Karamunting is a plant that contains cholesterol. Cholesterol is needed by mud crabs as precursors of reproductive hormones in ovarian development. This study aims to determine the effect of karamunting leaf extract which is injected with mangrove crabs (Scylla sp) on the level of ovarian maturity. The stages of the research included extraction of karamunting leaves using ethanol 70%, experiments by injecting the parent crabs consisting of 3 treatments A (control), B treatment (0.25 mg / g body weight), C treatment (0.5 mg / g body weight) maintained for 20 days by feeding trash fish and measuring parameters including morphological ovarian development, somatic gonad index (GSI), and hepatosomatic index (HSI). The results showed that by giving karamunting extract at the end of the GSI study on control (1.70%), treatment A (5.61%), treatment B (4.24%) while on ovarian development with morphological observation at the end of the study on treatment ovarian control develops in the TKG II phase, A treatment in the TKG V phase while treatment B in TKG IV. This shows that by giving karamunting extract can provide the development of mud crab ovaries.

Katuk leaves are plants that contain sterols. Sterols are steroid precursors used for the development of mud crab ovaries. This study aims to determine the development of mangrove crab ovaries by giving katuk leaf extract. Stages of research include katuk leaf extraction, experiments by injecting the mother crab consisting of 3 treatments consisting of treatment A (control), treatment B (0.25 mg / g body weight), treatment C (0.5 mg / g body weight) ) maintained for 20 days by feeding trash fish and measuring parameters including morphological ovarian development, gonad somatic index, and hepatosomatic index. The results of the study on C treatment on the 20th day of observation by ovarian morphology have developed in the TKO IV phase, the final B TKO III treatment and the A-phase TKO III treatment. GSI on the 20th day in treatment C showed the highest GSI value of 16.1% and the lowest in treatment A was 12.2%. It can be concluded that the administration of katuk extract can accelerate the ovarian maturity of mud crabs.

The medicinal plants in aquaculture are currently being studied quite extensively by researchers. Medicinal plants can act as an inducer or aphrodisiac and an antifertility agent in gonad maturation. The aimed of the experiment was to known side effects of the herb M. malabathricum L. on maturation process in female mud crab (S. olivacea). This study used 40 crabs with an average weight of 200-250g. The experiment consists of two treatment groups. The controls group (A) that received 100 μl of aquadest and group treatment (B) received 100 μl of 2 mg/g of ethanol extract M. malabathricum L. were studied. For 5, 10, 15, and 20 days of experiment, crabs were anesthetized and sacrified. The ovary was collect for GSI and histology. The end of experiment days (20 day), the hemolymph was collect to used ELISA assay. The result showed, significant differences in treatment group and control groups (p<0.05) in terms the macroscopic morphology of ovary, the mean GSI and the diameter of oocytes. Histological showed the influence of extract M. malabathricum L. on inhibited the ovary development compared with the control groups. Based on this results, it is concluded that ethanol extract of M. malabathricum L. leaves at a dose of 2 mg/g act as an antifertility agent for ovary maturation of S. olivacea.

Neem leaves is one part of the neem tree which can be used as a traditional medicinal herb. The purpose of this study was to examine the effect of neem leaf ethanol extract on hepatosomatic index (HSI) and hepatocyte diameter in female mice. The study used a Complete Randomized Design consist of 4 treatments and 5 replications including K (aquadest), P1, P2 and P3 (neem leaf ethanol extract with dose of 8.4,11.2 and 14 mg/ kg BW/day. The treatment was administered orally with a volume of 0.2 mL for 21 days. Body weight were measured every 7 days. The liver was isolated, weighed and histologically prepared with paraffin method, HE staining and 5 μm incision thickness. HSI value determined from body weight and liver weight. Hepatocyte diameter were measured on 7 hepatocytes per lobule. The data was analyzed using ANOVA at 95% confidence level. The results showed that neem leaf ethanol extract had no significant effect (p> 0,05) on  body weight, liver weight, HSI value and hepatocyte diameter, but the liver microanatomy showed hepatocyte damage. It can be concluded that exposure of neem leaf ethanol extract for 21 days still safe to be used for reference of traditional medicine. Keywords: Neem leaf, HSI, hepatocyte diameter

