 Objective: Green synthesis of nanoparticles has been used as an alternative, efficient, less expensive, and ecofriendly method. Ancient approaches employed for nanoparticle fabrication were chemical and physical methods having various disadvantages as they are costly and potentially harmful to the environment, use of harsh chemicals and stringent protocol for synthesizing nanoparticles. The present study is focused on the synthesis of silver nanoparticles by bio-availed as well as chemical route and evaluation of their wound healing potential in Wistar rat model.Methods: Extraction of Syzygium cumini was done and was used for silver nanoparticle synthesis. The synthesized nanoparticles were characterized by ultraviolet-visible spectroscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, and transmission electron microscopy, which confirms the availability of nanosilver particles with marginally spherical morphology. The particles were then carried forward for treating impaired diabetic wounds in Wistar rat models. Regular photography was done and wound healing parameters were monitored throughout the study along with analyzing other parameters such as biochemical, hematological, and histological parameters.Results: The study showed that the sizes of the synthesized nanoparticles are below 100 nm. The results obtained from in vivo studies showed efficient wound healing potential of silver nanoparticles as compared to pre-existing drug povidone-iodine, i.e., the percentage reduction in wound area after therapy is 96.09% in case of biosynthesized silver nanoparticle-treated group, 97.7% reduction in chemically synthesized silver nanoparticle-treated group, 64.28% reduction in case of pre-existing drug povidone-iodine-treated group, 37.5% reduction in case of diabetic control group (diabetic), and 97.5% reduction in normal control group (non-diabetic). Results showed that biosynthesized silver nanoparticles showed less toxicity with respect to liver and kidney functions. Skin histology results showed increased sign of wound healing in biosynthesized silver nanoparticles. Hematology results showed no such variation.Conclusion: The study will help to synthesize new economically viable potential biosynthesized nanoparticles along with providing the approach to develop the medication at nanoscale level.

This research was considered to evaluate the antidiabetic effect of silver nanoparticle (AgNps)and following experimental diabetic. In the present study. Thirty healthy swiss mice aged between 7-8 weeks, old male mice and divided into six groups of five animals. Diabetic induced mice by using intraperitoneal (IP) injection of alloxan (180 mg lkg). Group 1 included non diabetic control, Group 2 Diabetic, Group 3 Diabetic +0.01 mg AgNps Group 4 Diabetic +0.05 AgNps, Group 5 Diabetic+ wound contaminated with Pseudomonas aeruginosa. Group 6 ( diabetic +contaminated wound + silver nanoparticle (Ag Nps). Silver nanoparticle show ample antibacterial activities. The result of the current study introduced an invivo silvernanoparticle accelerate by effects on the treatment of Pseudomonas aeruginosa infected skin wound. The present study was conducted to synthesis the AgNps biologically and evaluate its antibacterial activity against Pseudomonas aeruginosa diabetes induced by Alloxan in mice. Adminstration of silver nanoparticle resulted in significance antidiabetic effects that is is improved glucose tolerance higher source. The current study results are presented for the first time which suggest for the development of AgNps as anantdiabetic factor in future. The broad spectrum of bioactivity of AgNPs makes them promising agent not only to fight infection, but to sterile the wound and accelerate wound healing. There were significant higher wound healing scores in Nanoparticle treated group. Compared with control group. These result suggest that nanoparticle may be useful in diabetic wound healing. Treatment with asingle dose of AGNPs produced amild reduction in blood glucose and some reduction in plasma insulin at 2 h.The present results revealed the potential of the synthesized Ag-NPs as safer bactericidal agents for the treatment of diabetes induced wound contaminated with P.aeruginosa.

