this study aimed to investigate the chronic administration of aqueous leaves extract of the plant.MATERIALS AND METHODS: Twenty-four rats were divided into: a control group administered with distilled water and three experimental groups, respectively, administered with the extract at doses of 500, 1000, and 2000 mg/kg orally for six months were investigated. Various hematological and biochemical parameters followed by histopathological analysis were evaluated.RESULTS: Treatment with the extract did not significantly affect most of the hematological parameters. However, there were a significant decrease of MCH at doses of 1000 mg/kg and 2000 mg/kg in male rats and increase of MCV at all doses in female rats. Levels of ALP at 2000 mg/kg and those of AST and ALT at 1000 and 2000 mg/kg were significantly increased in male rats. Furthermore, significant decrease in urea and increase in creatinine levels at the dose of 2000 mg/kg occurred in female rats. Mild histopathological changes were also observed in the liver of male rats and kidney of female rats treated with the extract, respectively at doses of 1000 and 2000 mg/kg, and 2000 mg/kg.CONCLUSION: Findings from the present study suggest that prolonged administration of extract of Moringa stenopetala at therapeutic doses is safe, but shows sign of mild toxicity as dose increases, with differential effect on male verses female rats.

Objective: Hematological disorders including anaemia are prevalent in alcoholics. The study was an attempt to find out the toxic effects of ethanol on blood values and semen parameters of albino rats. Material & Methods: 15 male albino rats with body weight (bwt) of 190 – 220 gm were used for the 2-phase study. 25% ethanol was administered via oral cannula to a group of 5 male rats each at daily dose of 0.6ml/200gm bwt respec-tively for 3 days during phase I. Phase II was a recovery study involving 5 male rats exposed to dose regimen as in phase I, and sacrificed after 3-day withdrawal of treatment. The control group of 5 male rats was given sterile water ad-libitum. Each animal was weighed before sacrifice to obtain difference in bwt relative to its basal value. Blood samples were collected by cardio-puncture from the rats for hematology and serum testosterone at the end of each phase. Semen parameters were determined and compared with controls. Results: Ethanol caused significant reduction (P< 0.05) in the hematological profile as well as in the serum testoster-one and semen parameters of the animals. Discontinuation of the drug use however showed gradual recovery of the depressed indices of the semen, serum testosterone and blood parameters. Conclusion: The ethanol could induce reversible changes in hematological profiles and semen parameters of rats, and by extension man. Hence, the study supported the use of alcohol with caution especially in infertile men and those prone to anemic tendencies. <p class="RSCB01ARTAbstract"><strong>Objective</strong>:<strong> </strong>The aim of this study was to characterise the resveratrol inclusion complex with β-cyclodextrin (RCD) and evaluate their toxicity in wistar rats.</p><p class="RSCB01ARTAbstract"><strong>Methods: </strong>The RCD were prepared in ultra-turrax. For characterization of the RCD were used: Fourier transform infra-red Spectroscopy, Nuclear Magnetic Resonance (NMR), Differential Scanning Calorimetry (DSC) and X-ray powder diffraction. The RCD and others 4 treatments were performed by the chronic oral administration in 35 rats during 60 ds. After the treatments they were euthanized and the serum blood were collected to analyzed some hemogram and biochemical parameters including aspartyl aminotransferase (AST); alanine aminotransferase (AST); phosphatase alkaline (ALP); total bilirubin (TB); direct bilirubin (DB); total protein (TP); total cholesterol (TC), triacylglycerol (TAG), very low-density lipoprotein (VLDL), high-density lipoprotein (HDL), calcium, iron and phosphate using fully automated biochemistry analyzer.</p><p class="RSCB01ARTAbstract"><strong>Results: </strong>The characterization results indicated a successful formation of the RCD. All hematological parameters analysed were within the normal values in all the groups. Furthermore, the hemogram and biochemical parameters were significantly (P&gt;0.05) similar to the control group.</p><p class="RSCB01ARTAbstract"><strong>Conclusion: </strong>The daily oral administration during 60 d of RCD are not harmful on blood parameters of Wistar rats. Thus, RCD can be used safely for treatment of some metabolic diseases.</p>

