The aim of the study was to define the chemical characteristics, antioxidant capacity, oxidative status, sensory properties, and the presence of polyphenols in ovine cheese obtained after dietary administration of spray-dried olive mill wastewater (SDP). SDP is a waste from olive oil production rich in bioactive molecules obtained by further processing the olive mill wastewater through a spray-drying system. Thirty-six sheep were randomly assigned to two experimental groups that received a standard diet based on hay and concentrate. The concentrate fed to the SDP group was supplemented with SDP at a rate 25 g/kg (as fed). The trial lasted 9 weeks. Milk from the two treatment groups was separately collected and used for manufacturing cheese. Cheese quality parameters and proximate composition were not affected by the dietary treatment, whereas the antioxidant status and oxidative stability of cheese were positively affected. Polyphenol analyses in cheese were performed through liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). The concentration of hydroxytyrosol and tyrosol, and their sulphate metabolites, were higher in cheese from supplemented sheep. These findings suggest that polyphenol metabolites can play a major role in the beneficial effects observed in food produced from sheep fed SDP.

Bladder cancer patients whose tumors develop resistance to cisplatin-based chemotherapy often turn to natural, plant-derived products. Beneficial effects have been particularly ascribed to polyphenols, although their therapeutic relevance when resistance has developed is not clear. The present study evaluated the anti-tumor potential of polyphenol-rich olive mill wastewater (OMWW) on chemo-sensitive and cisplatin- and gemcitabine-resistant T24, RT112, and TCCSUP bladder cancer cells in vitro. The cells were treated with different dilutions of OMWW, and tumor growth and clone formation were evaluated. Possible mechanisms of action were investigated by evaluating cell cycle phases and cell cycle-regulating proteins. OMWW profoundly inhibited the growth and proliferation of chemo-sensitive as well as gemcitabine- and cisplatin-resistant bladder cancer cells. Depending on the cell line and on gemcitabine- or cisplatin-resistance, OMWW induced cell cycle arrest at different phases. These differing phase arrests were accompanied by differing alterations in the CDK-cyclin axis. Considerable suppression of the Akt-mTOR pathway by OMWW was observed in all three cell lines. Since OMWW blocks the cell cycle through the manipulation of the cyclin-CDK axis and the deactivation of Akt-mTOR signaling, OMWW could become relevant in supporting bladder cancer therapy.

The present study was undertaken to evaluate the efficacy of aspirin as anti-heat stressor. Broiler chicks were reared on a standard diet up to 14 days of age. Thereafter, the chicks were randomly distributed into three dietary treatment groups viz., T1 (Control group: Standard diet), T2 (Standard diet with aspirin@250 mg/kg) and T3 (Standard diet with aspirin@500 mg/kg). Each treatment was having five replicates of eight birds per replicate. Experiment was carried out during hot-dry (April-May, 30.0±0.70 to 37.0±1.40C, Rh, %: 58.05±1.32 to 70.11±0.82) summer. Feed intake and live weight gain was comparable among the treatments but FCR, protein and energy efficiency improved significantly (P less than 0.001) on aspirin supplementation. The relative yield of thymus and bursa at 4th and 6th week of age improved significantly (P less than 0.05). The percentage of protein, aspartate transaminase (AST) and alanine transaminase (ALT) increased significantly (P less than 0.001) due to aspirin supplementation at 4th as well as 6th week of age. While H:L ratio, serum corticosteron and serum cholesterol (only at 42nd day of age) decreased significantly (P less than 0.001) in aspirin supplemented groups at 28th as well as 42nd day of age. Supplementation of aspirin at 500 mg/kg reduced relative expression of HSP70 in jejunum tissues during 28th or 42nd day of age. It can be concluded that aspirin supplementation 500 mg/kg in diet of heat stressed broilers improved performance as well as welfare..

Olive mill wastewater (OMW) is the aqueous waste derived from the production of virgin olive oil. OMW typically contains a wide range of phenol-type molecules, which are natural antioxidants and/or antibacterials. In order to exploit the bioactive molecules and simultaneously decrease the environmental impact of such a food waste stream, OMW has been intercalated into the host structure of ZnAl layered double hydroxide (LDH) and employed as an integrative filler for the preparation of poly(butylene succinate) (PBS) composites by in situ polymerization. From the view point of the polymer continuous phase as well as from the side of the hybrid filler, an investigation was performed in terms of molecular and morphological characteristics by gel permeation chromatography (GPC) and X-ray diffraction (XRD); also, the thermal and mechanical properties were evaluated by thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and dynamic thermomechanical analysis (DMTA). Antibacterial properties have been assessed against a Gram-positive and a Gram-negative bacterium, Staphylococcus aureus and Escherichia coli, respectively, as representatives of potential agents of foodborne illnesses.