Objective: Achillea fragrantissima L. (Asteraceae) is a traditionally used medicinal herb in the rural communities of Jordan. Methods: The present study evaluated the efficacy of the ethanol extract of this species on angiogenesis in both, ex vivo using rat aortic ring assay and in vivo using rat excision wound model. Results: In concentrations of 50 and 100 µg/ml, the ethanol extract showed angiogenic stimulatory effect and significantly increased length of capillary protrusions around aorta rings of about 60% in comparison to those of untreated aorta rings. In MCF-7 cells, the ethanol extract of A. fragrantissima stimulates the production of VEGF in a dose-dependent manner. 1% and 5% of ethanol extract of A. fragrantissima containing vaseline based ointment was applied on rat excision wounds for six days and was found to be effective in wound healing and maturation of the scar. Both preparations resulted in better wound healing when compared to the untreated control group and vaseline-treated group. This effect was comparable to that induced by MEBO, the positive control. Conclusion: The results indicate that A. fragrantissima has a pro-angiogenic effect, which may act through the VEGF signaling pathway.

Cold plasma can be beneficial for promoting skin wound healing and has a high potential of being effectively used in treating various wounds. Our aim was to verify the effect of cold plasma in accelerating wound healing and investigate its underlying mechanism in vitro and in vivo. For the in vivo experiments, 2 full-thickness dermal wounds were created in each mouse (n = 30). While one wound was exposed to 2 daily plasma treatments for 3 min, the other wound served as a control. The wounds were evaluated by imaging and histological analyses at 4, 7, and 11 days post the wound infliction process. Immunohistochemical studies were also performed at the same time points. In vitro proliferation and scratch assay using HaCaT keratinocytes and fibroblasts were performed. The expression levels of wound healing–related genes were analyzed by real-time polymerase chain reaction and western blot analysis. On day 7, the wound healing rates were 53.94% and 63.58% for the control group and the plasma-treated group, respectively. On day 11, these rates were 76.05% and 93.44% for the control and plasma-treated groups, respectively, and the difference between them was significant ( P = .039). Histological analysis demonstrated that plasma treatment promotes the formation of epidermal keratin and granular layers. Immunohistochemical studies also revealed that collagen 1, collagen 3, and alpha-smooth muscle actin appeared more abundantly in the plasma-treated group than in the control group. In vitro, the proliferation of keratinocytes was promoted by plasma exposure. Scratch assay showed that fibroblast exposure to plasma increased their migration. The expression levels of collagen 1, collagen 3, and alpha-smooth muscle actin were elevated upon plasma treatment. In conclusion, cold plasma can accelerate skin wound healing and is well tolerated.

Objective. The study was designed to evaluate pharmacological potential of hydroalcoholic leaves extract of Chrozophora tinctoria intended for wound healing in diabetic rats’ model. Methods. The method used to evaluate the pharmacological potential of hydroalcoholic leave extract was physical incision rat model. In this model, cutting of the skin and/or other tissues with a sharp blade has been made and the rapid disruption of tissue integrity with minimal collateral damage was observed shortly. Animals used in the study were divided into four groups that consist of six animals in each group. Group I serves as normal control, Group II serves as disease control, Group III was used as standard treatment (Povidone iodine 50 mg/kg b.w.), and Group IV was used for test drug (C. tinctoria 50 mg/kg b.w.). Result. The hydroalcoholic leave extract of Chrozophora tinctoria has been significantly observed to heal the wound (98%) in diabetic rats within 21 days, while standard drug (Povidone iodine) healed the wound about 95% in the same condition. The oral dose (50 mg/kg b.w.) of Chrozophora tinctoria was also found to improve the elevated blood glucose level in comparison to disease control group, which increased after the oral administration of Streptozotocin. Conclusion. The Chrozophora tinctoria has significant wound healing potential in the animal having physically damaged tissue in diabetic condition.