Ikan baung (Hemibagrus nemurus) telah berhasil didomestikasi oleh Balai Riset Perikanan Budidaya Air Tawar dan Penyuluhan Perikanan, Bogor. Hasil penelitian domestikasi, populasi Cirata memiliki fekunditas dan pertumbuhan yang lebih bagus, populasi ini dikembangkan hingga tiga generasi (G1, G2 dan G3) namun, ada informasi terbatas mengenai perbandingan antara perkembangan gonad dari berbagai generasi ikan. Tujuan penelitian ini adalah untuk mengukur pertumbuhan dan mengamati perkembangan gonad generasi pertama (G-1), kedua (G-2), dan ketiga (G-3). Parameter yang diukur dan diamati meliputi pengukuran panjang, berat, laju pertumbuhan spesifik, morfologi dan berat gonad, diameter oosit, dan indeks gonad somatik (IGS). Pengambilan sampel dilakukan setiap dua bulan dengan mengumpulkan sepuluh sampel ikan dari setiap generasi. Jaringan gonad disiapkan secara histologis dan diwarnai larutan asetokarmin sebagai zat pewarna. Pengamatan menggunakan mikroskop dengan perbesaran 4 x 10. Hasil penelitian menunjukkan bahwa pada usia 222 hari, gonad jantan dan betina secara fisik masih dalam bentuk benang halus. Namun, karakteristik sekunder pada ikan jantan seperti papila sudah mulai tampak meski tidak terlalu jelas. Pada usia 283 hari, ukuran oosit berkisar antara 0.025-0.05 mm. Pada usia 345 hari, telur mulai terlihat dengan ukuran oosit G-1, G-2, dan G-3 masing-masing berkisar antara 0.59 ± 0.24, 0.39 ± 0.15, dan 0.48 ± 0.15 mm. Setelah usia 406 hari, perkembangan gonad mencapai TKG-III dengan diameter rata-rata oosit G-1, G-2, dan G-3 berkisar antara 1.13 ± 0.11, 0.92 ± 0.18, dan 1.11 ± 0.10 mm dengan IGS dari G-1, G-2, dan G-3 dari 8.20%, 0.98 %, dan 4.8%, masing-masing. Tidak ada perbedaan yang signifikan antara diameter oosit G-1, G-2, dan G-3 (P£0,05). Keturunan kedua (G-2) merupakan generasi terbaik untuk dijadikan kandidat budidaya. Asian redtail catfish (Hemibagrus nemurus) has been successfully domesticated by the Research Institute for Freshwater Aquaculture and Fisheries Extension, Bogor. The results of the domestication of Cirata population have better fecundity and growth, this population has been developed for up to three generations (G1, G2 and G3) however, there is limited information regarding the comparison between the ovarian development of different generations of the fish]. This study’s aim was to measure the growth and observe the ovarian development of the first (G-1), second (G-2), and third generations (G-3). The parameters measured and observed included the measurements of length, weight, specific growth rate, morphology and gonad weight, oocyte diameter, and somatic gonad index (IGS). The sampling was carried out every two months by collecting ten fish samples of each generation. Gonad tissues were histologically prepared and stained asetocarmine solution as the coloring agent. The observations used a microscope with a magnification of 4 x 10. The results of the study showed that at the age of 222 days, the male and female gonads were physically still in the form of fine threads. However, secondary characteristics in male fish such as papillae have begun to appear though not very clear. At the age of 283 days, oocytes’ sizes ranged between 0.025-0.05 mm. At the age of 345 days, the eggs were starting to be visible with the oocyte sizes of G-1, G-2, and G-3 ranged between 0.59 ± 0.24, 0.39 ± 0.15, and 0.48 ± 0.15 mm, respectively. After the age of 406 days, the development of gonads reached gonad maturating level III with the average diameter of the oocytes of G-1, G-2, and G-3 ranged between 1.13 ± 0.11, 0.92 ± 0.18, and 1.11 ± 0.10 mm with the IGS of G-1, G-2, and G-3 of 8.20%, 0.98%, and 4.8%, respectively. There was no significant difference among the oocyte diameters of G-1, G-2, and G-3 (P£0.05). The 2nd generation (G2) is the best generation to be a candidate aquaculture.

AbstractDoxorubicin (DOX) is a broad-spectrum chemotherapeutic drug used in the treatment of cancers. It acts by generating reactive oxygen species in target cells. The actions are, however, not limited to cancerous cells as it attacks healthy cells, killing them. This study investigated the benefits of the antioxidant, tert-butylhydroquinone (tBHQ), on testicular toxicity following DOX therapy. Twenty-four adult male albino rats were assigned randomly into four groups (n = 6), namely: normal control (NC), tBHQ, DOX and tBHQ + DOX groups. tBHQ (50 mg/kg body weight in 1% DMSO) was administered orally for 14 consecutive days, while a single DOX dose (7 mg/kg body weight) was administered intraperitoneally on Day 8. DOX decreased sperm count, motility and viability, and decreased the levels of steroidogenesis-related proteins, and reproductive hormones. Furthermore, DOX decreased the expression of antioxidant cytoprotective genes, and decreased the protein level of proliferating cell nuclear antigen in the testis. Conversely, DOX increased the expression of pro-inflammatory and pro-apoptotic genes in the testis. These negative effects were ameliorated following the intervention with tBHQ. Our results suggest that tBHQ protects the testis and preserves both steroidogenesis and spermatogenesis in DOX-treated rats through the suppression of oxidative stress, inflammation and apoptosis.

