AbstractRice bran supplementation provides nutrients, prebiotics and phytochemicals that enhance gut immunity, reduce enteric pathogens in mice and diarrhea in neonatal pigs, and warranted attention for improvement of environmental enteric dysfunction (EED) in children at risk. EED is a condition that drives childhood stunting via intestinal dysbiosis and impaired nutrient metabolism. This study investigated effects of rice bran supplementation on growth, EED biomarkers, gut microbiome and metabolome in weaning infants from 6 to 12 months old in Nicaragua and Mali. Healthy infants were randomized to a control group or rice bran group that received daily supplementation at increasing doses each month. Stool microbiomes were characterized using 16S rDNA amplicon sequencing. Stool metabolomes were analyzed using ultra-high-performance liquid-chromatography tandem mass-spectrometry. Statistical comparisons were completed at 6, 8, and 12 months of age. Daily consumption of rice bran was safe and feasible for infant growth, decreasing alpha-1 antitrypsin levels, and modulating gut microbiome and metabolome when compared to control. This study investigated the effects of rice bran supplementation on weight for age and length for age z-scores (WAZ, LAZ), EED stool biomarkers, as well as microbiota and metabolome signatures in weaning infants from 6 to 12 months old that reside in Nicaragua and Mali. Healthy infants were randomized to a control (no intervention) or a rice bran group that received daily supplementation with increasing doses at each month (1–5 g/day). Stool microbiota were characterized using 16S rDNA amplicon sequencing. Stool metabolomes were analyzed using ultra-high-performance liquid-chromatography tandem mass-spectrometry. Statistical comparisons were completed at 6, 8, and 12 months of age. Daily consumption of rice bran was safe and feasible to support changes in LAZ from 6–8 and 8–12 months of age in Nicaragua and Mali infants when compared to control. WAZ was significantly improved only for Mali infants at 8 and 12 months. Mali and Nicaraguan infants showed major differences in the overall gut microbiota and metabolome composition and structure at baseline, and thus each country cohort demonstrated distinct microbial and metabolite profile responses to rice bran supplementation when compared to control. Rice bran is a practical dietary intervention strategy that merits development in rice-growing regions that have a high prevalence of growth stunting due to malnutrition and diarrheal diseases. Rice is grown as a staple food, and the bran is used as animal feed or wasted in many low- and middle-income countries where EED and stunting is prevalent.

High Performance Liquid Chromatography (HPLC) was used to determine the vitamin and mineral contents of some locally sourced ingredients such as fish liver oil, Adansonia digitata, Telfaria occidentalis, Celosia spp, Amaranthus cruentas leaves, Xylopia aetiopica, capsicum frutenscen, Saccharomyces cerevicae, rice bran, palm oil and colostrums. The results indicated that fish liver oil was the most potent source of vitamins A, D, E and B12 represented by 9x10 6 iu/100g, 48000icu/100g, 30mg/100g, and 40.5mg/100g, respectively. The B-Complex vitamins were present among the spices and vegetables analyzed. Saccharomyces cerevicae was observed to a potent source of thiamin (9.4mg /100g) and niacin (41.8 mg / 100g). Capsicum spp and Telfaria occidentalis had ample amounts of riboflavin, thiamin and niacin. Rice bran had the highest amount of pyridoxine, 28.6mg/100g, while panthothenate was present more in the milk (8.2mg/100g) than other materials analysed. The mineral composition of the materials analysed showed that they have fairly uniform potency. Amaranthus cruentas was found to contain more mineral with 3.0, 1.4, 5.13, 1.10, 5.04 and 1.09% for copper (cu), Zinc (Zn), potassium (K), Sodium (Na), Calcium (ca), and magnesium (mg), respectively. The results of the analysis indicated that vitamin and mineral potencies of these ingredients can be harnessed for the production of vitamin and mineral premix for animal feed formulation

The increasing popularity of pike in angling and fish farming has created a need to increase pike production. However, intensive pike farming is subject to limitations due to diseases and pathogens. Sodium chloride (NaCl) could be a good alternative to chemotherapeutics, especially for protecting the fish against pathogens and parasites at early life stages. However, the impact of high salinity on the symbiotic bacteria inhabiting freshwater fish is still unclear. Therefore, our objective was to analyze the gut microbiome to find possible changes caused by salinity. In this study, the influence of 3‰ and 7‰ salinity on pike fry was investigated. High-throughput 16S rRNA gene amplicon sequencing was used to profile the gut microbiome of the fish. It was found that salinity had a statistically significant influence on pike fry mortality. Mortality was highest in the 7‰ salinity group and lowest in the 3‰ group. Microbiological analysis indicated that Proteobacteria and Actinobacteria predominated in the pike gut microbiome in all examined groups, followed by lower percentages of Bacteroidetes and Firmicutes. There were no statistically significant differences in the percent abundance of bacterial taxa between the control group and groups with a higher salinity. Our results suggest that salinity influences the gut microbiome structure in pike fry, and that 3‰ salinity may be a good solution for culturing pike at this stage in their development.