Bone loss, either by trauma or other diseases, generates an increasing need for substitutes of this tissue. This study evaluated Bioglass as a bone substitute in the regeneration of the alveolar bone in mandibles of dogs by clinical, surgical and radiological analysis. Twenty-eight adult dogs were randomly separated into two equal groups. In each animal, a bone defect was created on the vestibular surface of the alveolar bone between the roots of the fourth right premolar tooth. In the treated group, the defect was immediately filled with bioglass, while in the control, it remained unfilled. Clinical evaluations were performed daily for a week, as well as x-rays immediately after surgery and at 8, 14, 21, 42, 60, 90 and 120 days post-operative. Most animals in both groups showed no signs of inflammation and wound healing was similar. Radiographic examination revealed a gradual increase of radiopacity in the region of the defect in the control group. In the treated group, initial radiopacity was higher than that of adjacent bone, decreasing until 21 days after surgery. Then it gradually increased until 120 days after surgery, when the defect became undetectable. The results showed that Bioglass integrates into bone tissue, is biocompatible and reduced the period for complete bone regeneration.

Introduction: Research on the use of papaya and aloevera sap on wound healing each been done, but effects the combination of the two extracts on wound healing in diabetes mellitus condition has not been done.Purpose: This study aims to determine the effect of a gel combination of papaya latex and aloevera flesh extracts on the traumatics ulcer healing process in diabetic rats.Methods: True experimental laboratory research with the post-test-only control group design that performed on 28 male wistar rats which were divided intofour groups: DM and non-DM rat that treatment with povidone iodine and gel combination of papaya latex and aloevera flesh extract. The treatment was done for 7 days with smearing that was given 2x/day. Traumatics ulcer healing seen from the number of fibroblasts that then were analyzed by One WayAnova and post hoc LSD test.Results: The results showed the number of fibroblasts in DM and non-DM rat with povidone iodine were 44.6 and 42.73; in the DM and non-DM rat with a gel combination of papaya latex and aloe vera flesh extracts were 61.10 and 77.03. One Way Anova test resulted p-value = 0.000, it was mean that there were at least a significant difference on the number of fibroblasts in the two groups. Differences of the number fibroblasts was shown in all groups, except in the group DM and non-DM rats with povidone iodine (p = 0.764).Conclusion: Conclusion of the study was the provision of a gel combination of papaya latex and aloevera flesh extracts effect on the traumatics ulcer healing process in diabetic rats.

Purpose. To explore the regulatory effects of liraglutide on the kidney and liver through the miR-34a/SIRT1 pathway with related factors in diabetic nephropathy (DN) rats. Methods. DN rats were randomly divided into two groups ( n = 10 ) and were injected with liraglutide or normal saline twice a day. The 24-hour urine microalbumin content and biochemical index levels were measured. qRT-PCR was performed to detect the expression of miR-34a in the kidney and liver tissues. The levels of SIRT1, HIF-1a, Egr-1, and TGF-β1 in kidney and liver tissues were determined using qRT-PCR, western blot, and immunohistochemistry. Electron microscopy and HE staining were used to observe the ultrastructure and pathological changes. Results. Liraglutide treatment in DN rats decreased blood glucose, 24-hour urine microalbumin, TC, TG, LDL-C, UA, Cr, UREA, ALT, and AST levels and increased the level of HDL-C ( P < 0.05 ). Compared with the control group, the miR-34a levels were significantly decreased in kidney and liver tissues followed by liraglutide treatment ( P < 0.05 ). The levels of SIRT1 in the liraglutide group are significantly higher than those in the control group with the kidney and liver tissues ( P < 0.05 ). Conversely, the contents of HIF-1a, Egr-1, and TGF-β1 were significantly lower in the liraglutide group than in the control group ( P < 0.05 ). Electron microscopy showed that the kidney of the liraglutide-treated group exhibited minor broadening of the mesangial areas, fewer deposits, and a well-organized foot process. HE staining revealed that the kidney of the liraglutide-treated rats had a more regular morphology of the glomerulus and Bowman sac cavity and lighter tubular edema. Additionally, the liraglutide-treated DN rats had a clear hepatic structure, a lower degree of steatosis, and mild inflammatory cell infiltration. Conclusion. Liraglutide, through its effect on the miR-34a/SIRT1 pathway, may have a protective role in the kidney and liver of DN rats.

