Abstract Background This study aimed to evaluate the effect of nanoemulsion containing peppermint and rosemary essential oils in rats with osteoarthritis (OA). Methods In this experimental study, we prepared a nanoemulsion containing peppermint and rosemary essential oils by spontaneous emulsification and evaluated the nanoemulsion’s dermal irritation and toxicity. Investigating the analgesic effect of the nanoemulsion, we randomly assigned 36 male rats to 6 groups: Control (saline injection into the knee), osteoarthritis (intra-articular injection of 2 mg monosodium iodoacetate), and four groups of OA treated with nanoemulsion gel, nanoemulsion solution, rosemary and peppermint essential oil gel, or diclofenac sodium. Treatments were administered topically at a dose of 1 ml daily. Using behavioral tests, we assessed pain on days 1, 4, 7, and 14 after injection. Finally, we did the histopathological and biochemical evaluation of rats’ knee joints. Results There were no irritation signs on the animals’ skin after receiving the nanoemulsion and no changes in the hematological and biochemical parameters of rats’ blood compared to the control group. Receiving nanoemulsion decreased the mechanical (P < 0.001) and thermal allodynia (P < 0.05), thermal hyperalgesia (P < 0.05), and ambulatory-evoked pain in comparison with the OA group. Also, the nanoemulsion receiving rats showed an increase in SOD and GPx activity and a decrease in MDA level. Histopathology of synovial tissues confirmed the results of behavioral and biochemical tests. Conclusion The nanoemulsion containing essential oils of peppermint and rosemary reduces osteoarthritis pain via increasing antioxidant capacity and improving the histopathological features of the rats’ knee joint.

The purpose of this work was to study the influence of “Bendamin” on the morphological and biochemical indices of rat blood in experimental modeling of heart failure. The investigation were performed on white sexually mature young  male-rats of Wistar line  weighing 180–200 g, kept at the standard ration of the institute vivarium of the State Research Institute of Veterinary Preparations and Fodder Additives. To create a model of doxorubicin-induced cardiomyopathy, 24 male rats were selected. Animals were divided into 3 groups of 6 animals in each: control group – intact animals; experimental group E1, in which animals were modeled doxorubicin-induced cardiomyopathy by intraperitoneal injected of doxorubicin at a dose of 2.5 mg/kg 3 times a week for two weeks; experimental group E2, in which animals, after injection of doxorubicin, the preparation “Bendamin” was injected in the dose 20 mg/kg intragastrically. For experimental modeling of heart a decrease in the number of red blood cells was found in the blood of rats of the first experimental group by 34.7 % and hemoglobin level – by 24.2 %, with a simultaneous increase in the average hemoglobin content in one erythrocyte by 16.2 %. Doxorubicin-induced cardiomyopathy in rats of the first experimental group was accompanied by a decrease in leukocyte count by 22.2 %, an increase in uric acid level by almost in 2 times, creatinine by 11.4 %, cholesterol by 61.1 %. In the research of the protein synthesizing function of the liver of rats in experimental modeling of heart failure, a decrease in total protein by 9.6 % was found. These changes were accompanied by a slight decrease in albumin levels and a slight increase in globulin levels. By intoxication with doxorubicin, rats of the first experimental group showed a violation of the functional state of the liver, indicating an increase in the activity of aminotransferases in their blood, namely: AlAT by 34.2 %, AsAT – by 21.7 %, accordingly. Positive action of cardio preparation “Bendamin” on rats organism under conditions of creation of a model of doxorubicin-induced cardiomyopathy was installed, manifested by the normalization of hematological and biochemical parameters, functional status and protein synthesis of the liver.

