The quality of methods and products are usually influenced by several input factors. Research has recently focused on understanding the effects of multidimensional and interconnected input factors on the results of pharmaceutical products and analytical methods using Design of Experiment (DoE). Furthermore, it examines how DOE may be implemented, both for students and teachers, as well as highlighting historical perspectives on DOE. A good experimental design can help you make the most use of the available resources and make the analysis of the results easier. Collaborations between researchers and practitioners that are pushing the boundaries of experimental design are examined. It provides an overview of the principles and applications of the most common screening and response surface design, as well as creating mixtures designs.

The design process of journal bearings of turbomachines is complex and time-consuming due to the many geometric and physical variables involved. This paper reports on the design of experiments (DOE) and the response surface design of experiments (RSDOE) methods employed on the design of the drive-end and free-end three-lobe journal bearings supporting a centrifugal compressor rotor. The suitability of each technique is discussed. The bearing design variables employed are bearing slenderness ratio, radial clearance, preload, and lubricant inlet temperature. The rotordynamic response variables selected were the critical speed location, the vibrations at critical speed and operating speed for both bearings, and the threshold speed of instability. The use of a nonlinear (quadratic) RSDOE model is justified. An optimization approach combining an SRDOE and rotordynamic finite element modeling is presented. This method leads to arrive to a multivariate model for multi-objective optimization with very few computations. Identification of the dominant design variables and their effects on several response variables allows establishing engineering feasible solutions with focus on manufacturing versus operating conditions tradeoff.

Concrete is a composite material that has complex mechanical properties. The mechanical properties of each of its components are different at the mesoscopic scale. Studying the relationship between the macroscopic and mesoscopic parameters of concrete can help better understand its mechanical properties at these levels. When using the discrete element method to model the macro-mesoscopic parameters of concrete, their calibration is the first challenge. This paper proposes a numerical model of concrete using the particle discrete element software particle flow code (PFC). The mesoscopic parameters required by the model need to be set within a certain range for an orthogonal experimental design. We used the proposed model to perform numerical simulations as well as response surface design and analysis. This involved fitting a set of mapping relationships between the macro–micro parameters of concrete. An optimization model was established in the MATLAB environment. The program used to calibrate the mesoscopic parameters of concrete was written using the genetic algorithm, and its macro-micro parameters were inverted. The following three conclusions can be drawn from the orthogonal test: First, the tensile strength and shear strength of the parallel bond between the particles of mortar had a significant influence on the peak compressive strength of concrete, whereas the influence of the other parameters was not significant. Second, the elastic modulus of the parallel bonding between particles of mortar, their stiffness ratio and friction coefficient, and the elastic modulus and stiffness ratio of contact bonding in the interfacial transition zone had a significant influence on the elastic modulus, whereas the influence of the other parameters was not significant. Third, the elastic modulus, stiffness ratio, and friction coefficient of the particles of mortar as well as the ratio of the contact adhesive stiffness in their interfacial transition zone had a significant influence on Poisson’s ratio, whereas the influence of the other parameters was not significant. The fitting effect of the response surface design was good.

Abstract Enterotoxigenic E. coli (ETEC) is a leading cause of moderate-to-severe diarrhoea. ETEC colonizes the intestine through fimbrial tip adhesin colonization factors and produces heat-stable and/or heat-labile (LT) toxins, stimulating fluid and electrolyte release leading to watery diarrhoea. We reported that a vaccine containing recombinant colonization factor antigen (CfaEB) targeting fimbrial tip adhesin of the colonization factor antigen I (CFA/I) and an attenuated LT toxoid (dmLT) elicited mucosal and systemic immune responses against both targets. Additionally, the toll-like receptor 4 ligand second-generation lipid adjuvant (TLR4-SLA) induced a potent mucosal response, dependent on adjuvant formulation. However, a combination of vaccine components at their respective individual optimal doses may not achieve the optimal immune profile. We studied a subunit ETEC vaccine prototype in mice using a response surface design of experiments (DoE), consisting of 64 vaccine dose-combinations of CfaEB, dmLT and SLA in four formulations (aqueous, aluminium oxyhydroxide, squalene-in-water stable nanoemulsion [SE] or liposomes containing the saponin Quillaja saponaria-21 [LSQ]). Nine readouts focusing on antibody functionality and plasma cell response were selected to profile the immune response of parenterally administered ETEC vaccine prototype. The data were integrated in a model to identify the optimal dosage of each vaccine component and best formulation. Compared to maximal doses used in mouse models (10 µg CfaEB, 1 µg dmLT and 5 µg SLA), a reduction in the vaccine components up to 37%, 60% and 88% for CfaEB, dmLT and SLA, respectively, maintained or even maximized immune responses, with SE and LSQ the best formulations. The DoE approach can help determine the best vaccine composition with a limited number of experiments and may accelerate development of multi-antigen/component ETEC vaccines.

