Application of Lemongrass Essential Oil as a Natural Preservative Agent for Pineapple Juice

Pineapple juice is frequently consumed for its extensive health benefits, but is prone to microbiological degradation. Pasteurization – as a current preservation technique – have been degrading the nutritional qualities of fruit juices, and the use of synthetic preservatives have been perceived negatively by consumers. In this study, lemongrass essential oil was applied for the preservation of pineapple juice. Agar Well Diffusion Method was used for antimicrobial activity screening of lemongrass essential oil against E. coli, S. cerevisiae, and A. niger, while Broth Macrodilution Method was used in determining Minimum Inhibitory Concentration (MIC) of lemongrass essential oil, which was evaluated to be 0.1%. Furthermore, the addition of 0.1% lemongrass essential oil into pineapple juice had a negative impact regarding the sensorial acceptability, but was able to significantly suppress the microbial count for bacteria, yeast, and mould during 5 days of storage time in chilling temperature (4oC). Vitamin C content degradation and pH changes during storage were not significantly affected by the addition oflemongrass essential oil. Furthermore, the addition of 0.1% lemongrass essential oil was observed to be not synergistic with the enzyme activity of pineapple juice, and has negatively impacted the enzyme stability of the pineapple juice.

Abstract: Endodontics is the a type of treatment that aims to keep the teeth in function. Disinfection of the root canal is very important in endodontic treatment. Root canal disinfection can be done by giving the material a root canal medication. One of the bacteria that causes the failure of root canal treatment is the bacterium Enterococcus faecalis. The lemongrass essential oil contains geranial, neral, and mirsen which have antimicrobial activity against Gram-positive and Gram-negative. This study aimed to determine whether the lemongrass essential oil was effective against bacteria Enterococcus faecalis. This was an experimental study using post test only control group design with agar plate diffusion method. Samples consisted of a group of lemongrass essential oil with several concentrations: 25%, 50%, 75%, 100%, each consisted of 4 samples. The 16 control groups consisted of positive and negative groups. Diameter of inhibition was determined by the ability to inhibit Enterococcus faecalis cultured on MHA agar. The results showed that the average inhibitory diameter of 25% lemongrass essential oil was 2.60 mm; of 50% was 4.73 mm; of 75% was 4.50 mm; and of 100% was 5.34 mm. Conclusion: Lemongrass essential oil showed an antibacterial effect to inhibit the growth of bacteria Enterococcus faecalis.Keywords: enterococcus faecalis, lemongrass oil, antibacerialAbstrak: Endodontik merupakan salah satu jenis perawatan yang bertujuan mempertahankan gigi agar tetap dapat berfungsi. Minyak atsiri sereh dapur mengandung geranial, neral dan mirsen yang memiliki aktifitas antimikrobapada gram positif dan gram negatif. Penelitian ini bertujuan untuk mengetahui apakah minyak atsiri sereh dapur memiliki efek terhadap bakteri Enterococcus faecalis. Penelitian ini menggunakan metode eksperimental dengan desain post test only control group design dan metode difusi lempeng agar. Sampel penelitian terdiri dari kelompok minyak atsiri sereh dapur dengan konsentrasi 25%, 50%, 75%, dan 100% yang masing-masing terdiri dari 4 sampel; 16 kelompok kontrol terdiri atas kelompok positif dan negatif. Diameter hambat ditentukan berdasarkan kemampuan menghambat Enterococcus faecalis yang dibiakkan pada media agar MHA. Hasil penelitian memperlihatkan rata-rata diameter hambat minyak atsiri sereh dapur dengan konsentrasi 25% sebesar 2,60 mm, 50% sebesar 4,73 mm, 75% sebesar 4,50 mm, dan 100% sebesar 5,34 mm. Simpulan: Minyak atsiri sereh dapur memiliki efek antibakteri untuk menghambat pertumbuhan bakteri Enterococcus faecalis.Kata kunci: enterococcus faecalis, sereh dapur, antibakteri