This study was carried out to investigate the effect of experimental infection by Escherichia coli on some blood parameters and histopathological changes in male rats to reach this objective twenty male rats (Rattus norvigicus) were divided into two groups, housed under same conditions of water, feed, temperature, light and humidity. The first one served as control group, while the second was experimentally infected with Ecoli by administration of 2ml / rat of (1×109) colony forming unit/ml of E-coli. Five days later blood were obtained from animals by cardiac puncture to measure the total and differential white blood cells count hemoglobin concentration packed cells volume red blood cells count, and thrombocytes count. One animal from each group was killed and histopathological examination was done for intestine kidney liver and spleen to detect the effect of infection on these organs. The results revealed a significant increase in total leukocytes count (P<005) in infected group while the differential count showed a significant increase in neutrophils and significant decrease in lymphocytes (P< 0.05) in same group. There were a significant decrease in hemoglobin concentration packed cell volume and non significant decrease in red blood cells count. While thrombocytes count showed a significant decrease in infected group compared with control group (P<0.05). The histopathological examination of infected male rats intestine was demonstrated mild diffuse inflammation which include increased numbers of neutrophile in tissue intraepithelial lymphocytes and goblet cells In kidney renal tubules showed extensive epithelial swelling with decreased lumen space and generalized necrotic changes with interstitial hemorrhage in renal cortex. In liver hepatocytes were swollen with decreased sinusoidal spaces and widely distributed necrotic foci were seen. There is extensive splenic necrosis with infiltration of leukocytes from white pulp to red pulp of spleen.

The article presents the results of studies of morphological and biochemical blood parameters of carp juveniles when adding the extract Quercus cortex (1, 2, 3 mg/kg of feed), probiotic preparation of Soya-bifidum (0.7 ml/kg of feed), antibiotics Ciprofloxacin hydrochloride in the composition of the drug Antibak 250 (100 mg/kg of feed). Analysis of hematological parameters of carp juveniles showed that adding Quercus cortex extract to the diet at a dosage of 2 mg/kg of feed contributed to increasing SGE by 66%, the average concentration of hemoglobin in the red blood cell by 20% compared to the control group. An increase in the level of leukocytes compared to the control was recorded only in the IV experimental group and made 13% (P≤0.05). An increase in glucose in the groups receiving Quercus cortex extract has been stated: in the I experimental group- by 4.2 times (P≤0.001), in II – by 3.4 (P≤0.001) and in III – by 3.3 (P≤0.001) compared to the control. Increasing glucose concentration indicates active metabolic processes and better stress resistance in the fish body. Iron content in carp blood was higher only in the first experimental group (by 3 times) compared to the control. Increasing activity of alanine aminotransferase was observed in all experimental groups, the highest rate of this enzyme being recorded in the IV experimental group (feed with probiotic additive Soya-bifidum) with a significant excess of control by 48%. Lactate dehydrogenase enzyme activity exceeded the control in all experimental groups, maximum increase (by 67%) was observed in the IV experimental group. Alkaline phosphatase was below the control in all experimental groups, minimum values being registered in the IV experimental group- by 37% less than the control. Indicators of p-amylase in the IV group exceeded the control by 2.6 times, in the V group – by 1.5 times. In the II group p-amylase was lower than the control by 44%, and in the III – by 1.9 times. According to the obtained hematological data, it has been established that including Quercus cortex extract into the diet of carp juveniles is promising due to the positive effect on the fish physiological status, immune status, metabolic processes, hematopoiesis and digestibility of feed.