An experimental design is one of the tools which are used in statistical quality control. The proper implementation of experimental design results in the improvement of technological outcomes of a manufacturing process, which in turn leads to the enhancement of economic results. Permutation tests, among other things, form a group of resampling methods which are used to verify statistical hypotheses. These tests, unlike parametric ones, do not entail the fulfilment of strict criteria and may be used for a small number of observations. The presented article deals with the use of permutation tests in the design of experiments. The proposed method will be presented with reference to selected empirical data.

In this review, quality by design (QbD) initiatives are described as applications for many types of processes, such as in the pharmaceutical industry, biotechnology field, and for analytical methods development. Design space (DS) and design of experiments (DoE) provide useful results for analytical methods development and simulation of advanced processes in traditional manufacturing relationships. The three topics, QbD, DS and DoE are the best way to achieve strong and efficient industrial production.

This objectives of this research were both to study and determine the best level of concentration of yeast Saccharomyces cereviseae and period of fermentation on the quality of tomi-tomi vinegar (Flacourtia inermis). A completely randomized experimental design with two factors of treatment was applied in this research. The first factor was concentration of yeast S. cereviseae having four levels of tretament, i.e.: without the addition of yeast 0.5, 1 and 1.5 g yeast. The second factor was period fermentation with 1, 2, 3, 4, and 5 weeks. The result indicated that the concentration of yeast S. cereviseae 1.5 g and period fermentation 5 week produced a good tomi-tomi vinegar with total acids 51.22%, total dissolved solids 8.35, total sugar 8.07% and pH 5.40.

ABSTRAK Tidur merupakan kebutuhan yang harus terpenuhi terutama pada fase perkembangan karena selama tidur akan terjadi perkembangan otak maupun tubuh, sehingga gangguan tidur merupakan masalah yang akan menimbulkan dampak buruk terhadap pertumbuhan dan perkembangan bayi. Kualitas tidur bayi yang baik dapat diciptakan dengan memberikan pemijatan bayi secara rutin. Penelitian ini bertujuan untuk membuktikan bahwa pemijatan dapat mempengaruhi kualitas tidur bayi umur 0-3 bulan. Penelitian ini menggunakan desain penelitian Quasy Eksperimental dengan metode One Group Pretest-Postest. Sampel 22 bayi yang dipilih dengan tehnik Total Sampling yang di observasi sebelum dan sesudah diberikan pemijatan. Variabel yang diukur dalam penelitian ini adalah kualitas tidur bayi 0-3 bulan. Hasil penelitian menunjukkan bahwa ada pengaruh pijat bayi terhadap kualitas tidur bayi umur 0-3 bulan (p value  0,008 < α = 0,05).Berdasarkan hasil penelitian ini disarankan agar keluarga dan masyarakat memberikan pemijatan secara rutin dan mandiri untuk meningkatkan kebutuhan tidur bayi yang berkualitas.   ABSTRACT Sleep is a human necessity that must be met, especially in the development phase because during sleep will occur the brain and body developments, so that sleep disturbance is a problem that would cause adverse effects on infants’ growth and development. The good quality of sleep can be created by providing the infants massage routinely. This study aimed to prove that the massage could affect the quality of sleep on the 0-3 months old baby. This study used Quasy-experimental design with One Group Pretest-Posttest. The sample 22 infants selected by total sampling technique observed on before and after the massage. The variables measured in this study are the quality of sleep. The results of study indicate that there is an effect of infant massage to the sleep quality on 0-3 months old babies (p value 0,008 < α = 0,05).Based on the results of this study it recommended for the families and communities to provide infant massage regularly and independently to increase the quality of sleep on the baby.

This study aimed to determine the effect of ovaprim hormone, aromatase inhibitor and pituitary on the quality of the catfish eggs (Clarias gariepinus). Experimental Design used was Completely Randomized Design (CRD) with four treatments, each with three replications. Treatment A: ovaprim; treatment B: Aromatase inhibitors, treatment C: hypophysis and treatment D: Control. The results showed that the difference in treatment gave highly significant effect on fertilization and hatching eggs but no significant effect on the survival rate of larvae. Aromatase inhibitor hormone was the best because it provided highly significant effect on fertilization (92.66%), hatchability of eggs (95%), and surviva rate (81.33%) of fish larvae.   Keywords : Clarias gariepinus.
