Impact of supplementation of probiotic, prebiotic and synbiotic on serum biochemical profile of crossbred calves

A study was conducted on 24 fifteen day old crossbred calves that were divided into 4 groups of 6 calves each. The calves were fed different dietary treatments viz T0 (control group) - basal diet, T1 (probiotic group) - basal diet + Lactobacillus acidophilus @ 1 g/calf/day (2 x 1010 cfu/ g), T2 (prebiotic group)- basal diet + Mannan oligosaccharide (MOS) @ 4 g per calf/day and T3 (synbiotic group) basal diet + Lactobacillus acidophilus @ 0.5 gm (2 x 1010 cfu/ g) + 2 g MOS per calf/day respectively for a period of 90 days. Blood samples were taken at monthly interval and serum was separated to estimate different biochemical parameters. The results of study indicated that probiotic (Lactobacillus acidophilus), prebiotic (MOS) and synbiotic supplementation were effective in reduction of serum cholesterol and triglycerides at 60th and 90th day. Serum creatinine, glucose, urea and bilirubin were unaffected by dietary supplementation of probiotic, prebiotic (MOS) and synbiotic. There was no effect of T1, T2 and T3 supplementation on Serum enzymes, Aspartate aminotransferase (AST) and Alanine Amino Transferase (ALT).

The objective of this study was to evaluate the effect of dietary supplementation with selenium (Se) and vitamin E (VitE) on milk yield, composition and renneting parameters in dairy Jonica breed goats. Forty multiparous lactating goats fed the same basal diet were randomly allocated to 1 of 2 dietary treatments: for 135 days, the experimental period. Basal diet was composed by ad libitum mixed vetch-oat hay and a definite amount of a pelleted concentrate. Dietary treatments included: (1) the control diet containing background Se and VitE only, and (2) the experimental diet containing sodium selenite as the Se supplement in concentrate (0·20 mg/head per day of Na-selenite and 20 mg/day per head of VitE). During the trial, no significant differences (P>0·05) were registered in dry matter intake and body weight gain among groups. Goats fed Se-VitE diet showed the highest milk production (P<0·05), and fat (P<0·01) and protein (P<0·05) yields and percentages. Somatic cell count were significantly lower in Se-VitE diet compared with control group (P<0·05). Significant differences (P<0·05) were observed in milk renneting properties. Based on our results, it was concluded that dietary supplementation with SE and VitE led to an increase of milk production and quality in Jonica goat, with positively improvement in milk coagulation aptitude.

Recent evidences suggest that gut microbiota plays an important role in regulating physiological and metabolic activities of pregnant sows, and β-carotene has a potentially positive effect on reproduction, but the impact of β-carotene on gut microbiota in pregnant sows remains unknown. This study aimed to explore the effect and mechanisms of β-carotene on the reproductive performance of sows from the aspect of gut microbiota. A total of 48 hybrid pregnant sows (Landrace × Yorkshire) with similar parity were randomly allocated into three groups (n = 16) and fed with a basal diet or a diet containing 30 or 90 mg/kg of β-carotene from day 90 of gestation until parturition. Dietary supplementation of 30 or 90 mg/kg β-carotene increased the number of live birth to 11.82 ± 1.54 and 12.29 ± 2.09, respectively, while the control group was 11.00 ± 1.41 (P = 0.201). Moreover, β-carotene increased significantly the serum nitric oxide (NO) level and glutathione peroxidase (GSH-Px) activity (P &lt; 0.05). Characterization of fecal microbiota revealed that 90 mg/kg β-carotene increased the diversity of the gut flora (P &lt; 0.05). In particular, β-carotene decreased the relative abundance of Firmicutes including Lachnospiraceae AC2044 group, Lachnospiraceae NK4B4 group and Ruminococcaceae UCG-008, but enriched Proteobacteria including Bilophila and Sutterella, and Actinobacteria including Corynebacterium and Corynebacterium 1 which are related to NO synthesis. These data demonstrated that dietary supplementation of β-carotene may increase antioxidant enzyme activity and NO, an important vasodilator to promote the neonatal blood circulation, through regulating gut microbiota in sows.

