The use of water-soluble form of ubikhinone for leveling oxidative stress in chicken embryos as a method for optimizing the hematological status

The incubation eggs were treated with solutions of a water-soluble form of ubiquinone in order to neutralize the negative consequences of the development of oxidative stress in chick embryos under conditions of artificial incubation to optimize the hematological status in their body. The results showed that the implementation, first of all, of the antioxidant properties of the claimed metabolite made it possible to obtain the values of the central hematological indices, indicating a decrease in the body of individuals of the best experimental group of endogenous intoxication, an increase in stress resistance, adaptive capabilities and stabilization of homeostasis.

The purpose of the work was to investigate the effect of the feed supplement “Butaselmevit Plus” on the antioxidant status of the organism in rats under conditions of oxidative stress. The research was carried out on white, sexually-mature, young male rats of the Wistar line, with a body weight of 180–200 g, which was kept on a standard diet of the Institute vivarium of the State Scientific-Research Control Institute of Veterinary Preparations and Feed Supplements. The animals were divided into three groups of 20 animals in each: 1st group (K) intact animals; Group 2 (R1) – rats, affected with tetrachloromethane; Group 3 (R2) – rats, affected with tetrachloromethane and used as a feed additive “Butaselmevit-Plus”. The feed supplement “Butaselmevit-Plus” includes the fruits of thistle spotted, methionine, sodium selenite and vitamins A, E, D3. Significant violation of oxidative-antioxidant balance in animals under oxidative stress conditions, which is characterized, primarily, by activation of processes of radical lipid oxidation with excessive accumulation of both intermediate and final products of lipid peroxidation oxidation and inhibition of antioxidant defense system activity, is established. The development of oxidative stress leads to the inhibition of the activity of the glutathione system of antioxidant protection of the body of rats. This is evidenced by the low activity of glutathione peroxidase and the low level of reduced glutathione in the blood of experimental rats. The feed supplement “Butaselmeib-Plus” contributed to the activation of the system of antioxidant protection of the body of rats for tetrachloromethane poisoning, as evidenced by the increased activity of glutathione peroxidase and the level of reduced glutathione. In addition, in the blood of experimental rats, inhibition of lipid peroxidation and formation of free radicals was observed. It was determined that the level of lipids hydroperoxides on the 20th day of the experiment in blood of rats of experimental group R2 decreased by 35.7%, and the level of TBK-active products – by 21.6% relative to the indices of the first experimental group of rats. Thus, the feed supplement “Butaselmevit-Plus” when fed to rats for the development of oxidative stress inhibited lipid peroxidation, as indicated by the low level of lipids hydroperoxides and TBK-active products in their blood. This may be due to the fact that the feed additive includes two strong antioxidants, such as vitamin E and selenium, which in turn enhance the action of each other. It should also be noted that the antioxidant properties of thistle blisters, which, according to the literature, also possess antioxidant properties. It consists of vitamins B, A, E, K, precursors of vitamin D, carotenoids, macroelements – potassium, calcium, magnesium, ferrum and microelements – Cuprum, Zinc, Manganese, Iodine. The combined action of these biologically important elements exhibits high hepatoprotective and antioxidant activity.