Thromboxane A2 and other eicosanoids such as isoprostanes contribute to vascular proliferation and atherosclerosis by binding to the thromboxane/prostaglandin endoperoxide receptors. The effects of terutroban, a thromboxane/prostaglandin endoperoxide receptor antagonist, on aorta remodeling were evaluated in spontaneously hypertensive stroke-prone rats (SHRSPs), a model of severe hypertension, endothelial dysfunction, vascular inflammation, and cerebrovascular diseases. Male SHRSPs were allocated to three groups receiving a standard diet ( n = 5) or a high-sodium permissive diet plus vehicle ( n = 6) or plus terutroban (30 mg·kg−1·day−1; n = 6). After 6 wk of dietary treatment, all of the animals were injected with bromodeoxyuridine and simultaneously euthanized for aorta collection. The aortic media thickness-to-lumen ratio significantly ( P < 0.0001) increased in the salt-loaded rats compared with the rats fed a standard diet, whereas terutroban treatment completely prevented media thickening ( P < 0.001). When compared with vehicle, terutroban was also effective in preventing cell proliferation in the media, as indicated by the reduced number of bromodeoxyuridine-positive ( P < 0.0001) and proliferating cell nuclear antigen-positive cells ( P < 0.0001). Severe fibrosis characterized by a significant accumulation of collagen and fibronectin in the vascular wall was observed in the vehicle-treated rats ( P < 0.01) but was completely prevented by terutroban ( P < 0.001). The latter also inhibited heat shock protein-47 ( P < 0.01) and TGF-1β expression ( P < 0.001), which were significantly increased by the high-salt diet. In conclusion, terutroban prevents the development of aorta hyperplasia and has beneficial effects on fibrotic processes by affecting TGF-β and heat shock protein-47 expression in SHRSPs. These findings provide mechanistic data supporting the beneficial effects of terutroban in preventing or retarding atherogenesis.

Olive oil production using three-phase decanter systems creates olive oil and two by-products: olive mill wastewater (OMWW) and pomace. These by-products contain the highest share of polyphenolic compounds that are known to be associated with beneficial effects on human health. Therefore, they are an attractive source of phenolic compounds for further industrial use in the cosmetic, pharmaceutical and food industries. The use of these phenolics is limited due to difficulties in recovery, high reactivity, complexity of the OMWW matrix and different physiochemical properties of phenolic compounds. This research, focused on OMWW, was performed in two phases. First, different polyphenol extraction methods were compared to obtain the method that yields the highest polyphenol concentration. Twenty-five phenolic compounds and their isomers were determined. Acidifying OMWW, followed by five minutes of ultrasonication, resulted in the highest measured polyphenol content of 27 mg/L. Second, the collection of polyphenolic compounds from OMWW via adsorption on unmodified iron (II, III) oxide particles was investigated. Although low yields were obtained for removed polyphenolic compounds in one removal cycle, the process has a high capability to be repeated.

An experiment was conducted on broiler chicks (n=120), reared in cages on a standard diet up to 14th day of age and thereafter up to 42nd day (14-42d) on test diets with or without MOS. The 14th day old chicks were randomly distributed into three dietary treatment groups viz., T1 (control diet), T2 (control diet with MOS @ 0.3%) and T3 (control diet with MOS @ 0.5%). Each dietary treatment comprised of four replicates of eight birds each. Experiment was carried out during hot-dry summer. It was found that production indices improved (P>0.001) significantly during all phases. Cellular and humoral immunity at 28th day of age improved significantly (P>0.05) due to MOS supplementation. The percentage of hemoglobin, protein, AST and ALT increased significantly (P>0.001) in MOS supplemented group at 28th as well as 42nd day of age. While H:L ratio, serum corticosterone and cholesterol levels decreased significantly (P>0.001) in MOS supplemented groups. Supplementation of MOS at the both levels caused significant (P>0.001) down regulation of HSP70 expression in jejunum tissues during 28th and 42nd day. Based on this study it was concluded that supplementation of 0.3% MOS in broiler diets significantly improved their performance and welfare during heat stressed conditions.

Olive mill wastewater is an important agro-industrial waste with no established treatment method. The authors have developed a phenol separation method that could potentially cover the treatment cost of the waste. The purpose of this study was to identify any economic hotspots in the process, the operational cost and examine the margin of profit for such a process. The equipment cost was scaled for different treatment capacities and then used to estimate the fixed capital investment and the yearly operational cost. The highest purchased equipment cost was identified for the membrane filtration system, while the cost for resin replacement was identified as the highest operational cost. The lifespan of the resin used in the adsorption step was identified as an economic hot spot for the process, with the phenols separation cost ranging from 0.84 to 13.6 €/g of phenols for a resin lifespan of 5–100 adsorption/desorption cycles. The lifespan of the resin proved to be the single most important aspect that determines the phenols separation cost. The price range that was calculated for the product of the process is very promising because of the typical value of antioxidants and the low concentration of phenols that are needed for food supplements and cosmetics.

This work aims to analyze the chemical and biological evaluation of two extracts obtained by olive mill wastewater (OMW), an olive oil processing byproduct. The exploitation of OMW is becoming an important aspect of development of the sustainable olive oil industry. Here we chemically and biologically evaluated one liquid (L) and one solid (S) extract obtained by liquid–liquid extraction followed by acidic hydrolysis (LLAC). Chemical characterization of the two extracts indicated that S has higher phenol content than L. Hydroxytyrosol and tyrosol were the more abundant phenols in both OMW extracts, with hydroxytyrosol significantly higher in S as compared to L. Both extracts failed to induce cell death when challenged with endothelial cells and vascular smooth muscle cells in cell viability experiments. On the contrary, the higher extract dosages employed significantly affected cell metabolic activity, as indicated by the MTT tests. Their ability to counteract H2O2-induced oxidative stress and cell death was assessed to investigate potential antioxidant activities of the extracts. Fluorescence measurements obtained with the reactive oxygen species (ROS) probe H2DCF-DA indicated strong antioxidant activity of the two OMW extracts in both cell models, as indicated by the inhibition of H2O2-induced ROS generation and the counteraction of the oxidative-induced cell death. Our results indicate LLAC-obtained OMW extracts as a safe and useful source of valuable compounds harboring antioxidant activity.