Background: The essential oil of methyl eugenol rich Cymbopogon khasianus Hack. was evaluated and its bioactivities were compared with pure methyl eugenol. So far, methyl eugenol rich essential oil of lemongrass was not studied for any biological activities; hence, the present study was conducted. Objective: This study examined the chemical composition of essential oil of methyl eugenol rich Cymbopogon khasianus Hack., and evaluated its antioxidant, anti-inflammatory, antimicrobial, and herbicidal properties and genotoxicity, which were compared with pure compound, methyl eugenol. Material and Methods: Methyl eugenol rich variety of Cymbopogon khasianus Hack., with registration no. INGR18037 (c.v. Jor Lab L-9) was collected from experimental farm CSIR-NEIST, Jorhat, Assam (26.7378°N, 94.1570°E). The essential oil wasobtained by hydro-distillation using a Clevenger apparatus. The chemical composition of the essential oil was evaluated using GC/MS analysis and its antioxidant (DPPH assay, reducing power assay), anti-inflammatory (Egg albumin denaturation assay), and antimicrobial (Disc diffusion assay, MIC) properties, seed germination effect and genotoxicity (Allium cepa assay) were studied and compared with pure Methyl Eugenol compound (ME). Results: Major components detected in the Essential Oil (EO) through Gas chromatography/mass spectroscopy analysis were methyl eugenol (73.17%) and β-myrcene (8.58%). A total of 35components were detected with a total identified area percentage of 98.34%. DPPH assay revealed considerable antioxidant activity of methyl eugenol rich lemongrass essential oil (IC50= 2.263 μg/mL), which is lower than standard ascorbic acid (IC50 2.58 μg/mL), and higher than standard Methyl Eugenol (ME) (IC50 2.253 μg/mL). Methyl eugenol rich lemongrass EO showed IC50 38.00 μg/mL, ME 36.44 μg/mL, and sodium diclofenac 22.76 μg/mL, in in-vitro anti-inflammatory test. Moderate antimicrobial activity towards the 8 tested microbes was shown by methyl eugenol rich lemongrass essential oil whose effectiveness against the microbes was less as compared to pure ME standard. Seed germination assay further revealed the herbicidal properties of methyl eugenol rich essential oil. Moreover, Allium cepa assay revealed moderate genotoxicity of the essential oil. Conclusion: This paper compared the antioxidant, anti-inflammatory, antimicrobial, genotoxicity and herbicidal activities of methyl eugenol rich lemongrass with pure methyl eugenol. This methyl eugenol rich lemongrass variety can be used as an alternative of methyl eugenol pure compound. Hence, the essential oil of this variety has the potential of developing cost-effective, easily available antioxidative/ antimicrobial drugs but its use should be under the safety range of methyl eugenol and needs further clinical trials.

Fungal food spoilage plays a key role in the deterioration of food products, and finding a suitable natural preservative can solve this problem. Therefore, antifungal activity of green mandarin (Citrus reticulata) essential oil (GMEO) in the vapor phase against the growth of Penicillium (P.) expansum and P. chrysogenum inoculated on wheat bread (in situ experiment) was investigated in the current research. The volatile compounds of the GMEO were analyzed by a gas chromatograph coupled to a mass spectrometer (GC–MS), and its antioxidant activity was determined by testing free radical-scavenging capacity (DPPH assay). Moreover, the disc diffusion method was used to analyze the antifungal activity of GMEO in in vitro conditions. The results demonstrate that the Citrus reticulata EO consisted of α-limonene as the most abundant component (71.5%), followed by γ-terpinene (13.9%), and β-pinene (3.5%), and it displayed the weak antioxidant activity with the value of inhibition 5.6 ±0.7%, which corresponds to 103.0 ±6.4 µg TEAC.mL-1. The findings from the GMEO antifungal activity determination revealed that values for the inhibition zone with disc diffusion method ranged from 0.00 ±0.00 (no antifungal effectiveness) to 5.67 ±0.58 mm (moderate antifungal activity). Finally, exposure of Penicillium strains growing on bread to GMEO in vapor phase led to the finding that 250 μL.L-1 of GMEO exhibited the lowest value for mycelial growth inhibition (MGI) of P. expansum (-51.37 ±3.01%) whose negative value reflects even supportive effect of the EO on the microscopic fungus growth. On the other hand, GMEO at this concentration (250 μL.L-1) resulted in the strongest inhibitory action (MGI: 54.15 ±1.15%) against growth of P. chrysogenum. Based on the findings it can be concluded that GMEO in the vapor phase is not an effective antifungal agent against the growth of P. expansum inoculated on bread; however, its antifungal potential manifested against P. chrysogenum suggests GMEO to be an appropriate alternative to the use of chemical inhibitors for bread preservation.