AbstractOxidative stress, associated with an imbalance between the oxidants (reactive oxygen species) and the antioxidants in the body, contributes to the development of many diseases. The body’s fight against reactive oxygen species is supported by antioxidants. Nowadays, there are too many analytical methods, but there is no one universal technique for assessing antioxidant properties. Moreover, the applied different ways of expressing the results lead to their incompatibility and unreasonable interpretation. The paper is a literature review concerning the most frequent ways of antioxidant activities expression and for an easy and universal method of the obtained results discussion. This paper is an attempt to point out their disadvantages and advantages. The manuscript can support the searching interpretation of the obtained results which will be a good tool for the development of a number of fields, especially medicine what can help in the future detection and treatment of many serious diseases. Oxidative stress is a result of disruption in the balance between antioxidants and pro-oxidants in which subsequently impacting on redox signaling, causing cell and tissue damages. It leads to a range of medical conditions including inflammation, skin aging, impaired wound healing, chronic diseases and cancers but these conditions can be managed properly with the aid of antioxidants. This review features various studies to provide an overview on how Carica papaya help counteract oxidative stress via various mechanisms of action closely related to its antioxidant properties and eventually improving the management of various oxidative stress-related health conditions. Carica papaya is a topical plant species discovered to contain high amounts of natural antioxidants that can usually be found in their leaves, fruits and seeds. It contains various chemical compounds demonstrate significant antioxidant properties including caffeic acid, myricetin, rutin, quercetin, α-tocopherol, papain, benzyl isothiocyanate (BiTC), and kaempferol. Therefore, it can counteract pro-oxidants via a number of signaling pathways that either promote the expression of antioxidant enzymes or reduce ROS production. These signaling pathways activate the antioxidant defense mechanisms that protect the body against both intrinsic and extrinsic oxidative stress. To conclude, Carica papaya can be incorporated into medications or supplements to help manage the health conditions driven by oxidative stress and further studies are needed to investigate the potential of its chemical components to manage various chronic diseases.

The basis of the pathogenesis of male infertility is the processes of peroxide oxidation of biological substrates, especially lipids and proteins. By destroying the sperm membrane, toxic peroxidation products reduce its motility and ability to fertilize the egg, which is determined by a decrease in the number of motile sperm in the ejaculate. These changes lead to complete or partial male infertility. The authors of the review found that is accompanied by a damaging effect on the structural and functional activity of the gonads and is manifested, in particular, by an imbalance in the hormonal background of the male body. Similar effects are characteristic of an increase in the content of reactive Nitrogen species and its metabolites, which cause nitrosative stress, which is also the cause of male hypofertility and is inseparable from the state of oxidative stress. In scientific work it is determined that the accumulation of harmful peroxidation products leads to damage and destruction of sperm DNA, reduced activity of acrosomal enzymes and mitochondrial potential of sperm, reduced overall antioxidant activity. This makes it impossible for an adequate response of the body. Multi component antioxidant defense system resists stress. It is represented by enzymatic and non-enzymatic links, which can neutralize harmful radicals and peroxidation products. It contributes to the full manifestation of reproductive function. The presence of powerful antioxidant properties of catalase, superoxide dismutase, and enzymes of the thiol-disulfide system, which form the enzymatic system of antioxidant protection, as well as selenium, zinc, copper, other trace elements, retinol, tocopherol, ascorbic acid, and vitamins as parts of the non-enzymatic system is shown. The efficiency of registration is substantiated thin biochemical shift detectors or complex methods, such as total antioxidant status of sperm or sperm plasma, mitochondrial membrane potential, etc along with simple markers of oxidative stress, such as diene conjugates, malonic dialdehyde, and metabolites of the Nitrogen Oxide cycle. Given the leading role of oxidative stress in the development of male hypofertility, the prospect of further research is the search for modern means for correction, especially among substances with pronounced redox activity

The results of research on the influence of the elaborated complex immunotropic drug containing butafosfan, interferon, thistle and fat-soluble vitamins A, D3, E in the form of a liposomal emulsion on the activity of T- and B-cell chains of immunity in rats under the conditions of action on the body of oxidative stress are presented. It has been established that the introduction of 50% tetrachloromethane into the rats of the first and second experimental groups, with a dose of 0.25 ml per 100 g of body weight, causes oxidative stress in them which negatively affects the cellular immunity and functional activity of immunocompetent blood cells. Immunosuppressive effects of oxidative stress were manifested by a decrease in the blood of rats in the first and second experimental groups of the number of T- and B-lymphocytes and their regulatory subpopulations mainly on the 2nd and 5th day of the study. At the same time, in the blood of rats of the first experimental group in all research periods a decrease in the relative number of common, active and theophylline-resistant T-lymphocytes, as well as B-lymphocytes was observed with a noticeable increase in the number of their undifferentiated forms. At the same time, the obtained data suggest the positive effect of butafosfan, interferon, thistle and vitamins A, D3, E in the liposomal preparation on the relative amount of T- and B-lymphocytes and on the redistribution of avidity in the direction of strengthening the receptor field of plasma membranes immunocompetent cells. It was found that the normalization on the 2nd day of blood level in the second experimental group of common T-lymphocytes occurred due to the secondary forms of the blood and active T-lymphocytes by changes in the number of low-avid forms. In addition to the indicators characterizing the cellular immunity of rats, the components of the liposomal preparation showed regulatory influence on the humoral link of the immune response. In particular, on the 10th day of research in blood of rats of the second experimental group a tendency towards an increase in the relative number of B-lymphocytes and an increase in the number of cells with low and medium density of receptors was found, which, under the conditions of oxidative stress, indicates an increase in the body's ability to actively synthesize protective antibodies

