Abstract              Azadirachta indica (Neem) has been shown to affect the fertility of mice by interfering with the synthesis of testosterone in mice. The aim of this study was to detect the testes protein expression of mice after exposure to the ethanolic Neem leaf extract. The laboratory animals of this study were 20 male Swiss Webster mice with three months in age and body weight ranging from 27.5 grams. The mice were divided into two treatment groups, namely K (control group, exposed with distilled water) and P (treatment group, exposed to etahnolic Neem leaf  extract with 14 mg/animal /day). The treated were given for 21 days and the testicular protein was carried out on the 22nd day. The variables observed were testes weight, concentration and expression of proteins isolated from the testes. The protein concentration is determined by a spectrophotometer at a wavelength of 450nm. The protein expression was observed and determined based on the results of protein electrophoresis (SDS-PAGE). The results showed that protein expression in the treatment group has a lower concentration compared to the control group. Those results  was confirmed by thinner bands in SDS-PAGE result. Those proteins thought to be a fertility determinant in mammals. Keywords : anti-fertility; Neem; protein expression Abstrak  Azadirachta indica (Mimba) telah terbukti mempengaruhi fertilitas mencit dengan cara mengganggu sintesis hormon testoteron pada mencit. Penelitian ini bertujuan untuk melacak ekspresi protein pada testis mencit setelah paparan ekstrak etanol daun Mimba. Hewan uji penelitian ini adalah 20 ekor mencit Swiss Webster jantan dengan umur tiga bulan dan bobot badan berkisar 27.5 gram. Hewan uji dibagi menjadi 2 kelompok perlakuan, yaitu K (kelompok kontrol, dipapar akuades) dan  P (kelompok perlakuan, dipapar dengan ekstrak etanol daun Mimba dengan dosis 14 mg/ekor/hari). Pemberian bahan uji dilakukan  secara oral selama 21 hari. Variabel yang diamati adalah bobot testes, konsentrasi serta eskpresi protein yang diisolasi dari testis. Isolasi protein testis dilakukan pada hari ke-22. Konsentrasi protein ditentukan dengan spectrofotometer pada panjang gelombang 450nm. Ekspresi protein diamati dan ditentukan berdasar hasil elektroforesa protein. Hasil penelitian menunjukkan bahwa ekspresi protein pada kelompok perlakuan menunjukkan konsentrasi yang lebih rendah dengan pita yang lebih tipis jika dibandingkan dengan kelompok kontrol. Kesimpulan penelitian ini adalah paparan ekstrak etanol daun Nimba menyebabkan gangguan ekspresi protein yang diduga berperan dalam menentukan fertilitas mamalia. Kata kunci : Mimba; anti-fertilitas; eskpresi protein;

The aim of this study was to determine the effect of neem leaf extract (Azadirachta indica) on parasitemia of rats infected with Trypanosoma evansi (T. evansi) Aceh local isolate. A total of 24 male rats aged three months were used in this study and randomly divided into six treatment groups equally. The negative control group (K0) without T. evansi infection and neem leaf extract, the positive control group (K1) was infected with T. evansi but no neem leaf extract given, group K2, K3, K4, and K5 were infected with 5x104 T. evansi and were given neem leaf extract after patent infection with dose of 50, 100, 400, and 800 mg/kg BW respectively. The extract was given orally for three consecutive days. On the fourth day, rat blood was drawn for parasitemia examination. The results showed that no T. evansi detected in rats in negative control group (K0), while parasitemia in group K1; K2; K3; K4; and K5 was 12,295 x106/mL; 10,495 x106/mL; 9,360 x106/mL; 5,080x106/mL; and 2,398x106/mL of blood, respectively. Percentage of inhibition of parasitemia in K2, K3, K4, and K5 reached 14.64, 23.78, 58.68, and 80.50%, respectively. Based on the result of the study, neem leaf extract of 800 mg/kg BW gave the highest reduction of parasitemia in rats infected with T. evansi.