Bazhen is a classic prescription used for the prevention of qi and blood deficiency. The present study aimed to investigate the effects of dietary supplementation with modified Bazhen powder (MBP) on sows during lactation. Forty pure-bred Yorkshire sows on day 100 of gestation were randomly fed a standard diet supplemented with 20 g MBP per sow per day (MBP group) or without (control group) during -14 to 7 days relative to parturition. Results showed that the serum levels of interleukin 2 (IL-2), immunoglobulin A (IgA), and IgG were higher, whereas IL-10 level was lower in sows fed with MBP diet than in controls on day 7 postpartum. A significantly elevated proportion of serum CD4+ T cells and a slight increase in the ratio of CD4+ to CD8+ T cells in the MBP group were also observed. Furthermore, MBP supplementation improved gastrointestinal function of postpartum sows, evidenced by increased levels of motilin, gastrin, and nitric oxide. Ultra high-performance liquid chromatography combined with a quadrupole time of flight and tandem mass spectrometer identified a total of 21 absorbed milk components. 16S rRNA gene amplicon sequencing data revealed that the microbiota diversity of the colostrum and transitional milk in the MBP group was increased. At the genus level, relative abundances of Enterococcus and Anaerostipes were significantly lower in the MBP group on day 0 of lactation. Metabolomic analysis showed that 38 metabolites were upregulated, and 41 metabolites were downregulated in the transitional milk; 31 metabolites were upregulated and 8 metabolites were downregulated in the colostrum in response to MBP. Metabolic pathways, protein digestion and absorption, and biosynthesis of amino acids were enriched in the colostrum and transitional milk. Our findings provide new insights into the beneficial effects of MBP, highlighted by the changes to the microbiota and metabolomic profile of breast milk from sows fed with an MBP-supplemented diet. Thus, MBP should be considered as a potential dietary supplement for lactating sows in pork production.

In recent years there has been increasing interest in the potential beneficial effects of probiotic bacteria, particularly concerning their immune-modulating effects. Considering the involvement of free radicals in immunological processes, we tried to verify and compare the effects of probiotic (Lactobacillus casei) and conventional yoghurt on antioxidant and oxidant parameters in plasma of humans. In this study female volunteers consumed 100 g/day of probiotic (n = 17) or conventional yoghurt (n = 16) for two weeks (T1–T2) and 200 g/day for another two weeks (T2–T3). A wash-out phase lasting two weeks followed. Total antioxidant capacity (TAC), albumin, and bilirubin were determined photometrically, uric acid was determined by enzymatic methods, and vitamin E, carotenoids, malondialdehyde (MDA), and conjugated dienes (CD) were measured using high-performance liquid chromatography (HPLC). In the period of continuous yoghurt intake (T1–T3), mean concentrations of the antioxidants vitamin E, lycopene, and zeaxanthin decreased significantly (p < 0.01) in the probiotic and in the control group. The average concentrations of lutein, ß-carotene, albumin, uric acid, and bilirubin decreased significantly (p < 0.05) in the probiotic group, only. These alterations led to a significant (p < 0.001) decrease of the average TAC values during the period T1–T3 in both tested groups. In the interval of daily yoghurt consumption (T1–T3) the mean plasma levels of oxidant parameters MDA and CD increased significantly in the probiotic (MDA: p < 0.01; CD: p < 0.001) and the control group (CD: p < 0.01). The average activity of the antioxidant enzyme superoxide dismutase (SOD) was quite constant throughout the study in both groups. The mean activities of GSH-Px and catalase decreased significantly (p < 0.001) in the probiotic group, only after consuming yoghurt daily for four weeks (T1–T3). Although several parameters changed significantly during the study, no significant differences were observed between both investigated groups. Therefore, the results indicate a possible influence of both probiotic and conventional yoghurt on the plasma levels of antioxidant and oxidant parameters.