Для получения препаратов и добавок, обладающих антиоксидантными свойствами, представляют интерес выжимки томатов, образующиеся при производстве сока прямого отжима с предварительным отделением семян. Проведена сравнительная оценка эффективности функциональных свойств образцов пищевой добавки, полученных из выжимок томатов по различным технологиям: образец добавки 1 получен по технологии, включающей ИК-сушку выжимок, их охлаждение и измельчение; образец добавки 2 – по инновационной технологии, включающей дополнительную предварительную обработку выжимок в ЭМП СВЧ перед ИК-сушкой. Исследования проводили в течение 30 дней на трех группах лабораторных белых крыс: контрольной, животные которой получали стандартный рацион, и двух опытных, в которых животные помимо основного рациона получали при кормлении индивидуально 1 раз в сутки 2 г добавки образца 1 (1-я опытная группа) или образца 2 (2-я опытная группа). Установлено, что степень снижения концентрации ферментов печени АсАт и АлАт в крови животных 2-й опытной группы соответственно на 10,6 и 11,0% выше, чем в крови животных 1-й опытной группы. Степень снижения содержания диеновых коньюгатов, кетодиенов и малонового диальдегида в крови животных 2-й опытной группы была соответственно на 22,1; 10,1 и 12,7% выше по сравнению с аналогичными показателями животных 1-й опытной группы. Таким образом, доказано, что пищевая добавка «Томатная», выработанная по инновационной технологии, проявляет ярко выраженные антиоксидантные и гепатопротекторные свойства, что обеспечивает нормализацию биохимических процессов в организме, повышает его антиоксидантную защиту и препятствует разрушению мембран клеток. For the obtaining of preparations and additives with antioxidant properties, tomato pomace, formed during the production of direct-pressed juice with preliminary separation of seeds, is of interest. A comparative evaluation of the effectiveness of the functional properties of food additive samples obtained from tomato pomace using various technologies was carried out. The sample of additive 1 was obtained using a technology that includes IR drying of pomace, cooling and grinding; the sample of additive 2 was obtained using a technology that includes additional pretreatment of pomace in microwave EMF before IR drying. The studies were carried out for 30 days on three groups of laboratory white rats: a control group, the animals of which were given a standard diet, and two experimental ones, in which the animals, in addition to the main diet, were given individually once a day with 2 g of an additive of sample 1 (1st experimental group) or sample 2 (2nd experimental group). It was found that the degree of decrease in the concentration of liver enzymes AsAt and AlAt in the blood of animals of the 2nd experimental group was respectively 10,6 and 11,0% higher, than in the blood of animals of the 1st experimental group. The degree of decrease in the content of diene conjugates, ketodienes and malondialdehyde in the blood of animals of the 2nd experimental group was higher respectively by 22,1; 10,1 and 12,7% compared with similar indicators of animals of the 1st experimental group. Thus, it is proved that the food additive «Tomatnaya», developed by innovative technology, shows pronounced antioxidant and hepatoprotective properties, which ensures the normalization of biochemical processes in the body, increases its antioxidant protection and prevents the destruction of cell membranes.