Abstract Objectives Aflatoxins are highly toxic, carcinogenic, mutagenic and teratogenic metabolites produced by some Aspergillus species that contaminate a wide range of nourishment cereal and pose serious health problems for human and livestock with nanograms limits of acceptability in foods. This study aimed to extract, identify and assess essential oils extracted from ginger rhizomes (Zingiber officinale Rose) (EOG) and flower head of Chamomile (Chamomilla recutita L.) (EOC) against aflatoxin producing fungi. Toxicity evaluation was conducted to ensure their safety. Methods Essential oils were extracted by hydrodistillation using Clevenger apparatus and identified by Gas Chromatography-Mass Spectrometry (GCMS). The fungicidal effect was assessed against penicillium chrysogenum, Aspergillus niger and Aspergillus flavus isolated from bean, corn and rice using agar disc diffusion method to determine maximum inhibition zone (MIZ) and minimum inhibitory concentration (MIC) for each extract. Toxicity evaluation was conducted on two groups of 10 male rats treated with EOG or EOC at a dose of 0.1 g/kg (1/50 LD) for 28 consecutive days against control group. Serum biochemical analysis, hematological parameters and liver histological investigations were considered. Results The phytochemical analysis identified 20 compounds in EOG and 26 compounds in EOC. The most common compounds were monoterpenes and sesquiterpenes in EOG and EOC respectively. The highest MIZ showed by EOC (5.15 ± 0.07 cm) and EOG (4.15 ± 0.07 cm) against P. chrysogenum. The MIC showed that EOG has greater inhibitory effect against P. chrysogenum (2.5 μg/ml) whereas EOC exhibit strong inhibitory effect against all tested fungi with lower MIC values ranged from 1.25 to 2.5 μg/ml. Serum biochemical analysis and hematological parameters indicated minor changes in EOC and EOC group while liver histological features showed marked improvement in hepatic tissues of EOC and EOG groups compared with control. Conclusions Promising fungicidal effects of both EOC and EOG against aflatoxin producing fungi have been demonstrated, however further evaluation is needed concerning its safety. Objectives: To investigate the tissue-protective effects of three gel formulations (chitosan, chitosan with taurine or chitosan with dexpanthenol) as active substances against an acute overdose of diclofenac sodium. Methods: White outbred conventional male rats were allocated to five experimental groups: the first is an intact group that did not receive any drug, the second group is a control group that received 50mg/kg of diclofenac sodium once orally, the third, fourth and fifth groups are an experimental group that received our studied drugs at a dose of 0.16ml/100mg b.w. once orally 1 hr. before diclofenac sodium, the third group received chitosan-based gel 1%, the fourth group received chitosan-based gel 1% with 4% taurine and the fifth group chitosan-based gel 1% with 0.43% dexpanthenol. Blood samples were taken for biochemical, hematological and blood coagulation system tests on day 7th after administration of diclofenac sodium. Results: An acute overdose of diclofenac sodium caused marked extensive tissue necrosis in the liver, bleeding in the gastrointestinal tract and inflammatory process, these marks were evidenced by different changes in the test of the blood samples. Significantly 73.6% of the blood indicators were improved by the administration of chitosan-based gel 1% with 0.43% dexpanthenol, while 57.8% were improved by chitosan-based gel 1% with 4% taurine and 68.4% by chitosan-based gel 1%. Conclusion: Chitosan-based gel 1% with dexpanthenol 0.43% can help in mitigating hepatic injury, gastrointestinal bleeding, and systemic and local intestinal inflammation caused by an acute overdose of diclofenac sodium.

ABSTRACTObjective: The objective of this study is to explore the effects of the 70% ethanolic extract of pearl grass on the immune system of the osteoarthritismodel rat, characterized by the number of leukocytes and lymphocytes, and the histology of the joint. Osteoarthritis is a degenerative diseasecharacterized by chronic inflammation in the joints. Based on the daily practice of herbal medicine in some community in Indonesia, pearl grassusually used for anti-inflammation but not a lot of data to support it.Methods: We used 36 male rats Sprague-Dawley strain divided into 6 groups. Normal group was given 0.5% of CMC, the negative control group was given0.025 ml of sodium iodoacetate in 0.9% saline, the positive groups control group was given a suspension of glucosamine-chondroitin 135 mg/200 g bb,three were given pearls grass extract in various dose 5.625 mg, 11.25 mg, and 22.5 mg, respectively. 28 days after sodium iodoacetate induction, the extractswere given orally once daily for 21 days. Measurement of inflammation of knee joint and the number of leukocytes and lymphocytes were counted on day14th, 28th, and 49th after sodium iodoacetate induction. After treatment, all rats were sacrified and all knee joints were collected to subject for histology.Results: The results showed that the extract of pearl grass in all doses was able to decrease the number of leukocytes and lymphocytes significantlyand prevent proteoglycan degradation. The results showed that the extract of pearl grass with a given dose variations have antiinflammation effectand been able to protect proteoglycan significantly.Conclusion: Doses 3 (22.5 mg/200 g BW) is the best result. These results indicate that pearl grass can be further investigated as a treatment for osteoarthritis.Keyword: Osteoarthritis, Sodium iodoacetate, Pearl grass, Hedyotis corymbosa L. The present study was designed to evaluate the effect of nanoselenium (Nano-Se) on hematological and biochemical parameters of rats experimentally intoxicated with lead (Pb). Thirty male rats were randomly divided into six groups as follows: the control, selenite, Nano-Se, Pb group, Pb + selenite, and Pb + Nano-Se groups. After 35 days, blood was collected from rats and hematology and serum biochemical parameters of oxidative stress were measured. The thiobarbituric acid reactive substances (TBARS) level of Pb group was significantly higher than other groups. Also, TBARS level was significantly lower in the Pb + Nano-Se group than Pb + selenite group. The serum superoxide dismutase activities were significantly lower in Pb group than the control, Pb + selenite, and Pb + Nano-Se groups. The catalase activities in the Pb group showed no significant change when compared to other groups. In the Pb group, packed cell volume was lower than the control group. A significant difference was observed between the control group and the Pb, Pb + selenite, and Pb + Nano-Se groups. In the Pb group, the numbers of white blood cell (WBC) decreased in comparison with the control group. Also, there was significant increase in WBC counts in the Pb + Nano-Se and Pb + selenite groups in comparison with Pb group. The number of lymphocytes in the Pb group decreased in comparison with the control group. By comparing the means of the Pb + Nano-Se and Pb + selenite groups together, it was determined that there were significant differences in the lymphocytes and neutrophil counts. In conclusion, usage of selenium compounds particularly Nano-Se particles inhibits the adverse effects of Pb on antioxidant activity and immune system function in the Pb poisoning.