Alternative growth promoters are able to not only effectively replace the traditional use of antibiotics but also provide additional health benefits for livestock and reduce food safety concerns. This study investigated the effects of dry Hydrastis canadensis on the laying performance and fecal microbial community of laying hens. Twenty-four Lohmann (LSL, white layer strain) hens were reared from 40 to 48 weeks of age and randomly allotted to four dietary treatments (six birds/treatment). The dietary treatments comprised a basal diet with no treatment as control, a basal diet plus 0.6% powder of dry Hydrastis canadensis roots (R) or leaves (L), and a basal diet plus 0.6% powder of a mixture of dry Hydrastis canadensis roots and leaves (1:1, LR). No mortality was observed in the whole experimental period. The results indicated that albumen height in the LR group was significantly greater than that in the control group. The diet supplemented with Hydrastis canadensis had no significant effects on egg production rate, egg weight, eggshell strength, eggshell thickness, Haugh unit, or yolk height during the whole experimental phase. However, principal coordinate analysis, comparative heat map analysis, and cluster dendrogram analysis of cecal microbiota showed distinct clusters among the groups treated with Hydrastis canadensis and the control group. Regarding blood biochemical parameters, serum cholesterol levels were significantly lower in all Hydrastis canadensis-treated groups compared with those in the control group. Moreover, serum low-density lipoprotein levels were lower in hens supplemented with the leaf of Hydrastis canadensis. The abundances of the phyla Fusobacteria and Kiritimatiellaeota were increased (p < 0.05) in laying hens fed with 0.6% Hydrastis canadensis leaves, whereas the abundance of the phylum Firmicutes in cecum digesta decreased in response to treatment with Hydrastis canadensis roots and leaves. The relative abundance of the Fusobacterium genus was higher in the LR group compared with that in the control. On the contrary, we found a different trend in the Synergistes genus. The potential influences of these microbiota on the performance of laying hens were discussed. The results demonstrate that Hydrastis canadensis can improve the egg albumen height and modulate the cecum digesta microbiota composition of laying hens.

AbstractPurslane is a widespread wild vegetable with both medicinal and edible properties. It is highly appreciated for its high nutritional value and is also considered as a high-quality feed resource for livestock and poultry. In this study, Sanhuang broilers were used to investigate the effect of feeding purslane diets on the growth performance in broilers and their gut microbiota. A total of 48 birds with good growth and uniform weight were selected and randomly allocated to four treatment groups A (control), B, C and D. Dietary treatments were fed with basal diet without purslane and diets containing 1%, 2% and 3% purslane. The 16S rDNA was amplified by PCR and sequenced using the Illumina HiSeq platform to analyze the composition and diversity of gut microbiota in the four sets of samples. The results showed that dietary inclusion of 2% and 3% purslane could significantly improve the growth performance and reduce the feed conversion ratio. Microbial diversity analysis indicated that the composition of gut microbiota of Sanhuang broilers mainly included Gallibacterium, Bacteroides and Escherichia-Shigella, etc. As the content of purslane was increased, the abundance of Lactobacillus increased significantly, and Escherichia-Shigella decreased. LEfSe analysis revealed that Bacteroides_caecigallinarum, Lachnospiraceae, Lactobacillales and Firmicutes had significant differences compared with the control group. PICRUSt analysis revealed bacteria mainly enriched in carbohydrate metabolism pathway due to the additon of purslane in the diet. These results suggest that the addition of purslane to feed could increase the abundance of Lactobacillus in intestine, modulate the environment of gut microbiota and promote the metabolism of carbohydrates to improve its growth performance. This study indicates that the effect of purslane on the growth-promoting performance of broilers might depend on its modulation on gut microbiota, so as to provide a certain scientific basis for the application of purslane in the feed industry.

Indole-3 carbinol (I3C) has shown dual effects on the promotion and progression stages of colon carcinogenesis while synbiotics (Syn) have exerted anti-carcinogenic activities in most rodent studies. This study aimed to investigate the effects of I3C given alone or together with a Syn intervention on 1,2-dimethylhydrazine (DMH)-induced colon carcinogenesis. All animals were given four subcutaneous DMH injections (4×40 mg/kg bodyweight, twice a week for two weeks) and then received either basal diet (G1), basal diet containing I3C (1g/kg chow) (G2) or basal diet containing I3C+Syn (I3C + inulin 50g/kg chow + Bifidobacterium lactis BB-12®), 2.5×1010 cfu/g of basal diet), (G3) for 21 weeks. Dietary I3C (G2) significantly increased tumour volume and cell proliferation when compared to the DMH control group (G1). Syn intervention (G3) significantly reduced tumour volume and cell proliferation when compared to I3C (G2). The colon tumours found were classified into well-differentiated tubular adenomas or adenocarcinomas. Dietary I3C or I3C+Syn did not significantly affect the incidence and the multiplicity of tumours in comparison with the DMH control group. Furthermore, Syn intervention (G3) increased Gstm1 and reduced Mapk9 gene expression in colonic tumours. The findings of the present study show that the dietary I3C shows a weak promoting activity, while the combination with Syn ameliorates I3C effects.