In this paper present the results of research of influence of the preparation «Kvatronan-Se» and carboxylates of Se, Cu, Mn, Cr, Ge in different combinations on haematological parameters of blood of experimental animals. Study was carried out on the basis of PC Haleks-Agro. It is located in the village Gul'sk of Novograd Volynskiy district of Zhytomyr region. In the farm to bred of Simmental cows. For the experiment was formed four groups. We took into account age, live weight of 650–700 kg and the average milk yield 5000–6000 kg. Animals is located in identical conditions of feeding and maintenance. Research experiment was carried out on animals couples counterparts.  Preparations introduced animals three times at 10–12 days after insemination. The cows first experimental group were injected complex nanokarboksylativ Ge, Cu, Mn and Cr, second – drug Kvatronan–Se and animals third experimental group was administered a complex composed of Se, Cu, Mn, Cr. On 13 day after insemination in experimental animals were taken blood from tail vein and examined the contents of hematological parameters. Established that injection nanokarboksylativ enhance the fertility of cows in the first and third group, the figure rose to 20% in the second by 40% compared with the control. Research hematological parameters blood  animals shows that after entering content drugs of blood cells had minor changes but remained within the physiological norm. The results show that in  concentration monocyte the second group  was at 5.53% (P > 0.01) higher compared to the control group and 2.3% compared to the first and third. The content of lymphocytes in this group decreased by 26.3% (P > 0.05), 22% and 16% relative to the control, the first and third groups. Also, it should be noted that the hemoglobin in the experimental groups decreased in group I by 7.9% in the II – 3.9% and III by 7.3% compared with the control group. Also, these groups distinguished downward trend eosinophils 1.8%, 1.5%, 2.05%. Then, as the number of segment-nuclear neutrophils contrast, in the first group increased by 18.2% in the second and third at 16.9% comparatively control. The number of monocytes in the second group doslfidniy the suspicion was higher by 66.3% and 0.3% higher than the physiological norm – it may indicate a high immune response.

Abstract Objectives Transport of blood tubes is mainly by car or pneumatic transport. The transportation of blood tubes by drones is a novel approach for rapid transportation of blood tubes over long distances. However, limited data on the stability of biochemical, coagulation and hematological parameters is available after transport of blood tubes by drone. Methods To investigate the effect of drone transport on the stability of blood parameters, four test flights were performed. Blood was drawn from 20 healthy individuals and 39 of the most frequently measured blood parameters were compared between 4 groups; immediate measurement (control), late measurement, transport by car and transport by drone. Total Allowable Error (TAE) of the EFLM Biological Variation Database was used to determine the clinical relevance of significant differences. Results The majority of blood parameters were not affected by drone transport. Eight of the measured parameters showed significant differences between all the groups; glucose, phosphate, potassium, chloride, hemoglobin, platelet count, APTT and Lactate dehydrogenase (LD). A clinically relevant increase for LD after transport and a decrease for glucose values in time and after transport compared with the control group was shown. Conclusions Transportation of blood tubes from healthy individuals by drones has a limited clinically relevant effect. From the 39 investigated blood parameters only LD and glucose showed a clinically relevant effect.

Background: Tartrazine (Yellow 5 or E102) is a synthetic food dye able to modify perception and behavior, causingagitation, confusion, rhinitis and can produce hyperactivity syndrome in children when is combined with benzoates.Additionally, it can trigger oxidative stress which consequently generates metabolic disorders. Therefore, the studywas designed to evaluate the harmful effects of the food additive tartrazine and to observe beneficial properties ofblackthorn fruits (Prunus spinosa) on the blood and organs of albino Wistar rats.Materials and methods: This study was carried out on 20 mature Wistar rats, randomly divided into four groupsof five animals. Over the course of the experiment, the control group received only food and drinking water, group Ireceived 75 mg of tartrazine dissolved in (250 ml) water group II was given 75 mg of tartrazine and 200 mg of driedblackthorn fruit powder 200 mg dissolved simultaneously in (250 ml) of tartrazine-water mixture (aiming to reducethe tartrazine toxicity) and group III received a higher dose of tartrazine (100 mg) in (250 ml) of water.Results: At the end of the experiment, values regarding kidney and liver weight were significantly increased, whilethe weight of the spleen was slightly decreased compared with the weight of the control group. Biochemical andhematological assays, of the blood samples show that the addition of tartrazine in the diet of rats caused significantchanges in all biochemical and hematological parameters of the blood. In the group II, which received (P. spinosa)powder combined with tartrazine, the biochemical and hematological parameters had average values similar to thecontrol group.Conclusions: Histopathological assay showed that the application of tartrazine in the group I, II and III producedlesions of the kidneys, spleen and the liver for all rodents. Tartrazine was able to generate histopathological changes,which caused significantly tissue lesions of the liver and significant changes in blood parameters. Blackthorn powdershowed a promising protective role for the blood parameters but demonstrated no significant benefits for the organs.