AbstractSunbirds feed on tobacco tree nectar which contains toxic nicotine and anabasine secondary metabolites. Our aim was to understand the effect of nicotine and anabasine on the gut microbiota composition of sunbirds. Sixteen captive sunbirds were randomly assigned to two diets: artificial nectar either with (treatment) or without (control) added nicotine and anabasine. Excreta were collected at 0, 2, 4 and 7 weeks of treatment and samples were processed for bacterial culture and high-throughput amplicon sequencing of the 16S rRNA gene. The gut microbiome diversity of the treated and control birds changed differently along the seven-week experiment. While the diversity decreased in the control group along the first three samplings (0, 2 and 4 weeks), it increased in the treatment group. The microbiota composition analyses demonstrated that a diet with nicotine and anabasine, significantly changed the birds’ gut microbiota composition compared to the control birds. The abundance of nicotine- and anabasine- degrading bacteria in the excreta of the treated birds, was significantly higher after four and seven weeks compared to the control group. Furthermore, analysis of culturable isolates, including Lactococcus, showed that sunbirds’ gut-associated bacteria were capable of degrading nicotine and anabasine, consistent with their hypothesised role as detoxifying and nutritional symbionts.

In modern poultry farming the most urgent tasks are to search for and test new environmentally safe feed and economically justifi ed additives that stimulate productivity, positively aff ect the health of poultry, and consequently increase the livability of livestock and other important zootechnical traits. In order to maintain physiologically normal processes of life, metabolism and increase the level of productivity of poultry it needs mineral components and their chelated compounds. The purpose of this work was to increase the egg productivity of poultry by using the silicon-containing poly additive Nabicat in feeding young hens and laying hens. The researches have been carried out on the poultry cross Highsex Brown under the environments in CJSC “Poultry Farm “Volzhskaya” in the Sredneakhtubinsky area in the Volgograd region. In this paper, The optimal dose of the introduction of the silicon-containing poly additive Nabicat in the diets of young and laying hens of the cross Highsex Brown has been scientifi cally justifi ed and the biological value of using an organic form of silicon has been determined. The positive infl uence of the optimal dose of poly additive on growth, consumption, and digestibility, nutrient metabolism in the poultry body, as well as egg productivity and quality of the resulting egg has been revealed. Thus, the average egg productivity per 1 hen in the control group during the experiment period was 321,3 pcs., and in the 1st, 2nd and 3rd experimental groups it was 322,6 pcs., 325,8 and 325,7 pcs., which was higher than in the control group by 0,40 %, 1,40 and 1,37 %, respectively. The average egg weight in the 1st experimental group exceeded the control by 0,43 g or 0,68 %, in the 2nd experimental group by 1,16 g or 1,84 %, in the 3rd experimental group by 0,81 g or 1,28 %. The economic eff ectiveness of using the organic silicon additive in the composition of Nabicat poly additive in the feeding of laying hens has been proved. The economic eff ect due to the use of diff erent doses of the introduction of silicon-containing poly additive Nabicat in compound feed for laying hens of the 1st experimental group was 141,66 rubles, the 2nd experimental group 962,76 rubles, and the 3rd experimental group 821,89 rubles.