<p align="center"><strong>ABSTRAK</strong></p><p><strong>Latar belakang: </strong>Trombositopenia merupakan suatu keadaan jumlah trombosit di bawah 150.000/mm<sup>3</sup>, hal ini dapat menyebabkan perdarahan yang apabila tidak ditangani dengan baik dapat berujung pada kematian. Beberapa tindakan yang dapat dilakukan untuk mengatasi  trombositopenia, salah satunya adalah dengan tanaman daun pepaya.</p><p><strong>Tujuan:</strong> untuk menganalisis pengaruh pemberian ekstrak daun pepaya terhadap <em>bleeding time</em> (waktu perdarahan).</p><p><strong>Metode : </strong>Desain penelitian ini menggunakan <em>true experimental</em> dengan rancangan <em>post test only control group design</em>. Some actions that can be done to overcome thrombocytopenia, one of which is with papaya leaf.</p><p><strong>Objectives:</strong> This study aims to analyze the effect of giving papaya leaf extract to bleeding time.</p><p><strong><em>Methods:</em></strong><strong> </strong>This research design uses true experimental with design of post-test only control group design. The sample in this study were 30 male mice (<em>Mus musculus</em>) divided into five groups. The first group as the negative control group (CMC Na 0.5%), the second group as the positive control (CMC Na 0.5%), the third group was given ethanol 96% extract papaya leaf dose 0.5g/kgBW, the fourth group was given ethanol 96% extract papaya leaf dose 1g/kgBW, the fifth group was given ethanol 96% extract papaya leaf dose 2g/kgBW. Data collection using measurement sheet of  bleeding time which were analyzed by ANOVA test.</p><p><strong><em>Results:</em></strong><strong> </strong>The results of this study showed that p&lt;0.0001 the fastest mean time occur between treatment groups that is treatment group with dose 2g/kgBW (2.74±0.14). In the bleeding time variables p&lt;0.0001 the fastest mean time occur between treatment groups that is treatment group with dose 2g/kgBW (2.74±0.14). This result shows that there are significant differences between every groups.</p><p><strong><em>Conclusion:</em></strong><strong> </strong>The conclusion of this research is giving papaya leaf extract can increase bleeding time.</p><p> </p><p><strong>KEYWORDS: </strong>leaf of papaya, bleeding time, thrombocytopenia.</p><p> </p>

BACKGROUND: Inflammatory bowel disease (IBD) is a chronic or relapsing inflammation of the intestine which consists of Crohn’s disease and ulcerative colitis. Interleukin (IL)-10 is an anti-inflammatory cytokine which plays important role in the pathogenesis of IBD. Neem (Azadirachta indica) is rich of azadiractoids which possesses anti-inflammatory activity. AIM: The objective of the study was to determine the effect of neem leaf extract on IL-10 expression and histological score in dextran sodium sulfate (DSS)-induced colitis rats. METHODS: The first phase compared 7 DSS-induced colitis rats (case group) with seven healthy rats (control group). All of them were sacrificed. The second phase compared 28 DSS-induced colitis rats treated with mesalazine (Group I) and 56 rats treated with neem leaf extract (Groups II and III). Seven rats from each group were sacrificed on days 7, 14, 21, and 28. Colon samples were harvested and underwent histopathological examination and immunohistochemical analysis to determine IL-10 expression and histological score. RESULTS: IL-10 expression and histological score were higher in case group compared to control group. There was similar IL-10 expression between Groups I and II on day 28 while the similarity was observed since day 7 between Groups I and III. Mean histological score was higher in Group II compared to Group I and it was similar between Group I and Group III on days 21 and 28. CONCLUSION: Neem leaf extract increased the expression of anti-inflammatory cytokine, in particular IL-10, and improved histological score in DSS-induced colitis rats.

Bleeding is when blood is removed from damaged blood vessels and can occur during dental procedures. Curry leaves (Murraya koenigii L.) contain tannins and flavonoids, which have the potential as a hemostatic agent to stop bleeding. The purpose of this study was to determine the effect of curry leaf extract (Murraya koenigii L.) on the bleeding time of Wistar rats (Rattus norvegicus). The test animals used were 5 Wistar rats (Rattus norvegicus), which were divided into a control group that was applied with distilled water and the treatment group with the extract of curry leaves. The first treatment group was given a concentration of 25%, the second treatment group had a concentration of 50%, the third treatment group had a concentration of 75%, and the fourth treatment group had a concentration of 100%. Bleeding time was calculated using the Duke method on the tail of the rats. The results showed that curry leaf extract concentrations of 25%, 50%, 75%, and 100% were able to shorten the bleeding time with an average time of 140 seconds, 81.67 seconds, 138.33 seconds, and 73.33, respectively. Second. One-way ANOVA data analysis showed that the bleeding time in all treatment groups was significantly different from the control group (p 0.05). This study concludes that the extract of curry leaves (Murraya koenigii L.) affects the bleeding time in Wistar rats (Rattus norvegicus).