BACKGROUND: Moringa stenopetala is used as nourishments, and treatment of various diseases. However, there is no much information on its safety. Hence, this study aimed to investigate the chronic administration of aqueous leaves extract of the plant.MATERIALS AND METHODS: Twenty-four rats were divided into: a control group administered with distilled water and three experimental groups, respectively, administered with the extract at doses of 500, 1000, and 2000 mg/kg orally for six months were investigated. Various hematological and biochemical parameters followed by histopathological analysis were evaluated.RESULTS: Treatment with the extract did not significantly affect most of the hematological parameters. However, there were a significant decrease of MCH at doses of 1000 mg/kg and 2000 mg/kg in male rats and increase of MCV at all doses in female rats. Levels of ALP at 2000 mg/kg and those of AST and ALT at 1000 and 2000 mg/kg were significantly increased in male rats. Furthermore, significant decrease in urea and increase in creatinine levels at the dose of 2000 mg/kg occurred in female rats. Mild histopathological changes were also observed in the liver of male rats and kidney of female rats treated with the extract, respectively at doses of 1000 and 2000 mg/kg, and 2000 mg/kg.CONCLUSION: Findings from the present study suggest that prolonged administration of extract of Moringa stenopetala at therapeutic doses is safe, but shows sign of mild toxicity as dose increases, with differential effect on male verses female rats.

Hypertension is one of the most common and most prevalent diseases in the world today. Chronic inflammation is one of the risk factors for cardiovascular disease (CVD). Since synovial inflammation plays a role in the initial stages of osteoarthritis (OA), therefore, the side effects of OA are the incidence of CVD. One of the most appropriate methods for treating this disease is the use of medicinal herbs that have a blood pressure modifying effect Due to OA as Papaver rhoeas. The present research shows The Effects of Extract of Papaver rhoeas on Blood Pressure of rats with OA and Its Interaction with nitrergic System. OA model in 35 male rats was created by injection of monosodium iodoacetate (MIA) into the right knee joint and then were gavaged with extract of Papaver rhoeas for 30 days.  On day 30, the veins and arteries of the animal were then cannulated for injection and measurement of blood pressure, respectively. Blood pressure parameters (systolic, diastolic, mean arterial pressure) were recorded in groups before and after L-NAME injection. The results were analyzed using SPSS software. Systolic, diastolic, and mean arterial pressure in response to extract of Papaver rhoeas was reduced compared to control(OA), as well as Significant decrease in systolic and diastolic pressure and mean arterial pressure and increased heart beat rate in the presence of extract and LNAME as compared with control group. (P <0/05) .We found that the aqueous extract of Papaver rhoeas has a decreasing effect on blood pressure in rats with OA. This effect may be due to inhibition of the nitrergic system.