Изучена целесообразность использования в рационах телят черно-пестрой породы комплексной минерально-витаминной кормовой добавки (КМВКД). Установлено пониженное содержание в кормах таких элементов, как кальций, фосфор, магний, сера, марганец, медь, цинк и кобальт. Соотношение питательных и минеральных веществ, степень их доступности для усвоения организмом не обеспечивают их потребность. Использование КМВКД способствовало улучшению всех гематологических показателей. Все испытываемые рецепты, имеющие в своем составе разные варианты макро-, микроэлементов и витаминов положительно влияют на обменные про- цессы в организме животных. У молодняка опытной группы, получавшей подкормку по рецепту 1, количество эритроцитов в сравнении с животными контрольной группы увеличилось на 6,6, гемоглобина на 6,93 соответственно, по рецепту 2 11,05 и 11,39 по рецепту 3 16,75 и 15,27, то есть соотношение макро- и микроэлементов, имеющихся в составе КМВКД и взаимодействие между собой меди, железа и кобальта сыграли особую роль на концентрацию форменных элементов. У молодняка, получавшего подкормку по рецепту 1 количество общего белка в сравнении с животными контрольной группы увеличилось на 1,99, кальция на 16,02, каротина на 10,92 соответственно, по рецепту 2 4,91, 25,96 и 19,12 по рецепту 3 на 3,58, 27,07 и 20,21. Значительное повышение содержания белка, кальция, фосфора и каротина по сравнению с животными контрольной группы объясняется тем, что недостаток их в рационах восполнялся за счет используемой подкормки, в результате их содержание увеличилось в сыворотке, но не выходило за пределы нормативных данных и соответствовало физиологическим нормам.Practicability of using a complex mineral and vitamin feed additive (CMVFA) in the ration of black-and-white calf breed was studied. Decreased content of such elements as calcium, phosphorus, magnesium, sulfur, manganese, copper, zinc, and cobalt was registered. The correlation of nutritive and mineral substances, and the level of their availability for digestion did not provide for their need. CMVFA use facilitated the increase of all hematologic parameters. Tested recipes which had different content of sapropel and zeolite, as well as various types of macronutrients, microelements, and vitamins, positively influenced the digestive processes in animal bodies. Young stock from the test group that was receiving food additives according to recipe No. 1 showed 6.6 increase in the quantity of red blood cells compared to control group animals, and 6.93 increase in hemoglobin quantity the ones who were fed according to recipe No. 2 demonstrated the increase by 11.05 and 11.39 recipe No. 3 showed 16.75 and 15.27 increase, which means that the ratio of macronutrients and microelements present in CMVFA and the mutual interaction of copper, iron, and cobalt played a special role in the concentration of formed elements due to the fact that they are integral elements of blood. Total protein of young stock that was receiving food additives according to recipe No. 1 was 1.99 higher than in test group animals calcium 16.02 higher carotene 10.92 as for recipe No. 2 the values were increased by 4.91, 25.96, and 19.12 recipe No. 3 3.58, 27.07, and 20.21. Substantial increase in the content of protein, calcium, phosphorus, and carotene compared to control group animals can be explained by the fact that their insufficiency in animal feed was replenished due to the use of feed additives, and as a result their content increased in the serum but did not go beyond the norm and corresponded to physiological norms.

Abstract Background Hispanics living in the USA may have unrecognized potential birthplace and lifestyle influences on the gut microbiome. We report a cross-sectional analysis of 1674 participants from four centers of the Hispanic Community Health Study/Study of Latinos (HCHS/SOL), aged 18 to 74 years old at recruitment. Results Amplicon sequencing of 16S rRNA gene V4 and fungal ITS1 fragments from self-collected stool samples indicate that the host microbiome is determined by sociodemographic and migration-related variables. Those who relocate from Latin America to the USA at an early age have reductions in Prevotella to Bacteroides ratios that persist across the life course. Shannon index of alpha diversity in fungi and bacteria is low in those who relocate to the USA in early life. In contrast, those who relocate to the USA during adulthood, over 45 years old, have high bacterial and fungal diversity and high Prevotella to Bacteroides ratios, compared to USA-born and childhood arrivals. Low bacterial diversity is associated in turn with obesity. Contrasting with prior studies, our study of the Latino population shows increasing Prevotella to Bacteroides ratio with greater obesity. Taxa within Acidaminococcus, Megasphaera, Ruminococcaceae, Coriobacteriaceae, Clostridiales, Christensenellaceae, YS2 (Cyanobacteria), and Victivallaceae are significantly associated with both obesity and earlier exposure to the USA, while Oscillospira and Anaerotruncus show paradoxical associations with both obesity and late-life introduction to the USA. Conclusions Our analysis of the gut microbiome of Latinos demonstrates unique features that might be responsible for health disparities affecting Hispanics living in the USA.

Bladder cancer (BC) is a common malignancy of the urinary system and a leading cause of death worldwide. In this work, untargeted metabolomic profiling of biological fluids is presented as a non-invasive tool for bladder cancer biomarker discovery as a first step towards developing superior methods for detection, treatment, and prevention well as to further our current understanding of this disease. In this study, urine samples from 24 healthy volunteers and 24 BC patients were subjected to metabolomic profiling using high throughput solid-phase microextraction (SPME) in thin-film format and reversed-phase high-performance liquid chromatography coupled with a Q Exactive Focus Orbitrap mass spectrometer. The chemometric analysis enabled the selection of metabolites contributing to the observed separation of BC patients from the control group. Relevant differences were demonstrated for phenylalanine metabolism compounds, i.e., benzoic acid, hippuric acid, and 4-hydroxycinnamic acid. Furthermore, compounds involved in the metabolism of histidine, beta-alanine, and glycerophospholipids were also identified. Thin-film SPME can be efficiently used as an alternative approach to other traditional urine sample preparation methods, demonstrating the SPME technique as a simple and efficient tool for urinary metabolomics research. Moreover, this study’s results may support a better understanding of bladder cancer development and progression mechanisms.