Background: Candida albicans (C. albicans) are dimorphic fungi in oral cavity, considered not only as normal flora, but also as pathogens. C. albicans have an ability to grow biofilm, which has a thick layer of outer skin structure, called as extracellular matrix. Jengkol leaves (Pithecellobium jiringa) contain alkaloids, flavonoids, terpenoids, and lectins, which have an ability as antifungal agent Purpose: This study aimed to analyze the optimum dose of jengkol leaf extract as antibiofilm against C. albicans biofilms. Method: C. albicans were cultured on yeast peptone dextrosa (YPD) media in 96-well microtiter plate flat bottom plates. There were one control group (without treatment) and three treatment groups. The first treatment group was given jengkol leaf extract at a dose of 100 mg/ ml. The second treatment group was given jengkol leaf extract at a dose of 200 mg/ ml. And, the third treatment group was given jengkol leaf extract at a dose of 400 mg/ ml. Semi quantitative method was applied to determine C. albicans biofilmsis using Crystal Violet staining technique. The absorbance of the cells then was calculated using a spectrophotometer with a wavelength of 570 nm. Result: The mean value of optical density in the control group was 1.23. The mean value of optical density in the treatment group with a dose of 100 mg/ ml was 0.2. Meanwhile, the mean value of optical density in the treatment group with a dose of 200 mg/ ml was 0.2, and 0.21 in the treatment group with a dose of 400 mg/ ml. The results also showed that there were significant differences between the control group and all of the treatment groups (p<0.05), but there was no significant difference between the treatment groups (p>0.05). Conclusion: The optimum dose of jengkol leaf extract used as antibiofilm against C. albicans biofilms is 100 mg /ml with an inhibitory percentage of 83.7%.

Background. Enterococcus faecalis is often found in persistent endodontic infection. Its high prevalence, about 24-77%, is due to resistance and virulence of these bacteria. NaOCl is commonly use as irrigation material, but it has toxicity effect and can irritate periapical tissues. Therefore, some studies to find natural materials that have antibacterial properties as an alternative root canal medicament need to be done. Neem (Azadirachta Indica A. Juss), also known as wonder tree for centuries has antibacterial properties, proven in the preliminary study which has been done that the MIC (Minimum Inhibitory Concentration) is 65% and MBC (Minimum Bactericidal Concentration) is 70%. Purpose. The aim of this study was to compare the effectiveness of antibacterial activity of the Neem leaf extract (Azadirachta Indica A. Juss) than NaOCl 2,5% against Enterococcus faecalis. Methods. This study is an experimental labolatory with post test only control group design usingEnterococcus faecalis ATCC 29212. Agar diffusion test was used to check the antimicrobial activity of 65% Neem leaf ectract, 70% Neem leaf ectract and NaOCl 2,5% by measuring the inhibition zone diameter of each treatment.Result. Diameter of bacterial inhibition zone formed Neem leaf extract is greater than NaOCl 2,5%. Conclusion. The antibacterial activity of Neem extract is greater than NaOCl 2,5% against Enterococcus faecalis.

Indikator klinis penyakit Diabetes Melitus adalah hiperglikemia. Mimba (Azadirachta indica A. Juss) merupakan salah satu tumbuhan yang berpotensi sebagai alternatif obat herbal hiperglikemia. Penelitian ini bertujuan untuk menganalisis pengaruh ekstrak etanol daun mimba terhadap struktur histologis pankreas pada tikus hiperglikemia. Penelitian ini mengunakan Rancangan Acak Lengkap (RAL) dengan jumlah tikus 24 ekor yang dibagi menjadi 6 kelompok perlakuan dan 4 kali ulangan. P0 (kontrol normal) adalah kelompok tikus normal yang diberi akuades, P1 (kontrol negatif) adalah tikus hiperglikemia yang diberi akuades. P2 (kontrol positif) adalah kelompok tikus hiperglikemia yang diberi glibenklamid dosis 2,25 mg/kg BB. P3, P4, dan P5 adalah kelompok tikus yang diberi ekstrak etanol daun mimba dosis 100, 200, dan 400 mg/kg BB. Data dianalisis dengan ANOVA pada signifikansi 95%. Hasil penelitian menunjukkan bahwa perlakuan ekstrak etanol daun mimba dosis 100, 200, dan 400 mg/kg BB tidak memberikan pengaruh nyata terhadap bobot pankreas, diameter, luas, dan densitas pulau Langerhans (P>0,05). Skoring struktur pulau Langerhans berdasarkan uji Mann-Whitneymenunjukkan hasil beda nyata pada kelompok  tikus yang diberi daun mimba (P≤0,05). Kesimpulan dari penelitian ini, pemberian ekstrak daun mimba dosis 400mg/kg BB menunjukkan adanya perbaikan morfologi pulau Langerhans. The clinical indicator of Diabetes mellitus was hyperglycemia. Azadirachta indica A. Juss was a plant has the potential to alternative medicine for hyperglycemia. The study was to analyze the ethanol neem leaf extract effect on histological structure of hyperglycemic rat pancreas. This study used a completely randomized design (CRD) with 24 rats were divided into 6 treatment groups and 4 replications. P0 (control) was a normal rats group were given distilled water, P1 (negative control) was a hyperglycemic rats group were given distilled water. P2 (positive control) was a hyperglycemic rats were given 2.25 mg/kg BW of glibenclamide. P3, P4, and P5 were rats were given 100, 200, and 400 mg/kg BW of ethanolic neem leaf extract.The data analyzed by ANOVA at 95% significance showed the treatment of 100, 200, and 400 mg/kg BW ethanolic neem leaf extract had no significant effect on the pancreatic weight, diameter, area, and density of Langerhans islet (P> 0.05). The score of Langerhans islet structure based on the Mann-Whitney test showed significant differences in the groups of mice given neem leaves (P≤0.05). Treatment of 400mg/Kg BW neem leaf extract showed an improvement in the morphology of the islets of Langerhans.