Purpose This study aims to analyze the antioxidant properties of the Mediterranean diet and describe methods that are used in clinical studies to assess its role in reducing oxidative stress. Design/methodology/approach The review presents the results of interventional and observational clinical trials aimed at assessing the influence of the Mediterranean diet on the level of enzymatic and non-enzymatic antioxidants, as well as the total blood antioxidant capacity. Findings The Mediterranean diet as a varied diet can be a better way to provide antioxidants to the body than supplements. Individual compounds administered in an isolated form can give the opposite effect to the expected, stimulating oxidative stress. The administration of antioxidants in the form of supplements instead of a varied diet is also associated with a lack of synergism of action. In studies on the importance of the Mediterranean diet in the reduction of oxidative stress, single markers are used to measure oxidative damage, the activity of enzymatic antioxidants and the concentration of individual non-enzymatic antioxidants. At the same time, the need to find markers that would assess the level of oxidative stress and the body’s antioxidant capacity more comprehensively is emphasized. Practical implications It should be taken into account that differences between in vivo and in vitro results may result from the fact of various factors, including genetic, smoking, intestinal microflora or diet composition. It is also necessary to answer the question about which marker or set of markers could in the most comprehensive way to assess the level of oxidative stress and the body’s antioxidant capacity. Originality/value The literature review shows not only the source of antioxidants in the Mediterranean diet. This paper also presents a critical approach to markers that allow the assessment of the antioxidant properties of the diet.

Oxidative stress is a major issue in a wide number of pathologies (neurodegenerative, cardiovascular, immune diseases, and cancer). Because of this, the search for new antioxidants is an important issue. One of the potential antioxidants that has been enthusiastically discussed in the past twenty years is fullerene and its derivatives. Although in aqueous solutions fullerene derivatives have shown to be antioxidants, their properties in this regard within the cells are controversially discussed. We have studied two different water-soluble fullerene C60 and C70 derivatives on human embryonic lung fibroblasts at a wide range of concentrations. Both of them cause a decrease in cellular ROS at short times of incubation (1 hour). Their prolonged action, however, is fundamentally different: derivative GI-761 causes secondary oxidative stress whereas derivative VI-419-P3K keeps ROS levels under control values. To gain a better understanding of this effect, we assessed factors that could play a role in the response of cells to fullerene derivatives. Increased ROS production occurred due to NOX4 upregulation by GI-761. Derivative VI-419-P3K activated the transcription of antioxidant master regulator NRF2 and caused its translocation to the nucleus. This data suggests that the antioxidant effect of fullerene derivatives depends on their chemical structure.

ABSTRACTThe water-soluble polyhydroxyfullerene (PHF) is a functionalized carbon nanomaterial with several industrial and commercial applications. There have been controversial reports on the toxicity and/or antioxidant properties of fullerenes and their derivatives. Conversely, metals have been recognized as toxic mainly due to their ability to induce oxidative stress in living organisms. We investigated the interactive effects of PHF and cadmium ions (Cd) on the model yeastSaccharomyces cerevisiaeby exposing cells to Cd (≤5 mg liter−1) in the absence or presence of PHF (≤500 mg liter−1) at different pHs (5.8 to 6.8). In the absence of Cd, PHF stimulated yeast growth up to 10.4%. Cd inhibited growth up to 79.7%, induced intracellular accumulation of reactive oxygen species (ROS), and promoted plasma membrane disruption in a dose- and pH-dependent manner. The negative effects of Cd on growth were attenuated by the presence of PHF, and maximum growth recovery (53.8%) was obtained at the highest PHF concentration and pH. The coexposure to Cd and PHF decreased ROS accumulation up to 36.7% and membrane disruption up to 30.7% in a dose- and pH-dependent manner. Two mechanisms helped to explain the role of PHF in alleviating Cd toxicity to yeasts: PHF decreased Cd-induced oxidative stress and bound significant amounts of Cd in the extracellular medium, reducing its bioavailability to the cells.