The regulation of glycerol permeability in the gastrointestinal tract is crucial to control fat deposition, lipolysis and gluconeogenesis. Knowing that the amino acid glutamine is a physiological regulator of gluconeogenesis, whereas cystine promotes adiposity, herein we investigated the effects of dietary supplementation with glutamine and cystine on the serum biochemical parameters of piglets fed on amino acid-enriched diets, as well as on the transcriptional profile of membrane water and glycerol channels aquaporins (AQPs) in the ileum portion of the small intestine and its impact on intestinal permeability. Twenty male piglets with an initial body weight of 8.8 ± 0.89 kg were allocated to four dietary treatments (n = 5) and received, during a four week-period, a basal diet without supplementation (control) or supplemented with 8 kg/ton of glutamine (Gln), cystine (Cys) or the combination of the two amino acids in equal proportions (Gln + Cys). Most biochemical parameters were found improved in piglets fed Gln and Cys diet. mRNA levels of AQP3 were found predominant over the others. Both amino acids, individually or combined, were responsible for a consistent downregulation of AQP1, AQP7 and AQP10, without impacting on water permeability. Conversely, Cys enriched diet upregulated AQP3 enhancing basolateral membranes glycerol permeability and downregulating glycerol kinase (GK) of intestinal cells. Altogether, our data reveal that amino acids dietary supplementation can modulate intestinal AQPs expression and unveil AQP3 as a promising target for adipogenesis regulation.

This study aimed to investigate the efficacy of compound probiotics on broiler performance, intestinal microflora, immune function and serum biochemical indicators. Four hundred one-d-old female broiler chickens were randomly divided into 4 groups with 4 replicates in each group and 25 chickens per replication. The chickens in groupI(control group) were fed a basal diet, experimental groups II, III and IVwere fed the basal diet supplemented with 0.1%, 0.2% and 0.4% compound probiotics. The results of this work were as follows: (1) during the experimental period, compared with control group, the average daily in group III was significantly increased (P<0.05), the feed/gain and average daily feed intake were significantly lower (P<0.05); (2) at the age of 14 and 28 days, the number of cecal Lactic acid in group II and III were significantly higher than the control (P<0.05) and the content of E.coli has the opposite results. (3) the anti-body titer in group III was the highest, and significantly higher compared to the control (P<0.05), but there were no significantly differences between group IV and control group (P>0.05). (4) serum biochemical indexes of broilers in group III and IVwere significantly increased than that in control group (P<0.05). In conclusion, the supplementation of 0.2% compound probiotics could promote the growth performance, improve the cecal mico-ecosystem, enhance the anti-body titer and biochemical indexes in serum.

Animals fed with a high amount of a wide range of antioxidants in their diet are significantly protected against oxidative stress. Powerful antioxidant substances such as vitamin E, vitamin C, and carotenoids are present naturally in red-hot pepper (RHP). This study hypothesized that using RHP may provide protection against oxidative stress and enhance animal physiological responses. Thus, this study aimed to investigate the effect of feeding New Zealand white rabbits with RHP-supplemented diets on their physiological and biochemical responses. New Zealand White rabbits (age = 6 weeks, n = 48) were split equally into three groups (n = 16 in each group). One group was fed a basal diet only (control group), with the other two groups fed a basal diet along with 1 and 2% RHP. Mass spectrometric analysis for the RHP methanolic extract showed some phenolic compounds, such as p-coumaric, sinapinic acids, vanillic, and luteolin, as well as catechin and its isomers. Hepatic antioxidant enzymes (SOD, GSH, GSH-Px, and CAT) were significantly elevated (p < 0.05) by feeding rabbits diets supplemented with 1 or 2% RHP. The addition of RHP significantly enhanced immune-responses; phagocytic activity, chemotaxis, TIg, IgG, IgM, and IgA increased when growing rabbits were fed RHP compared with the control group. In conclusion, dietary supplementation of 1 or 2% RHP may play a role as an enhancer of growth and immune response in growing rabbits.

