Recently, the substantial progress has been made toward understanding the human microbiota as an ecosystem that functions as a separate organ in a macro-organism. More and more data on the microbiome as the aggregate of microorganism genes that reside on or within the human body appear. Deterioration of the environment, changes in food production technologies, widespread use of antibiotics result in serious human microbiota disorders, most often in the form of gut dysbiosis of varying severity. The investigation into the causes and the correct choice of a probiotic containing the identified microorganisms with the indication of genus, species, strain and their quantitative composition is the criterion for the success of dysbiosis therapy. The purpose of this article is to discuss the criteria for rational probiotic therapy and prevention of gut dysbiosis and associated diseases. It should be considered that the treatment of diseases accompanied by dysbiosis should be comprehensive and include the elimination of the cause of the disease, the correction of nutrition, the elimination of pathogenic agents and their toxins in the gastrointestinal tract, as well as the restoration of the microbiota composition and functional activity. Probiotics play an important role in restoring the composition and functional activity of microbiota. According to the current guidelines of the World Gastroenterological Organization (WGO), a certain quantity of bacteria (KOE of microorganisms) that is necessary to achieve a  positive effect is an important criterion for the efficacy of probiotic drugs.

The results of tests of a new combined feed additive Bioprotektin-KD based on milk thistle extract with the addition of a complex of bacteria of the genus Lactobacillus and Bifidobacterium to restore intestinal microbiota of animals are presented. Preclinical studies on the effectiveness of the feed additive were conducted on mice with experimental antibiotic-associated dysbacteriosis. Studies on the intestinal microbiota of mice revealed varying degrees of effect of different doses of feed additive on the recovery of resident microflora. The results of pre-clinical studies have proven the harmlessness, non-toxicity and efficacy of the feed additive. Clinical trials of the additive were carried out on productive animals. Its effect on the productivity of black-motley breed cows in a livestock farm in the Belgorod region was studied. The use of the functional feed additive Bioprotectin-KD had a positive effect on the milk productivity of black-motley breed cows. Cows in the experimental groups that received the feed additive had increased their milk yield per lactation by 209.41-499.01 kg compared to the control animals, which was 4.01-8.78%. The quality indicators of milk have increased. The composition of the microflora of the experimental (feed supplement recipients) and control calves was studied. It was found that the quantitative composition of probiotic flora in the intestines of young calves of the control group remained practically unchanged throughout the experiment. The number of indigeneous microflora (lactobacilli and bifidobacteria) increased in the experimental animals. The content of lactic acid microorganisms in the experimental calves began to increase on day 6 of feed supplementation and reached physiological norm on day 30. The safety index of the product has been calculated. The use of the functional feed additive Bioprotektin-KD in conditions of a livestock complex has improved such production indicators as calf survival, average daily weight gain, and milk productivity of cows in the experimental groups of animals as compared to the control ones.

Demands for higher production and consumer demand for healthier food have encouraged intensive research for alternative animal growth promoters in recent years. Research trends are focused on the development of new products enriched with feed additives to improve animal health and enhancing production. Many of these compounds are unstable in the presence of light, air, water, or high temperatures and need to be protected during processing, storage, and application. By encapsulatied in microparticles, feed additives are protected from harmful external influences, and their stability and functionality are not diminished. Microencapsulation technology is used to protect payload and improve bioavailability by controlled and targeted delivery to the digestive tract. It is particularly suitable for the addition of feed additives in ruminant’s nutrition, because the correct choice of microparticle material allows the release of the feed additives in the small intestine, and not in the rumen. With proper use, microparticle formulations are an effective tool in animal nutrition that delivers nutrients and/or drugs to a specific site at the desired rate. The paper summarizes laboratory studies on the application of microencapsulation technology in feeding ruminants and monogastric animals. Numerous results of the application of encapsulated feed additives have shown a positive effect on animal health, increased productivity without negative effects on the final product, and environmental protection.

The existing data indicate the multifactorial mechanisms of development of the overactive bladder (OAB) symptom, but the issue of OAB pathogenesis remains unclear. In more recent times, the neurogenic theory of OAB genesis has being accompanied by the increasing attention to the study of morphological changes that occur in the smooth myocytes of the detrusor and their interaction with the extracellular matrix. Therefore, the objective of our study became the evaluation of distribution of interstitial Cajal cells (ICC) and basophilic granulocytes (BG) in the structural elements of the bladder wall under stress urinary incontinence and its overactivity before and after treatment with Mirabegron, Spasmex, Quercetin and combination thereof with testosterone and estradiol, using histochemical and immunohistochemical methods. The experimental models of OAB and stress urinary incontinence (SUI) presented the increase in the amount and functional activity of BG revealed by histological and immunohistochemical methods, as well as ICC at all terms of OAB monitoring, while the SUI presented with high concentration and functional activity of BG only after 14 days of the experiment. After 28 days, we observed a sharp decrease of the parameters, indicating decompensation and depletion of the functional activity. The number of ICC decreased under SUI after both 14 days and 28 days of the experiment. The group of experimental animals receiving Spasmex and its combination with hormones, presented no significant effect on the quantitative and qualitative composition of BG and ICC at OAB and SUI on Day 14 of the experiment, but the combination with testosterone demonstrated statistically reliable (p<0.001) reduction of BG and ICC expression in the muscle layer of the bladder after 14 days, and unreliable after 28 days of the experiment (p>0.05). The administration of Mirabegron alone and especially its combination with testosterone and estradiol presented positive trends in histochemical and immunohistochemical expression of BG and ICC. The experiment proved high efficacy of Quercetin in combination with testosterone and estradiol under OAB and SUI, confirmed by stabilization of the functional activity of BG and ICC quantitative composition.