<p><strong>Objective: </strong>To evaluate the chemical composition, antibacterial and antioxidant properties of stem bark essential oil of <em>Litsea glutinosa </em>C. B. Rob.</p><p><strong>Methods: </strong>The essential oil isolated from stem bark of <em>L. glutinosa </em>and their chemical composition was analyzed by gas chromatography coupled with mass spectrometry detector. The <em>in vitro </em>antibacterial activity of the stem bark essential oil was investigated against eight human pathogenic bacterial clinical isolates using agar disc diffusion method and MIC value was determined by modified resazurin microtitre-plate assay. The antioxidant activity of essential oil was measured by 2, 2-diphenyl-1-picrylhydrazyl radical (DPPH), 2, 2-azinobis-3-ethylbenzothiazoline-6-sulphonate radical cation (ABTS) and β-carotene bleaching assay.</p><p><strong>Results: </strong>GC-MS analysis of stem bark essential oil resulted in the identification of 37 compounds, off which 9,12-octadecadienoic acid (62.57%), hexadecanoic acid (12.68%), stigmast-5-en-3-ol (6.87%) and vitamin E (2.51%) were the main constituents representing 84.63% of the oil. The determination of <em>in vitro</em> antibacterial activity of stem bark essential oil resulted in significant inhibition zone (15.00±0.57 mm) and MIC value (0.15±0.15×10<sup>-2</sup> mg/ml) against the pathogenic bacteria <em>Vibrio cholera</em> followed by <em>Pseudomonas aeruginosa</em> and <em>Salmonella typhi. </em>The results of DPPH radical scavenging (IC<sub>50</sub>:4.540±0.06 µg/ml), ABTS (IC<sub>50</sub>:256.02±0.06 µg/ml) and β-carotene bleaching assay (%I: 78.51±0.42 <strong>%</strong>) showed significant <em>in vitro</em> antioxidant property.</p><p><strong>Conclusion: </strong><em>L. glutinosa</em> stem bark essential oil showed potential antibacterial activity against the <em>Vibrio cholera</em>. The results of this investigation supported the ethnomedical claim of essential oil as a demulcent, antidiarrheal and antioxidant drug.</p>

Abstract The objective of this work was to evaluate the partial replacement of gum arabic by modified starches on the spray-drying microencapsulation of lemongrass (Cymbopogon flexuosus) essential oil. The ultrasound-assisted emulsions were prepared with 30% (w/w) of wall material, 7.5% (w/w) of oil load, and 1:1 (w/w) replacement ratio for all treatments. After 16 hours, the incompatibility observed between gum arabic and octenyl succinic anhydride (OSA) starch did not affect the obtained microparticles, since the treatment with OSA starch, partially replacing gum arabic, showed the best results for the process yield and for the oil charge retention after spray-drying process, and the treatment showed Newtonian viscosity close to that of the treatment prepared with gum arabic. Maltodextrin dextrose equivalent 10 (10DE) shows an oil load similar to that of the treatment with gum arabic, while the presence of maize maltodextrin DE20 reduces the content of encapsulated oil and the efficiency of the drying process due to the adherence of particles to the chamber. Therefore, the partial substitution of gum arabic is an alternative for the formation of emulsions, for the spray-drying microencapsulation of lemongrass essential oil.

Citrus species of plants are among the most commercially cultivated crops, mainly for their fruit. Besides, the generally consumed flesh inside the fruit, the peel is quite important too. Essential oils extracted from the peel have a history of being used by humankind for centuries. These essential oils are rich in antioxidants and antimicrobial agents. Comparative investigation of volatile constituents, and antioxidant and antimicrobial activities were undertaken. The essential oils were evaluated through gas chromatography-mass spectrometry (GC–MS), and enantiomeric composition by chiral GC–MS. Similarly, the antioxidant properties were evaluated by 2,2-diphenyl-1-picrylhydrazyl scavenging assay, and antimicrobial activities were assayed using the disk diffusion method. The highest extraction yield of 1.83% was observed in Citrus sinensis Osbeck. GC–MS analysis showed limonene (63.76–89.15%), γ-terpinene (0.24–6.43%), β-pinene (0.15–6.09%), linalool (0.35–3.5%), sabinene (0.77–2.17%), myrcene (0.74–1.75%), α-terpineol (0.28–1.15%), and α-pinene (0.2–0.58%) as the major constituents of the essential oil of the Citrus species studied. For the first time, through our study, chiral terpenoids have been observed from Citrus grandis Osbeck essential oil. The order of antioxidant activity is as follows: Citrus grandis Osbeck red flesh > Citrus reticulata Blanco > Citrus sinensis Osbeck > Citrus grandis Osbeck white flesh. Except for Citrus grandis Osbeck white flesh (52.34 µL/mL), all samples demonstrated stronger antioxidant activities than those of the positive control, quercetin (5.60 µL/mL). Therefore, these essential oils can be used as a safe natural antioxidant to prevent product oxidation. Likewise, citrus peel essential oil showed antimicrobial activity against tested bacterial strains, albeit marginal.