Objective: The aim of this study was to investigate hypoglycemic and hypolipidemic effects of Annona muricata leaf ethanol extract. This study also investigated phytochemical analysis of the extract and improvement in the islet of Langerhans.Methods: Hypoglycemic and hypolipidemic effects were evaluated using alloxan induced diabetic rats. Twenty-five Wistar rats devided into five groups, that are normal group, control group that induced by alloxan 125 mg/kg body weight and given CMC (carboxymethyl cellulose), and three treatment group that induced by alloxan and given Annona muricata leaf ethanol extract (AMLEE) with 50, 100 and 200 mg/kg body weight. Blood glucose and total cholesterol levels were measured before and after alloxan induction, and 21 d after AMLEE treatment. At the end of the study, all of the animals experiment were sacrificed for histopathological examination.Results: Phytochemical analysis revealed that flavonoids, tannins, saponins, phytosterols and phenols were present in ethanol extract of Annona muricata L. leaf. At the end of the study, blood glucose and total cholesterol levels in all AMLEE treated group were decreased significantly (p<0.05). Final blood glucose level in the groups that given AMLEE 50 mg/kg body weight (86.7±14.6 mg/dl) was almost the same with that of the normal group (91.25±28.38 mg/dl). The result of the histopathological examination is not showed an improvement of the islet of Langerhans in AMLEE treated groups.Conclusion: In conclusion, the ethanol extract of Annona muricata L. leaf have hypoglycemic and hypolipidemic effects. However, there was no improvement in the islet of Langerhans damage.

Acid gelugur fruit (Garcinia atroviridis) is one of the Garcinia families that has long been used as traditional medicine. Gelugur acid has been widely used as a cooking spice by Malay people, but it also has the ability to reduce cholesterol. Besides that, it has antioxidant properties and can reduce body weight and cholesterol. The purpose of this study was to establish specific parameter standards and non-specific parameters from the ethanol extract of acid gelugur fruit. The extract was obtained by maceration method using 70% ethanol with a rendament of 37.15%. Specific parameters include observations of organoleptic extracts of tamarind fruit showed, dark brown, distinctive smelling, and sour. Contains several sekendur metabolites such as flavonoids and saponins. The level of the compound that is soluble in water is 3.6%, while the content of the compound dissolved in ethanol is 3.9%. Total ash content of 2.99%. Non-specific parameters include acid insoluble ash content of 1.03%. The drying rate of geluguric acid simplicia was 7.20%. Water content of sour fruit gelugur extract was 13%.

Background: The ethanol extract of Combretum dolichopentalum (EECD) is employed in Nigeria to stabilize the uterus after parturition. The ability of EECD to confer protection on rats destabilized by moderate concentrations of carbon tetrachloride (CCl4) was evaluated. Methods: Fifty rats were assigned to 5 groups of 10 rats each. The experimental animals after acclimatization were handled accordingly: Groups 1 and 2 respectively were maintained on food and water only throughout the study. Group 3, 4, and group 5 were pre-treated with 250 mg/kg and 500 mg/kg body weight of EECD and 50 mg/kg of silymarin for 28 respectively. All groups except group 1 were intoxicated to 0.2 ml/kg body weight of CCl4, administered via an intraperitoneal route on day 29. Serum pipetted from the blood of the rats after cardiac puncture was assayed for antioxidant enzymes, lipid peroxidation product and serum iron, zinc and biocarbonate. Haematological analysis was also conducted. Results: Administration of CCl4 at 0.2 ml/kg b.w slightly increased the oxidizing species as indicated in the concentration of malondialdehyde in the rats while reducing the antioxidant enzymes; it increased the Iron and zinc concentrations and also the haematological parameters except for the white blood cells. However, this was corrected by pre-treatment with the EECD dosedependently. Conclusion: These characteristics portends that the crude ethanol extract of C. dolichopentalum could be employed to correct minor oxidative perturbation induced by CCl4 intoxication

Background: Takokak fruit (Solanum torvum) is a type of eggplant containing solasodin, solamargin, solasonin and other phytochemicals components with anti-inflammatory and anti-cancer properties. The purpose of this study was to examine the effects of takokak ethanol extract on TNF-α, IL6, and SOD levels.Methods: Experimental factorial study design, with the effect of takokak extract concentration factor (0, 400 and 800 mg/kgBW), takokak extract administration time factor including preventive (weeks 1-7) and curative (weeks 6-12) administration, and necropsy factor (necropsy in weeks 12 and 16). The data obtained included rats body weight, TNF-α, IL-6 and SOD levels.Results: The result showed that the interaction between takokak concentration and handling has significant effect on the increase of TNF-α levels (p=0.003) and the decrease of IL-6 levels (p=0.000). Interaction between takokak concentration, handling and necropsy has significant effect on the increase of SOD levels (p=0.010).Conclusion: Takokak ethanol extract has significant effect on the increase of TNF-α, and SOD levels, and the decrease of IL-6 levels.