Abstract Background: Several studies indicated that dietary organic selenium (Se) usually absorbed better than an inorganic source, with high retention and bioavailability. Dietary Se as an antioxidant element affects the immune system and hematological status in animals. Therefore, the aim of this study was to evaluate the effect of dietary supplementation of bacterial selenium as an organic source on hematology, immunity response, selenium retention, and gut morphology in broiler chickens. Results: The present results revealed that supplementation of inorganic Se was associated with the lowest level of RBC, HB, and PCV with significant difference than ADS18-Se. In the starter stage, both T2 and T5 were associated with the significantly highest IgG level compared to the basal diet, while all supplemented groups showed higher IgM levels compared to the control group. In the finisher phase, all Se supplemented groups showed significant (P ˂ 0.05) increases in IgG, IgA, and IgM levels compared to T1. Birds fed bacterial-Se showed high intestinal villus height and better Se retention more than sodium selenite. The organic selenium of ADS18 had a superior action in improving Se retention compared to ADS1 and ADS2 bacterial Se. Conclusion: Bacterial organic Se had a beneficial effect on the villus height of small intestine led to high Se absorption and retention. Thus, it caused a better effect of Se on hematological parameters and immunity response.

Abstract Objectives Aflatoxins are highly toxic, carcinogenic, mutagenic and teratogenic metabolites produced by some Aspergillus species that contaminate a wide range of nourishment cereal and pose serious health problems for human and livestock with nanograms limits of acceptability in foods. This study aimed to extract, identify and assess essential oils extracted from ginger rhizomes (Zingiber officinale Rose) (EOG) and flower head of Chamomile (Chamomilla recutita L.) (EOC) against aflatoxin producing fungi. Toxicity evaluation was conducted to ensure their safety. Methods Essential oils were extracted by hydrodistillation using Clevenger apparatus and identified by Gas Chromatography-Mass Spectrometry (GCMS). The fungicidal effect was assessed against penicillium chrysogenum, Aspergillus niger and Aspergillus flavus isolated from bean, corn and rice using agar disc diffusion method to determine maximum inhibition zone (MIZ) and minimum inhibitory concentration (MIC) for each extract. Toxicity evaluation was conducted on two groups of 10 male rats treated with EOG or EOC at a dose of 0.1 g/kg (1/50 LD) for 28 consecutive days against control group. Serum biochemical analysis, hematological parameters and liver histological investigations were considered. Results The phytochemical analysis identified 20 compounds in EOG and 26 compounds in EOC. The most common compounds were monoterpenes and sesquiterpenes in EOG and EOC respectively. The highest MIZ showed by EOC (5.15 ± 0.07 cm) and EOG (4.15 ± 0.07 cm) against P. chrysogenum. The MIC showed that EOG has greater inhibitory effect against P. chrysogenum (2.5 μg/ml) whereas EOC exhibit strong inhibitory effect against all tested fungi with lower MIC values ranged from 1.25 to 2.5 μg/ml. Serum biochemical analysis and hematological parameters indicated minor changes in EOC and EOC group while liver histological features showed marked improvement in hepatic tissues of EOC and EOG groups compared with control. Conclusions Promising fungicidal effects of both EOC and EOG against aflatoxin producing fungi have been demonstrated, however further evaluation is needed concerning its safety.