In this study, we aimed to investigate the antidiabetic effects of Euphorbia nivulia (En), native to Cholistan Desert area of Bahawalpur, Pakistan. First, we performed high-performance liquid chromatography analysis and found that this plant contains ferulic acid, gallic acid, quercetin, benzoic acid, polyphenols, and flavonoids. Then, we performed in vitro and in vivo studies to assess its effects on diabetic Wistar rat model. The experiments were performed and compared with control drug glibenclamide. The 70% hydroalcoholic extract of En exhibited 97.8% in vitro α-glucosidase inhibitory effect at a dose of 1.0 mg/mL. We orally administered the extract of En and control drug to the streptozotocin (STZ)-induced diabetic rats and analyzed its antidiabetic effects. We found that the extract of En with a dose of 500 mg/kg/body weight exhibited significant effect to reduce blood glucose in STZ-induced rats as compared with the control group ( P < .001). Our histological data also showed that the extract significantly improved the histopathology of pancreas. Collectively, both in vitro and in vivo studies revealed that En possesses α-glucosidase inhibitory, antioxidant, and anti-hyperglycemic effect in STZ-induced diabetic rats.

Acupuncture regulates inflammation process and growth factors by increasing blood circulation in affected areas. In this study, we examined whether acupuncture has an effect on wound healing in injured rat. Rats were assigned randomly into two groups: control group and acupuncture group. Acupuncture treatment was carried out at 8 sites around the wounded area. We analyzed the wound area, inflammatory cytokines, proliferation of resident cells, and angiogenesis and induction of extracelluar matrix remodeling. At 7 days after-wounding the wound size in acupuncture-treat group was decreased more significantly compared to control group. In addition, the protein levels of proinflammatory cytokines such as tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) were significantly decreased compared to the control at 2 and 7 days post-wounding. Also, we analyzed newly generated cells by performing immunostaining for PCNA and using several phenotype markers such as CD-31,α-SMA, and collagen type I. In acupuncture-treated group, PCNA-positive cell was increased and PCNA labeled CD-31-positive vessels,α-SMA- and collagen type I-positive fibroblastic cells, were increased compared to the control group at 7 days post-wounding. These results suggest that acupuncture may improve wound healing through decreasing pro-inflammatory response, increasing cell proliferation and angiogenesis, and inducing extracellular matrix remodeling.

BACKGROUND: Acalypha indica Linn. has been used as traditional medicine, it contains flavonoids, alkaloids, tannins, saponins, steroids, triterpenoids, and essential oils. AIM: This study aimed to determine the bio-larvicide effects of A. indica Linn. leaf stew and the silver nanoparticles against Anopheles sp. larvae. METHODS: The fresh leaves of A. indica Linn. extracted using distilled water at 100°C for 30 min. The silver nanoparticles were made by mixing a solution of silver nitrate with the stew, which acts as a reducing agent. The resulting silver nanoparticles were characterized by particle size analyzer and UV-vis spectrophotometer. The bio-larvicide effects against Anopheles sp. larvae performed using a completely randomized design. There were eight groups consisted of ten larvae and three replications. Treatment groups of stew and silver nanoparticle for concentrations 0.05%, 0.5%, and 5%, respectively. The negative control group was distilled water and the positive control group was the 0.01% abate solution. Assessment of larvicide activity was carried out every hour for 6 h and continued if there were larvae that live up to 24 h. The LC50 value was calculated based on Probit analysis. RESULTS: The results showed that the A. indica Linn. leaf stew can be made into silver nanoparticles preparations, optimal results were obtained from a mixture of 1% stew and 3 mM AgNO3. The result of bio-larvicides effect test against Anopheles sp. larvae showed that the LC50 value of the A. indica Linn. leaf stew was 727,3 ppm and the LC50 value of silver nanoparticles was 3.366 ppm. CONCLUSION: It can be concluded that A. indica Linn. is a promising larvicidal plant and can be made into silver nanoparticle preparations.