BACKGROUND<br />Lead acetate may inhibit the enzyme aminolevulinate dehydratase (ALAD) resulting in decreased heme synthesis (and consequently in anemia) but in increased number of reticulocyte cells. IR Bagendit paddy leaf water extract has a high metallothionein protein content which acts to bind to lead. The study objective was to determine whether aqueous IR Bagendit rice leaf extract dosage variations prior to lead exposure decreases reticulocyte count in lead-exposed rats.<br /> <br />METHODS<br />The study was of randomized post test only control-group design involving a sample of 28 rats, that were randomized into 4 groups consisting of 1 control group and 3 treatment groups, daily administered with aqueous IR Bagendit rice leaf extract of respectively 0.2; 0.4; 0.8 mg using a feeding tube up to week 13. Lead exposure was also given using a feeding tube to both control and treatment groups at a dose of 0.5 g/kg BW/day, up to week 13. The reticulocyte count was then examined using supravital brilliant cresyl blue staining. The reticulocyte count was determined per 1000 erythrocytes and then converted into a percentage. Kruskal Wallis test followed with Bonferroni test was conducted to figure out the differences between groups. <br /><br />RESULTS<br />Mean reticulocyte count decreased significantly, starting from the control group up to the third treatment group (15.48 ± 3.41; 12.25 ± 03.28; 10.45 ±1.47; 9.10 ± 2.35 average per unit) (p=0.02). The Bonferroni test showed that the reticulocyte count was significantly decreased in the third treatment group (p=0.004). <br /><br />CONCLUSION<br />Aqueous rice leaf extract significantly decreases reticulocytes in rats exposed to lead.

Burn therapy involves elimination of causative factors and symptomatic therapy. Kenitu leaves (Chrysophyllum cainito L.) are known to contain β-amylin acetate, Gallic acid, ursolic acid, quercetin, β-sitosterol, and other substances that have the potential to be antibacterial, antioxidant, and anti-inflammatory. The use of kenitu leaves (Chrysophyllum cainito L.) as an alternative to wound healing has low side effects and is easy to obtain, especially in Jember Regency. The aim of this study was to examine the effect of kenitu leaves (Chrysophyllum cainito L.) leaf extract gel on re-epithelialization in buccal mucosal burns of wistar rats. This type of research is experimental laboratories. This research used 32 samples which divided into four control groups and four treatment groups. Burns are made on the left buccal mucosa. In the control group the CMC placebo gel was given and the treatment group was given 2% leaf extract gel. Euthanasia was performed on days 4, 7, 10, and 13 then histological preparations were made. Measurement using software optilab viewer 2.2 and image raster 3.0. The average yield of epithelial thickness was lowest in the control group on day 4 and highest in the treatment group on day 13. The results of the One Way Anova statistical test showed a significant difference in epithelial thickness between the control group and the treatment group. This study proved that the 2% leaf extract can accelerate the process of reepithelialization in the healing of wistar mucosal mucosal burns.

Atikaningrum DA, Ediningsih E, Utari CRS. 2013. Analgesic effectiveness comparison between red betel leaf extract (Piper crocatum) and therapy dosage of aspirin in mice. Biofarmasi 11: 1-6. The aim of this research was to find out an analgesic effectiveness of red betel leaf extract (Piper crocatum) compared to the therapy dosage of aspirin in mice. This research used a completely randomize experimental design. The subject in this research were 30 male mice of Swiss which in age of 2-3 months with 20-30 grams weight. The subjects were divided into five treatment groups: (i) negative control group (aquadest), (ii) positive control group (aspirin), (iii) first treatment group (3.64 mg red betel leaf extract), (iv) second treatment group (7.28 mg red betel leaf extract), and (v) third treatment group (14.56 mg red betel leaf extract). Analgesic effect was determined by counting the mice jump on 42oC in hotplate during 5 minutes, 2 hours after treatment. The obtained data were tested statistically by Anova and Pos-Hoc processed by Lead Significance Difference. The Anova test showed that there were significant differences among five treatment groups, while the LSD test showed that there were significant difference between negative control group and treatment groups, and the second and third treatment groups showed no significant difference with positive control group. The red betel leaf extract had an analgesic effect when it was given orally in mice. The treatment groups that had the same efficacy with the aspirin treatment group were the second (7.28 mg red betel leaf extract) and the third groups (14.56 mg red betle leaf extract).