In the present study, chemical composition and antibacterial properties of essential oil obtained from the aerial parts of the Melicope pteleifolia (Champ. ex Benth.) T.G Hartley in Dalat were evaluated. Essential oil was isolated through hydro-distillation. Twenty-nine constituents comprising 100% of the essential oil were characterized by gas chromatography/mass spectrometry (GC-MS) techniques. The major compounds in the essential oil were (+)-Sabinene (34.73%), Cis-α-bergamotene (13.15%), Z-α-trans-bergamotol (5.28%), β-mycrene (4.98%), and 1,3,6-octatriene, 3,7-dimethyl-(4.71%). Antibacterial activities of Melicope pteleifolia essential oil were investigated against Gram-positive and-negative bacteria. Results showed significant activities against Streptococcus pyogenes and Escherichia coli using an agar well diffusion method. The application of this essential oil in preventing and eliminating bacteria could be useful in fields as medicine and cosmetics.

ABSTRAK                                                                        Imbuhan pakan unggas dapat berasal dari bahan herbal yang mengandung berbagai komponen aktif yang bermanfaat bagi pertumbuhan ternak.Temu putih dan jahe gajah dapat dimanfaatkan sebagai imbuhan pakan karena mengandung minyak atsiri yang dapat berperan sebagai agen antibakteri. Penelitian ini bertujuan untuk mengetahui komponen penyusun minyak atsiri dan aktivitas antimikroba pada rimpang temu putih dan jahe gajah. Penelitian dilakukan dengan percobaan in vitro menggunakan temu putih dan jahe gajah yang diolah menjadi bentuk ekstrak minyak atsiri temu putih dan jahe gajah sebagai materi uji komposisi penyusun minyak atsiri serta bentuktepung dan enkapsulasi sebagai materi uji aktivitas antimikroba. Komposisi minyak atsiri temu putih terdiri dari lima komponen penyusun dengan cis-1,7-octadien-3-yl acetat sebagai komponen utama. Komposisi minyak atsiri jahe gajah terdiri dari tujuh komponen dan benzene,1-(1,5-dimethyl-4-hexenyl)-4-methyl-(CAS) ar-curcumene sebagai komponen utama. Minyak atsiri yang terkandung pada temu putih dan jahe gajah mempunyai peran dalam menghambat mikroba. Uji komposisi penyusun minyak atsiri menggunakan alat GC-MS dan uji aktivitas antimikroba menggunakan metode disc diffusion dan. Hasil dari uji aktivitas antimikroba menunjukkan bahwa temu putih dan jahe gajah dalam bentuk tepung dan enkapsulasi memiliki perbedaan yang sangat nyata (P<0,01) terhadap aktivitas antimikroba pada bakteri asam laktat, Escherichia coli dan Salmonella sp. Campuran temu putih dan jahe gajah (1:1) menunjukkan kemampuan terbaik dalam menghambat pertumbuhan bakteri patogen dengan diameter zona hambat 5,70±0,14 mm  (Escherichia coli) dan 6,88±0,45 mm (Salmonella sp.).Kata Kunci : antimikroba, fitobiotik, jahe gajah, minyak atsiri, temu putihABSTRACTThe poultry feed additives can contain herbal ingredients that contain various beneficial components for livestock growth. White turmeric and giant ginger can be used as feed additives because they contain essential oils that can be used as antibacterial agents. This study aims to determine the constituent components of essential oils and antimicrobial activity in white turmeric and giant ginger rhizomes. The study was carried out by in vitro experiments using white turmeric and giant ginger which were processed into the form of essential oil extract as material for the composition of essential oils test, and powder and encapsulation form as antimicrobial activity test material. The composition of essential oils of white turmeric consists of five constituent components with cis-1,7-octadien-3-yl acetate as the main component. The composition of giant ginger essential oil consists of seven components with benzene, 1- (1,5-dimethyl-4-hexenyl) -4-methyl- (CAS) ar-curcumene as the main component. Essential oils contained in the white turmeric and giant ginger have a role in inhibiting microbes. The composition of the essential oil tested using GC-MS and the antimicrobial activity test used the disc diffusion method. The results of the antimicrobial activity test showed that white turmeric and giant ginger in powder and encapsulation form had significant differences (P <0.01) on antimicrobial activity in lactic acid bacteria, Escherichia coli and Salmonella sp. The mixture of white turmeric and giant ginger (1: 1) showed the best ability to inhibit the growth of pathogenic bacteria with inhibitory zone diameters of 5.70 ± 0.14 mm (Escherichia coli) and 6.88 ± 0.45 mm (Salmonella sp.).Keywords: antimicrobial, essential oil, giant ginger, phytobiotic, white turmeric
