Meningococcal infections remain particularly difficult to treat. Despite antibiotic therapy, the state of the patients often rapidly deteriorates. Early clinical studies suggest that meningococci acquire a form of resistance to antibiotic treatments during infections. Taking advantage of a humanized animal model of infection, we confirm that adherent bacteria become highly resistant to antibiotic treatments as early as 3-6 hours post infection, although fully sensitive in vitro. Within this time frame, meningococci adhere to the endothelium via their type IV pili, proliferate and eventually fill the vessel lumen. Using intravital imaging, we show that rapidly upon infection blood flow is dramatically decreased, thus limiting antibiotic access to infected vessels. Concomitantly, fibrin is deposited inside infected vessels in proximity to bacterial aggregates. Pharmacologically impairing thrombin generation by inhibiting Factor X activity not only improves blood flow in infected vessels, but also enhances the efficacy of the antibiotic treatment. Our results indicate that the combined administration of anticoagulants together with antibiotics might represent a therapeutic approach to treat meningococcal sepsis more efficiently.

The results of this study indicate that: 1) The implementation of the supervision of the madrasah principal in developing the guidance and counseling program has been well implemented and can help, guide, and provide direction to BK teachers in compiling and developing guidance and counseling programs. The steps are carried out in several steps, including preparing a schedule and supervision instrument, notifying the BK teacher who will be supervised, carrying out supervision to the BK teacher by checking the program and monitoring its implementation, and evaluating or assessing the BK program. The technique used in the implementation of the supervision of the madrasah principal is individual and group techniques. Meanwhile, the approach to the supervision of the head of the madrasah is a direct and indirect approach. 2) Factors supporting the implementation of the supervision of the madrasah Principal in developing guidance and counseling programs are cooperation and support from all parties in the school, the existence of a program or supervision schedule, and BK tools in the form of programs and RPL. While the inhibiting factor is the existence of an agenda or schedule for the madrasa principal that coincides with the schedule for the implementation of supervision for BK teachers so that supervision can be delayed. 3) The solution to overcome obstacles in implementing the supervision of the madrasa principal in developing a counseling guidance program is to arrange or change the supervision schedule to another time and coordinate with the BK teacher so that supervision can still be carried out.

The Covid-19 pandemic that has lasted for more than a year has changed many things. One thing that has changed is family communication, especially one whose members have been confirmed positive for Covid-19. This study aims to determine: 1) the communication patterns of the families of covid-19 survivors; 2) the supporting and inhibiting factors in healing psychological trauma due to Covid-19; and 3) the communication patterns of the survivors before, during, and after recovering from Covid-19. The research method used is a case study conducted in Sawitan Village, Banyumas Regency, and Rikmayung Village, Cilacap Regency. The study results show that: 1) family members are the prominent supporters of communication during survivors experiencing symptoms of Covid-19, and during the initial recovery period and socializing with the community; 2) the supporting factor for healing psychological trauma is family communication with a positive tone and good communication with neighbours and the environment, while the inhibiting factor is communication avoidance, and 3) Covid-19 survivors tend to communicate more empathy with other sufferers and are more careful in carrying out health protocols.

Opioids are the most potent widely used analgesics, primarily, but not exclusively, in palliative care. However, they are associated with numerous side effects, such as tolerance, addiction, respiratory depression, and cardiovascular events. This, in turn, can result in their overuse in cases of addiction, the need for dose escalation in cases of developing tolerance, and the emergence of dose-related opioid toxicity, resulting in respiratory depression or cardiovascular problems that can even lead to unintentional death. Therefore, a very important challenge for researchers is to look for ways to counteract the side effects of opioids. The use of peptides and their related compounds, which have been shown to modulate the effects of opioids, may provide such an opportunity. This short review is a compendium of knowledge about the most important and recent findings regarding selected peptides and their modulatory effects on various opioid actions, including cardiovascular and respiratory responses. In addition to the peptides more commonly reported in the literature in the context of their pro- and/or anti-opioid activity—such as neuropeptide FF (NPFF), cholecystokinin (CCK), and melanocyte inhibiting factor (MIF)—we also included in the review nociceptin/orphanin (N/OFQ), ghrelin, oxytocin, endothelin, and venom peptides.

The purpose of this research is to analyze a BUM Desa development strategy in Lumajang Regency. The research was conducted at BUM Desa Labruk Lor according to recommendations from the Lumajang Regency Government of East Java Province. The business units managed by BUM Desa Labruk Lor are fisheries, trade, agriculture, and culinary tourism businesses. Data were analyzed using the FFA (Force Field Analysis) approach by considering the driving factors for the development of BUM Desa (D) and the factors inhibiting the development of BUM Desa (H). The results show that the driving factor (key) is having formal legality (Village Regulations and AD/ART) and the formation of management is carried out democratically, while the inhibiting factor (key) is that business activities are still limited to the provision of production facilities and marketing services for fish-based processed products of catfish. The conclusion of this research is that the value of the strength field is smaller than the field of weakness, so it requires certain efforts to develop BUM Desa Labruk Lor. The recommendation for the BUM Desa development strategy is by expanding the network of cooperation with suppliers of fishery production facilities and the use of information technology as a medium for processed marketing.