The aim: to determine the effect of a preparative form of an antiemeric agent based on maduramycin and nicarbazine on the body of white rats and broiler chickens under the conditions of a subacute experiment. Materials and methods. The formulation based on maduramycin and nicarbazine was administered to rats and broiler chickens with feed for 28 days. One control and three experimental groups were formed for the experiment: Group I – animals received a complete diet without admixture of the formulation (control group), Group II – animals that were administered the formulation (by the amount of active substances) at a dose of 5.0 mg / kg, III – 25.0 mg / kg and IV – 50.0 mg / kg of feed, respectively. In order to establish the toxic effect of the formulation on the body of experimental birds on 7, 14, 28 days of the experiment and 7 days after discontinuation of the formulation, 5 heads from each group were killed under light ether anesthesia, blood samples were taken for hematological and biochemical studies. Results. During the study of the general clinical condition of rats and broiler chickens of the experimental groups, no significant changes in behavior and appearance were detected, compared with the control. Hematological parameters of broiler chickens it was found that the receipt of the formulation with feed at doses of 5, 25 and 50 mg/kg body weight, causes a decrease in hemoglobin and an increase in hematocrit in broiler chickens on day 7 of the experiment. It was found that the changes are reversible, on the 28th day of the experiment the value of these indicators was within the physiological norm. In the study of hematological and biochemical parameters of rats, it was found that the receipt of the formulation with food in doses of 25.0 and 50.0 mg/kg for 28 days, causes a decrease in hemoglobin and increase hematocrit, total protein, albumin, creatinine and urea in male rats on the 7th day of the experiment. Conclusions. Under conditions of repeated oral administration of the preparation mixture to rats and poultry at doses of 5, 25 and 50 mg/kg body weight in the absence of clinical signs of poisoning, insignificant fluctuations in hematological and biochemical parameters were identified

Abstract. Alterations of essential elements in the brain are associated with the pathophysiology of many neuropsychiatric disorders. It is known that chronic/overwhelming stress may cause some anxiety and/or depression. We aimed to investigate the effects of two different chronic immobilization stress protocols on anxiety-related behaviors and brain minerals. Adult male Wistar rats were divided into 3 groups as follows ( n = 10/group): control, immobilization stress-1 (45 minutes daily for 7-day) and immobilization stress-2 (45 minutes twice a day for 7-day). Stress-related behaviors were evaluated by open field test and forced swimming test. In the immobilization stress-1 and immobilization stress-2 groups, percentage of time spent in the central area (6.38 ± 0.41% and 6.28 ± 1.03% respectively, p < 0.05) and rearing frequency (2.75 ± 0.41 and 3.85 ± 0.46, p < 0.01 and p < 0.05, respectively) were lower, latency to center area (49.11 ± 5.87 s and 44.92 ± 8.04 s, p < 0.01 and p < 0.01, respectively), were higher than the control group (8.65 ± 0.49%, 5.37 ± 0.44 and 15.3 ± 3.32 s, respectively). In the immobilization stress-1 group, zinc (12.65 ± 0.1 ppm, p < 0.001), magnesium (170.4 ± 1.7 ppm, p < 0.005) and phosphate (2.76 ± 0.1 ppm, p < 0.05) levels were lower than the control group (13.87 ± 0.16 ppm, 179.31 ± 1.87 ppm and 3.11 ± 0.06 ppm, respectively). In the immobilization stress-2 group, magnesium (171.56 ± 1.87 ppm, p < 0.05), phosphate (2.44 ± 0.07 ppm, p < 0.001) levels were lower, and manganese (373.68 ± 5.76 ppb, p < 0.001) and copper (2.79 ± 0.15 ppm, p < 0.05) levels were higher than the control group (179.31 ± 1.87 ppm, 3.11 ± 0.06 ppm, 327.25 ± 8.35 ppb and 2.45 ± 0.05 ppm, respectively). Our results indicated that 7-day chronic immobilization stress increased anxiety-related behaviors in both stress groups. Zinc, magnesium, phosphate, copper and manganese levels were affected in the brain.

Abstract. It has been suggested that treatment of intact male rats with oestradiol benzoate (OeB) causes an interference with testosterone (T) production by the testes by a direct inhibitory effect on steroidogenesis. To test this hypothesis, different doses (5, 10 or 25 IU) of hCG were administered concomitantly with 50 μg of OeB to adult intact or hypophysectomized male rats. The testicular and plasma testosterone, and serum hCG levels were determined. The sex accessory weights were recorded. In the intact OeB-treated group of animals, hCG stimulated both the secondary sex organs and plasma testosterone levels above the intact control group. However, in hypophysectomized animals, although plasma testosterone levels increased above that of intact controls, their secondary sex organ weights did not. Moreover, inspite of high circulating hCG levels, the testicular testosterone content and concentration remained suppressed in OeB-treated animals. The reason for such dichotomy of hCG action on OeB-treated animals is not clear at present.