Abstract This study was performed to investigate the effects of dietary supplementation with garlic and turmeric powder as growth promoter agents on performance, carcass traits, serum biochemistry, and antioxidant enzyme activities of growing rabbits. A total of 112 New Zealand White rabbits (NZW) at 5 weeks of age were randomly assigned to seven treatments with four replicates. The dietary treatments consisted of 7 groups as follows; the basal diet as control, phytogenic additives groups were supplemented with 2, 4, and 6 g/kg garlic or turmeric powder added to the basal diet. There were no linear and quadratic differences (P<0.05) in growth performance after garlic or turmeric supplementation at all studied ages. Compared with the control group, supplementation of diets with garlic or turmeric linearly and quadratically elevated immunity biomarkers such as total protein (TP), albumin (AL) and immunoglobulin (IgG) levels and decreased (linearly and quadratically, P<0.05) aspartate aminotransferase (AST), alanine aminotransferase (ALT ), triglyceride (TG) and total cholesterol (TC) levels in rabbit serum. However, TP, AL, globulin (GL), IgG and IgM levels were linearly and quadratically enhanced with increasing turmeric levels versus the control diet. Hepatic superoxide dismutase (SOD), catalase (CAT ) and glutathione peroxidase GSH-Px activities as well as reduced glutathione (GSH) concentrations were linearly and quadratically (P<0.05) improved in garlic or turmeric additives fed groups. While MDA concentration was statistically (linearly, P= 0.022) reduced in comparison with the control group. It could be concluded that garlic or turmeric supplementation (2, 4 and 6 g/kg) did not linearly and quadratically affect growth performance but improved the immunity responses and lowered the lipid profile in blood and lipid peroxidation in liver and increased hepatic antioxidant activity in treated rabbits.

This study was conducted to investigate the effects of dietary supplementation with different types of Bacillus subtilis (B. subtilis) on the growth and gut health of weaned piglets. A total of 160 piglets were randomly assigned into four groups: control group (a basal diet), BS-A group (a basal diet supplemented with B. subtilis A at 1 × 106 CFU/g feed), BS-B group (a basal diet supplemented with B. subtilis B at 1 × 106 CFU/g feed), and BS-C group (a basal diet supplemented with B. subtilis C at 1 × 106 CFU/g feed). All groups had five replicates with eight piglets per replicate. On days 7, 21, and 42 of the trial, blood plasma and intestinal tissues and digesta samples were collected to determine plasma cytokine concentrations, intestinal morphology, gut microbiota community and metabolic activity, and the expression of genes related to gut physiology and metabolism. The results showed that dietary B. subtilis supplementation improved (P &lt; 0.05) the body weight and average daily gain (in BS-B and BS-C groups) of weaned piglets and decreased (P &lt; 0.05) the diarrhea rates (in BS-A, BS-B, and BS-C groups). In the intestinal morphology analysis, B. subtilis supplementation improved (P &lt; 0.05) the size of villus height and villus height to crypt depth ratio in the ileum of weaned piglets. Firmicutes, Bacteroidetes, and Tenericutes were the most dominant microflora in piglets' colon whatever the trial group and time of analysis. Dietary BS-C supplementation increased (P &lt; 0.05) the relative abundances of Anaerovibrio and Bulleidia and decreased (P &lt; 0.05) the relative abundances of Clostridium and Coprococcus compared with the control group. In addition, dietary B. subtilis supplementation increased (P &lt; 0.05) the indicators of intestinal health, including plasma levels of interleukin (IL)-2 and IL-10, as well as the colonic levels of short-chain fatty acids. Furthermore, dietary B. subtilis supplementation also up-regulated (P &lt; 0.05) the expression of genes involved in metabolic pathways related to intestinal microbiota maturation. In conclusion, these findings suggest that a diet containing BS-B or BS-C can efficiently promote growth performance, decrease diarrhea incidence, and ameliorate several indicators of intestinal health through the modulation of gut microbiota composition and metabolic activity in weaned piglets.