The immune system as a whole plays a leading role in the formation of an adequate immune response during the onset and development of tumors. The aim of the study was to reveal the features of the change in the morphology and level of biogenic amines of the thymus of rats during experimental carcinogenesis of breast cancer by the injection of N-methyl-N-nitrosourea. In the work used luminescence-histochemical, general histological and immunohistochemical methods, which made it possible to obtain important information about the functional activity of the cellular elements of the thymus, the quantitative composition of macrophages, dendritic cells, lymphocytes and the qualitative characteristics of mast cells. The experiment was carried out on 85 white nonlinear female rats, which were injected with the carcinogen N-methyl-N-nitrosourea, which induces a breast tumor. Animals were divided into two groups: intact (n=30) and rats injected with a carcinogen (n=45). The thymus structure was examined 1, 2, and 3 months after the end of the injections. During the study, changes in the morphometric parameters of the organ are noted. A month after the injection of the drug, the area of the medulla of the thymus and the thickness of the cortex substance of the organ increases, which decrease with an increase in the study period. Replacement of the thymus parenchyma with adipose and connective tissue begins after 2 months and progresses with an increase in the study period. A tendency to wavelike changes in biogenic amines was revealed, after 1 month there is a significant increase in the level of histamine and a decrease in serotonin and catecholamines, after 2 months an increase in all biogenic amines, especially histamine (1.5 times or more) in premedullary and mast cells, thymocytes of the cortex. After 3 months, on the contrary, there is a decrease in histamine and an increase in the level of monoamines. The ratio (serotonin+histamine)/catecholamines after 1 month indicate a decrease in the functional activity of the thymus, and after 3 months - about its increase. At all stages of the study, degranulated forms of mast cells predominate. In the dynamics of the experiment, an increase in the number of dendritic cells, lymphocytes, and twice the number of macrophages and Bcl-2+cells was revealed. The data obtained in the course of the study suggested that the introduction of N-methyl-N-nitrosourea and the subsequent development of a breast tumor is reflected in the change in the morphofunctional state of the thymus. In this case, the greatest changes were observed 2 months after the end of the administration of the carcinogen, even before the formation of a breast tumor. The data obtained can have a certain value in determining the immune status in models of experimental carcinogenesis.

Introduction. Dry eye syndrome (DES) is a multifactorial disease of the ocular surface, characterized by changes in its homeostasis and accompanied by ocular symptoms, the etiology of which is associated with destabilization of the tear film, hyperosmolarity, inflammation, damage to the structures of the ocular surface and neurosensory disorders.Objective. To study the effect of a drug containing plastoquinonyldecyltriphenylphosphonium bromide (PDTP, SkQ1) on dynamics of osmolarity and reparative properties of the cornea.Materials and methods. The study included 23 patients (46 eyes) aged 23 to 49 years. All patients presented complaints of dryness, burning sensation, cramps and discomfort in the eyes, increased sensitivity with instillation of drops, periodic blurred vision. Patients underwent standard and special ophthalmic examinations. The patients were divided into two groups comparable in gender and quantitative composition.Results. Based on the results of the examination at the starting visit, all patients were diagnosed with DES mild or moderate study. After one week of therapy, a significant decrease in subjective complaints was noted patients who received the drug with PDTF. All patients tolerated the drug well. In the group of patients who received a tear replacement therapy with hypromellose, in most patients, while complaints of dryness persist, decreased a feeling of pain in the eyes. A month later, complaints persisted only in patients of this group, but their intensity was known significantly reduced compared to the visit after a week. Against the background of therapy with the drug with PDTP, significant improvement of the Schirmer test, Norn test and corneal staining, as well as with the help of a in vivo confocal microscopy shows a decrease in the number of Langerhans cells.Conclusion. The results obtained indicate a positive effect of PDTP on the structure of the ocular surface even with its short-term use due to its keratoprotective and anti-inflammatory action.

AbstractDiabetes mellitus is a very important comorbidity in patients with heart failure, as the common presence of both diseases significantly worsens the prognosis of patients. In order to improve the outcome of these patients, it is essential to diagnose both diseases at an early stage and to treat them in accordance with guidelines. In particular, a differentiated medication plays a crucial role. The therapy of heart failure does not differ in patients with diabetes and patients without diabetes. In the treatment of diabetes mellitus, however, it is very important to choose substances that have a positive effect on the cardiovascular outcome of patients. First-line treatment of diabetes in patients with cardiovascular diseases should be metformin, followed by a SGLT-2 inhibitor or GLP-1 receptor agonist with proven cardiac benefit. A rigorous adjustment of risk factors according to current guidelines reduces cardiovascular mortality and hospitalization rates. Glitazones and saxagliptin are associated with increased hospitalization rates and should be avoided in heart failure.

As a result of the work, adequately new data were obtained on the change in the intestinal microbiocenosis of pigs during their entire life. The degree of influence of the probiotic, prebiotic and synbiotic on the change in the microbiocenosis of the intestines of pigs is determined. A scheme for the introduction of lactic starter cultures, "Baliz-V" and lactic starter cultures and "Baliz-V" together. The effect of the new diet on growth indicators, microbiocenosis, immunity and the quality of raw meat was studied. The positive effect of the synbiotic effect on the quantitative composition of lactic acid bacteria in the intestines of young pigs was 3-4 orders of magnitude higher than in the control. The use of a probiotic, prebiotic and synbiotic led to an improvement in intestinal microbiocenosis, strengthening the body's resistance, and an increase in live weight gain, respectively, by 8.4; 9.6 and 10.7%.
